Vitis vinifera cell suspension cultures carried out in shake flasks were closely examined for biomass growth and cell division in relation to carbohydrate, NH(4), NO(3)PO(4), and dissolved oxygen (DO)consumption. After inoculation, the oxygen uptake rate of the cultures measured on-tine was observed to increase continuously to a maximum value of 3.8 mmol O(2)L(-1)h(-1) at day 7 when cell division ceased and dissolved oxygen reached its lowest level of 17% air saturation. During this first phase of growth, the specific oxygen uptake rate remained constant at approximately 0.6 mmol 02 O(2) g(-1) dw h(-1)or approximately 2.2 mumol O(2), (10(6) cells)(-1) h(-1) whereas dry biomass concentration increased exponentially from 1.5 to 6.0 g dw L(-1). Thereafter, dry biomass concentration increased linearly to approximately 14 g dw L(-1) at day 14 following nitrate and carbohydrate uptake. During this second phase of growth, the biomass wet-to-dry weight ratio was found to increase in an inverse relationship with the estimated osmotic pressure of the culture medium. This corresponded to inflection points in the dry and wet biomass concentration and packed cell volume curves. Furthermore, growth and nutrient uptake results suggest that extracellular ammonium or phosphate ion availability may limit cell division. These findings indicate that cell division and biomass production of plant cell cultures may not always be completely associated, which suggests important new avenues to improve their productivity. 该研究通过生物信息学方法,从桑树基因组中获得了8个花青素生物合成调控关键转录因子(MYB)候选基因,利用转录组数据及实时荧光定量PCR技术,分析了各基因在不同组织及果实发育过程中的表达。聚类分析结果显示,4个MYB基因与葡萄、水稻和玉米花青素调控相关MYB基因聚为一类,仅1个MYB基因与拟南芥、苹果花青素调控相关MYB基因聚为一类。转录组数据显示多数基因在雄花中高水平表达。实时荧光定量PCR结果表明,2个MYB基因(MnMYBJMnMYB4)在果实发育过程中持续下调,1个MYB基因(MnMYB330)在果实发育过程中显著上调,分别与花青素在桑椹中的积累成负相关和正相关关系。因此,桑树MYB基因家族对花青素的积累可能存在正调控与负调控两种机制。  相似文献

Aziridinylphosphinothionates were prepared from optically active ethyl hydrogen phenylphos- phonothionates and l-bromo-2-alkanamines derived from leucine or valine. The aziridine ring was opened by the action of some nucleophiles. Refluxing the aziridinylphosphinothionates in acetone with sodium iodide caused hydrolysis accompanied by the rearrangement of the sulfur atom to give β -mercaptoethylphosphonamidates. The reaction mechanism was discussed with stereochemical considerations. The insecticidal activity of the products was also examined.  相似文献

Immunohistochemical methods combined with progressive plasmolysis were used to localize chalcone synthase (CHS), an important enzyme for plant metabolism of aromatics in hypocotyls of illuminated buckwheat (Fagopyrum esculentum M) seedlings. Illumination of etiolated seedlings with white light results in anthocyanin synthesis in the epidermal layer of the hypocotyl. Anthocyanin-containing epidermal peels, after fixation for 30 min in 4% paraformaldehyde, 2.5% glutaraldehyde, 0.1% caffeine, were treated with a specific rabbit anti-buckwheat CHS antibody and a 20 nm goat anti-rabbit IgG gold conjugate. CHS is specifically shown in epidermal cells as pink to dark red deposits. Progressive plasmolysis combined with our immunohistochemical method showed that CHS was located exclusively in the cytoplasm of the epidermal cells of buckwheat hypocotyls except for the guard cells, which contained no detectable CHS.  相似文献

二氢黄酮醇4-还原酶(DFR)是植物花色素苷合成途径中的关键酶,在植物花色的形成过程中起重要作用。依据七彩红竹转录组数据设计特异引物,采用ImPcR技术从七彩红竹中克隆获得了一个新的DFR基因cDNA全长,命名为IhDFR1(登录号为KF728205)。序列分析结果表明,IhDFR1基因cDNA全长945bp,编码314个氨基酸。生物信息学预测显示,该基因编码的蛋白具有典型的DFR蛋白功能结构域,存在2个特异结合位点,属于非Asn/Asp型DFR酶,与禾本科植物中的DFR具有较高的相似性。对不同发育时期七彩红竹的IhDFR1基因进行时空表达的结果显示,只有在竹秆颜色呈现红紫色时,IhDFR1基因才有表达。以上结果初步显示IhDFR1蛋白可能作为一个重要的酶参与竹秆花色素苷的代谢调控,同时为进一步研究七彩红竹花色素苷产生的分子机理和综合开发利用奠定了基础。  相似文献

The anthocyanin composition of berries of Maqui [Aristotelia chilensis (Mol.) Stuntz] was determined by HPLC with photodiode array and MS detection. Eight pigments corresponding to the 3-glucosides, 3,5-diglucosides, 3-sambubiosides and 3-sambubioside-5-glucosides of delphinidin and cyanidin were identified, the principal anthocyanin being delphinidin 3-sambubioside-5-glucoside (34% of total anthocyanins). The average total anthocyanin content was 137.6 +/- 0.4mg/100g of fresh fruit (211.9 +/- 0.6 mg/100g of dry fruit). The relative high anthocyanin content and the important presence of polar polyglycosylated derivatives makes the fruits of A. chilensis an interesting source of anthocyanin extracts for food and pharmaceutical uses.  相似文献

The investigations reported here focus on an in‐depth characterization of the secondary metabolite profile of Sanguisorba officinalis flowers. For this purpose, fresh flowers were extracted with MeOH/H2O and EtOH/H2O and the resulting crude extracts fractionated using CH2Cl2, AcOEt, and BuOH. Individual compounds were characterized by high performance liquid chromatography and gas chromatography coupled with mass spectrometric detection (HPLC‐DAD‐MSn and GC/MS). MeOH/H2O extraction and LC/MSn investigations revealed the occurrence of flavonoid glycosides (quercetin, kaempferol), ellagitannin glycosides and four anthocyanins. Among the latter, two components, i. e., cyanidin‐malonyl‐glucose and cyanidin‐galloyl‐hexose, have not been reported for S. officinalis so far. Furthermore, phenylethylamine was characterized for the first time in Sanguisorba by pH value dependent extraction with CH2Cl2. In addition, AcOEt and BuOH extracts were analyzed by GC/MS both prior to and after acid hydrolysis of secondary metabolites. For this purpose, the extracts were treated with 1 n HCl solution (105 °C, 1 h) and derivatized with BSTFA. Analyses revealed the occurrence of several classes of phenolic compounds, such as gallic acid, hydroxybenzoic acid, hydroxycinnamic acid and ellagic acid derivatives. Additionally, the most prominent ursane‐type triterpenoid (ziyu‐glycoside I) from Sanguisorba and its corresponding aglycone isomers were detected and assigned based on their characteristic fragmentation patterns.  相似文献
