The effect of waveguiding property (i.e., the intensity distribution) of the photoreceptor on the number of photons absorbed in a photoreceptor has been studied. It has been found that the effect is significant only for large values of the exposure and the maximum effect is less than 11% in the case of human rod photoreceptor. In the analysis, the funnelling effect, which follows from the coupling between the interior and exterior fields, has not been considered.Work partially supported by the Department of Science and Technology (India)B. Aquaspirillum magnetotacticum MS-1 cells cultured microaerobically (dissolved O2 tension 1% of saturation), expressed proteins with superoxide dismutase (SOD) activity. The majority (roughly 95%) of total cell superoxide dismutase activity was located in the cell periplasm with little or no activity in the cell cytoplasm. Irontype SOD (FeSOD) contributed 88% of the total activity activity detected, although a manganese-type SOD (MnSOD) was present in the periplasm as well. Cells cultured at a higher dissolved O2 tension (10% of saturation) expressed increased activity of the MnSOD relative to that of the FeSOD.  相似文献

The dielectrophoretic (DEP) crossover method has been applied to the detection of cell responses to toxicants. Time and dose responses of the human cultured leukemia (HL-60) line were measured for paraquat, styrene oxide (SO), N-nitroso-N-methylurea (NMU) and puromycin. These toxicants were chosen because of their different predominant mechanisms of action, namely membrane free radical attack, simultaneous membrane and nucleic acid attack, nucleic acid alkylation, and protein synthesis inhibition, respectively. For all treatments, the specific membrane capacitance (Cmem) of the cells decreased while the specific membrane conductance (Gmem) increased in dose- and time-dependent manners. The DEP responses correlated sensitively with alterations in cell surface morphology, especially folds, microvilli, and blebs, observed by scanning electron microscopy. The DEP method was more sensitive to agents that had a direct action on the membrane than to agents for which membrane alterations were secondary. The responses to paraquat and SO, which directly damaged the cell membrane, could be detected 15 min after exposure, while those for puromycin and NMU, which acted on intracellular targets, could be detected after 30 min. The detection times and dose sensitivity results showed that the DEP method is much faster and more sensitive than conventional cell and higher organism viability testing techniques. The feasibility of producing small instruments for toxicity detection and screening based on cellular dielectric responses is discussed.  相似文献

The Optically Detected Magnetic Resonance spectrum of lysozyme has been shown to consist of a multiplet of narrow components, at -1565 MHz, 1585 MHz, and 1620 MHz. The 1585 MHz component is the strongest feature of the spectrum. This is consistent with earlier reports which apparently resolved only this principal component in lysozyme. The linewidths reported here are the narrowest ever reported for tryptophan in proteins. Using Microwave-Induced Phosphorescence techniques, the dominant 1585 MHz line is seen to be coupled to a "narrow" phosphorescence emission component at about 4134A. This component has a bandwidth of about 25A compared to 42A for the normal O-O band for tryptophan in lysozyme.  相似文献

Summary Changes in the chord conductanceG and the membrane electromotive forceE m in the so-called breakdown region of large negative potential of theChara plasmalemma were analyzed in more detail. In addition to the increase inG, the voltage sensitivity of the change inG increased, which was the cause of marked inductive current in the breakdown region. The breakdown potential, defined as a critical potential at which both low and high slope conductances of theI–V m relationship cross, almost coincided with the potential at which an inductive current began to appear. This breakdown potential level changed with pH o in a range between 5 and 9. TheChara plasmalemma was electrically most tolerant around pH o 7.In some cellsE m shifted to a positive level as large as +50+70 mV during the breakdown phenomenon. Such a large positive shift ofE m is caused mainly by the increase in conductance of Cl and partly Ca2+ and K+.  相似文献

Abstract: Several studies have reported declines in brain total and free magnesium concentration after a traumatic insult to the CNS. Although the evidence suggests that this magnesium decline is associated with eventual neurologic outcome after trauma, the duration of free magnesium decline and its impact on related bioenergetic variables are relatively unknown. The present study has therefore used phosphorus magnetic resonance spectroscopy to determine the length of time that free magnesium remains suppressed after traumatic brain injury in rats. Immediately after the traumatic event, brain intracellular free magnesium declined to <60% of preinjury values and remained significantly depressed (50 ± 8%; p < 0.001) for 5 days before recovering to preinjury levels by day 8. Cytosolic phosphorylation ratio and mitochondrial oxidative capacity also significantly decreased ( p = 0.008) and increased ( p = 0.002), respectively, after trauma. However, unlike the time of maximum magnesium change, the maximum changes in these bioenergetic variables occurred at 16–24 h after trauma and thereafter remained stable until after the magnesium had recovered. We conclude that free magnesium decline after trauma precedes changes in bioenergetic variables. Furthermore, therapies targeted at reestablishing magnesium homeostasis after trauma may require administration over a 1-week period.  相似文献

Plants of Gracilaria sp.(chorda type), which grow along the coast of Uranouchi Inlet in Tosa Bay, southern Japan, showed the highest biomass in the summer (26 °C to 31 °C) and spring season (15.1 °C to 24.9 °C). Maximum biomass was 6952 g m–2 in July, but gradually decreased in the autumn (30.5 °C in September to 20 °C in November) and winter (19.5 °C in December to 14.9 °C in February). Variation in yields and gel strength of the agars, were shown to depend on the time in the season. After alkali treatment (5% NaOH, 2 h) at three different temperatures (70, 80, and 90 °C), the agars showed gel strengths essentially that of commercial grade agars, with the best gel obtained at 80 °C. Maximum gel strength (1455 g cm–2 of 1.5% agar gel) occurred in winter when the biomass and agar yield were low. Minimum gel strength was in spring. Gel strength was inversely correlated with agar yield, but was positively correlated with apparent viscosity. Maximum viscosity was 40 cP. in December. Gelling temperatures, pH of 1.5% agar gel, and moisture content in agars showed little variation.  相似文献

1.  This study examined spatial learning and memory in breeding adult male and female meadow voles, Microtus pennsylvanicus, and the effects of brief exposure to weak (0.1 mT or 1.0 Gauss rms) 60 Hz magnetic fields on their spatial performance.
2.  There were significant sex differences in the spatial performance of the polygynous meadow voles. Reproductive male voles displayed significantly better spatial learning and retention than reproductive females in a Morris water maze task, whereby individual voles had to learn and remember the location of a submerged hidden platform using extramaze visual cues. Males showed over 9 days of daily blocks of 4 trials both a faster rate and greater overall level of acquisition (shorter latency to find hidden platform) of the spatial task than did the females.
3.  Brief (maximum 5 min) exposure to 60 Hz magnetic fields during acquisition of the water maze task significantly enhanced the spatial performance of both the male and female meadow voles. Females showed a relatively greater facilitation of spatial acquisition and retention than did the males, resulting by day 9 in the elimination of sex differences in water maze performance.
4.  The time-varying magnetic fields may be : (i) functioning as orientation cues and providing directional cues, and/or (ii) affecting neuromodulatory systems that are involved in the mediation of spatial learning and memory. Exposure to weak time-varying magnetic fields has been shown to attenuate the activity of endogenous opioid systems, enhance protein kinase C (PKC) activity, and alter calcium ion flux. These effects have been shown to affect spatial learning and memory and are consistent with an enhancement of water maze performance.
