We describe the enantioseparation of functionalized derivatives of the Tr?ger base by HPLC on commercially available chiral stationary phases. Cellulose-derived Chiralcel OJ and brush-type Whelk O1 are demonstrated to be complementary to each other in their scope. On the basis of the results obtained, the separation of selected compounds was successfully transferred onto semipreparative columns. We believe that the availability of enantiopure functionalized derivatives of the Tr?ger base will stimulate the further use of this interesting molecular scaffold in molecular recognition, asymmetric catalysis, and related areas of research and technology.  相似文献

Accurate characterisation of the scanner's point spread function across the entire field of view (FOV) is crucial in order to account for spatially dependent factors that degrade the resolution of the reconstructed images. The HRRT users' community resolution modelling reconstruction software includes a shift-invariant resolution kernel, which leads to transaxially non-uniform resolution in the reconstructed images. Unlike previous work to date in this field, this work is the first to model the spatially variant resolution across the entire FOV of the HRRT, which is the highest resolution human brain PET scanner in the world. In this paper we developed a spatially variant image-based resolution modelling reconstruction dedicated to the HRRT, using an experimentally measured shift-variant resolution kernel. Previously, the system response was measured and characterised in detail across the entire FOV of the HRRT, using a printed point source array. The newly developed resolution modelling reconstruction was applied on measured phantom, as well as clinical data and was compared against the HRRT users' community resolution modelling reconstruction, which is currently in use. Results demonstrated improvements both in contrast and resolution recovery, particularly for regions close to the edges of the FOV, with almost uniform resolution recovery across the entire transverse FOV. In addition, because the newly measured resolution kernel is slightly broader with wider tails, compared to the deliberately conservative kernel employed in the HRRT users' community software, the reconstructed images appear to have not only improved contrast recovery (up to 20% for small regions), but also better noise characteristics.  相似文献

Object tracking is an instrumental tool supporting studies of cellular trafficking. There are three challenges in object tracking: the identification of targets; the precise determination of their position and boundaries; and the assembly of correct trajectories. This last challenge is particularly relevant when dealing with densely populated images with low signal‐to‐noise ratios—conditions that are often encountered in applications such as organelle tracking, virus particle tracking or single‐molecule imaging. We have developed a set of methods that can handle a wide variety of signal complexities. They are compiled into a free software package called Diatrack. Here we review its main features and utility in a range of applications, providing a survey of the dynamic imaging field together with recommendations for effective use. The performance of our framework is shown to compare favorably to a wide selection of custom‐developed algorithms, whether in terms of localization precision, processing speed or correctness of tracks.   相似文献

目的:探讨宝石能谱CT用于冠状动脉支架内成像质量评估的应用价值。方法:回顾分析56例冠状动脉支架植入后患者,按照支架厚度及直径分成四组。采用宝石能谱CT对冠状动脉成像及同时进行冠状动脉造影患者的影像学资料,通过分析宝石能谱CT的冠状动脉的成像质量,与冠状动脉造影进行对比,得出宝石能谱CT用于冠状动脉介入治疗术后支架内成像质量评估的价值。结果:宝石能谱CT显示支架内再狭窄的敏感度为100%,特异度为98.6%,阴性预测值为100%。直径≥3 mm的支架内成像质量优于直径3 mm的支架。但支架直径和厚度对宝石能谱CT支架内成像质量的可评估率没有差异。结论:宝石能谱CT均能提供优秀的冠状动脉支架内成像质量,采用宝石能谱CT替代冠状动脉造影术可对冠脉支架术后支架内影像进行评估。  相似文献

Candida rugosa lipase-catalyzed esterification of ibuprofen with 1-propanol was conducted in seven ionic liquids and the results were compared with those in isooctane. Although the enzyme showed comparable or higher activity in some ionic liquids compared to that in isooctane, only in the case of [BMIM]PF6 was the enantioselectivity (E = 24.1) almost twice that (E = 13.0) of isooctane. In another six ionic liquids the enzyme enantioselectivity was much poorer (E = 1.1-6.4). At the same conversion of 30%, E of [BMIM]PF6 is more than triple that of isooctane. The lipase stability in [BMIM]PF6 was improved by 25% of that in isooctane. It was concluded that [BMIM]PF6 could be applied to substitute the conventional organic solvent (isooctane) in the esterification of ibuprofen.  相似文献

Substituted racemic 1-phenylethylamine derivatives were reacted in molten phase with chiral N-substituted 1-phenylethylamine dicarboxylic acid derivatives (4-9), and their mixtures (e.g., 4+6, 7+8, etc.) in different ratios, and the crystalline quasi-racemic diastereomers so formed were separated. Nonlinear behavior of the mixtures of resolving agents having related structure and considerable positive and negative chiral-chiral interactions were observed.  相似文献

1. Possible impacts of water‐resource development on assemblages of freshwater macroinvertebrates were investigated in the upper Darling River and some of its tributaries in north‐western New South Wales (Australia), an arid and semi‐arid region of low relief where alteration of river flows has intensified through expansion of irrigated agriculture. 2. Study sites were grouped into four hydrological regimes resulting from impoundment, flow regulation, water abstraction and natural variation, namely (i) intermittent flow with relatively little hydrological alteration from water‐resource development, (ii) intermittent flow with substantial alteration, (iii) near‐perennial flow with substantial alteration but unimpounded and (iv) near‐perennial flow with substantial alteration plus impoundment by weirs that stabilise water levels. 3. Macroinvertebrates were sampled with three methods (a quantitative cylinder sampler, handnet sampling and baited traps) in three periods with differing hydrology (recessional low flow in June 2003, high flow in March 2004 and increasing flow after drought in December 2004). 4. Taxonomic richness, assemblage composition and catch per unit effort of the crayfish Cherax destructor differed significantly among the site groups, but total macroinvertebrate density and the AUSRIVAS O/E (Australian River Assessment System observed‐over‐expected) index did not. The principal spatial differences were between the intermittent and near‐perennial rivers, and apparent effects of water‐resource development and impoundment were more subtle. Temporal differences in richness, abundance and composition were substantial and appeared to be related mainly to variations in discharge and temperature. 5. Current macroinvertebrate‐based methods for assessing the ‘condition’ or ‘health’ of Australian dryland rivers are inadequate. Such assessments might be improved with (i) reference data that take adequate account of antecedent hydrological conditions, (ii) consideration of long‐term taxonomic richness as well as richness on individual sampling occasions, (iii) evaluation of invertebrate population sizes, (iv) analysis of assemblage data by trait composition and (v) adoption of the genus as the default level of taxonomic resolution.  相似文献

The enantioseparation of the sherry aroma components 5-oxo-4-hydroxyhexanoic acid γ-lactone (solerone) and 4,5-dihydroxyhexanoic acid γ-lactone (solerole) is achieved, using Chiraspher (Merck) as the chiral HPLC phase and the optical purity ascertained directly by HRGC with heptakis(3-O-acetyl-2,6-di-O-pentyl)-β-cyclodextrin (Lipodex D) as the chiral stationary phase. The absolute configurations of 4,5-dihydroxyhexanoic acid γ-lactones are assigned by 1H-NMR spectral data of diastereomeric α-methoxy-α-trifluoromethylphenylacetic acid (MTPA) esters, according to Mosher's model. The absolute configuration of three 4‐aryl‐3,4‐dihydro‐2(1H)‐pyrimidones (Biginelli compounds, DHPMs) was established by comparison of the typical circular dichroism (CD) spectra of individual enantiomers with reference samples of known absolute configuration. The enantiomers were obtained by semipreparative separation of racemic mixtures on a Chiralcel OD‐H chiral stationary phase. The method was used to establish the enantiopreference of various lipases in biocatalytic kinetic resolution experiments employing activated DHPM esters.
