Galactosylglycerolipids (GGLs) and chlorophyll are characteristic components of chloroplast in photosynthetic organisms. Although chlorophyll is anchored to the thylakoid membrane by phytol (tetramethylhexadecenol), this isoprenoid alcohol has never been found as a constituent of GGLs. We here described a novel GGL, in which phytol was linked to the glycerol backbone via an ether linkage. This unique GGL was identified as an Alkaline-resistant and Endogalactosylceramidase (EGALC)-sensitive GlycoLipid (AEGL) in the marine green alga, Ulva pertusa. EGALC is an enzyme that is specific to the R-Galα/β1-6Galβ1-structure of galactolipids. The structure of U. pertusa AEGL was determined following its purification to 1-O-phytyl-3-O-Galα1-6Galβ1-sn-glycerol by mass spectrometric and nuclear magnetic resonance analyses. AEGLs were ubiquitously distributed in not only green, but also red and brown marine algae; however, they were rarely detected in terrestrial plants, eukaryotic phytoplankton, or cyanobacteria.  相似文献

The coccolithophorid Emiliania huxleyi is a bloom-forming marine phytoplankton thought to play a key role as a biological pump that transfers carbon from the surface to the bottom of the ocean, thus contributing to the global carbon cycle. This alga is also known to accumulate a variety of polyunsaturated fatty acids. At 25 °C, E. huxleyi produces mainly 14:0, 18:4n − 3, 18:5n − 3 and 22:6n − 3. When the cells were transferred from 25 °C to 15 °C, the amount of unsaturated fatty acids, i.e. 18:1n − 9, 18:3n − 3 and 18:5n − 3, gradually increased. Among the predicted desaturase genes whose expression levels were up-regulated at low temperature, we identified a gene encoding novel ?15 fatty acid desaturase, EhDES15, involved in the production of n − 3 polyunsaturated fatty acids in E. huxleyi. This desaturase contains a putative transit sequence for localization in chloroplasts and a ?6 desaturase-like domain, but it does not contain a cytochrome b5 domain nor typical His-boxes found in ?15 desaturases. Heterologous expression of EhDES15 cDNA in cyanobacterium Synechocystis sp. PCC 6803 cells increased the level of n − 3 fatty acid species, which are produced at low levels in wild-type cells grown at 30 °C. The orthologous genes are only conserved in the genomes of prasinophytes and cryptophytes. The His-boxes conserved in orthologues varied from that of the canonical ?15 desaturases. These results suggested the gene encodes a novel ?15 desaturase responsible for the synthesis of 18:3n − 3 from 18:2n − 6 in E. huxleyi.  相似文献

Laurdan (6-lauroyl-2-dimethylaminonaphthalene) fluorescence spectroscopy has been applied to probe the physical status of the thylakoid membrane upon conversion of violaxanthin to zeaxanthin. So far, only phospholipid-dominated membranes have been studied by this method and hereby we report the first use of laurdan in mono- and digalactosyldiacylglycerol-dominated membrane systems. The generalised polarisation (GP) of laurdan was used as a measure of the structural effect of xanthophyll cycle pigments in isolated spinach (Spinacia oleracea) thylakoids and in model membrane vesicles composed of chloroplast galactolipids. Higher GP values indicate a membrane in a more ordered structure, whereas lower GP values point to a membrane in a less ordered fluid phase. The method was used to probe the effect of violaxanthin and zeaxanthin in thylakoid membranes at different temperatures. At 4, 25 and 37 °C the GP values for dark-adapted thylakoids in the violaxanthin-form were 0.55, 0.28 and 0.26. After conversion of violaxanthin to zeaxanthin, at the same temperatures, the GP values were 0.62, 0.36 and 0.34, respectively. GP values increased gradually upon conversion of violaxanthin to zeaxanthin. Similar results were obtained in the liposomal systems in the presence of these xanthophyll cycle pigments. We conclude from these results that the conversion of violaxanthin to zeaxanthin makes the thylakoid membrane more ordered.  相似文献

Primary leaves of Phaseolus vulgaris show concomitant changes in phospholipid, galactolipid, chlorophyll and fresh weight during leaf development from 3 to 32 days after planting. Phosphatidyl choline, phosphatidyl ethanolamine, and phosphatidyl inositol show only small changes on a mole per cent lipid phosphate basis during leaf development. The chloroplast lipids, phosphatidyl glycerol, monogalactosyl diglyceride (MGDG) and digalactosyl diglyceride (DGDG) all show marked increases and decreases which are coincident with chloroplast development. The decline in the leaf content of chloroplast polar lipids and chlorophyll become evident upon reaching maximal leaf size. The molar ratio of galactolipids (MGDG/DGDG), reaches a maximum value of 2.3 in expanding leaves, but steadily declines during senescence to a minimum value of 1.5 at abscission. The declining ratio is caused by a preferential loss of MGDG in the senescing leaves.  相似文献

A comparison of the chemical composition and physical states of chloroplast lipids, of atrazine-resistant (R) and sensitive (S) biotypes of Conyza canadensis L. (horseweed), in the rosetta stage showed: (1) the R biotype contains lower amounts of polar lipids in its thylakoids, as expressed on a chlorophyll basis, than the S biotype. (2) The chloroplasts of the R biotype have higher contents of monogalactosyl diacylglycerol (MGDG) and lower contents of digalactosyl diacylglycerol (DGDG) and phosphatidylglycerol (PG), than those of the S biotype. (3) The chloroplast total lipids exhibit a higher degree of unsaturation in the R biotype. This is due to a higher level of linolenic acid, and a lower level of palmitic acid in the glycolipids. The fatty acid compositions of the phospholipids, except that of PG, do not differ significantly. (4) The lipid matrix of the thylakoid membranes of the R biotype is more fluid than that of the S biotype, as measured by the fluorescence polarization technique. The results are discussed in terms of whether these differences are responsible for the herbicide resistance.  相似文献

The elaborate compartmentalization of plant cells requires multiple mechanisms of protein targeting and trafficking. In addition to the organelles found in all eukaryotes, the plant cell contains a semi-autonomous organelle, the plastid. The plastid is not only the most active site of protein transport in the cell, but with its three membranes and three aqueous compartments, it also represents the most topologically complex organelle in the cell. The chloroplast contains both a protein import system in the envelope and multiple protein export systems in the thylakoid. Although significant advances have identified several proteinaceous components of the protein import and export apparatuses, the lipids found within plastid membranes are also emerging as important players in the targeting, insertion, and assembly of proteins in plastid membranes. The apparent affinity of chloroplast transit peptides for chloroplast lipids and the tendency for unsaturated MGDG to adopt a hexagonal II phase organization are discussed as possible mechanisms for initiating the binding and/or translocation of precursors to plastid membranes. Other important roles for lipids in plastid biogenesis are addressed, including the spontaneous insertion of proteins into the outer envelope and thylakoid, the role of cubic lipid structures in targeting and assembly of proteins to the prolamellar body, and the repair process of D1 after photoinhibition. The current progress in the identification of the genes and their associated mutations in galactolipid biosynthesis is discussed. Finally, the potential role of plastid-derived tubules in facilitating macromolecular transport between plastids and other cellular organelles is discussed.  相似文献

The geographical distribution of the red alga Pterosiphonia complanata (Clem.) Falkenb. is described. New records confirm that it is restricted to the warmer waters of the North Atlantic and extend its known distribution in the British Isles where it is at the northern limits of its range. The apparent scarcity and restricted distribution of the species in the British Isles and the absence of sexual plants throughout its range are discussed in relation to the habitat and possible life history of the species.  相似文献

The negative charge at the root surface is mainly derived from the phosphate group of phospholipids in plasma membranes (PMs) and the carboxyl group of pectins in cell walls, which are usually neutralized by calcium (Ca) ions contributing to maintain the root integrity. The major toxic effect of aluminum (Al) in plants is the inhibition of root elongation due to Al binding tightly to these negative sites in exchange for Ca. Because phospholipid and pectin concentrations decrease in roots of some plant species under phosphorus (P)-limiting conditions, we hypothesized that rice (Oryza sativa L.) seedlings grown under P-limiting conditions would demonstrate enhanced Al tolerance because of their fewer sites on their roots. For pretreatment, rice seedlings were grown in a culture solution with (+P) or without (−P) P. Thereafter, the seedlings were transferred to a solution with or without Al, and the lipid, pectin, hemicellulose, and mineral concentrations as well as Al tolerance were then determined. Furthermore, the low-Ca tolerance of P-pretreated seedlings was investigated under different pH conditions. The concentrations of phospholipids and pectins in the roots of rice receiving −P pretreatment were lower than those receiving +P pretreatment. As expected, seedlings receiving the −P pretreatment showed enhanced Al tolerance, accompanied by the decrease in Al accumulation in their roots and shoots. This low P-induced enhanced Al tolerance was not explained by enhanced antioxidant activities or organic acid secretion from roots but by the decrease in phospholipid and pectin concentrations in the roots. In addition, low-Ca tolerance of the roots was enhanced by the −P pretreatment under low pH conditions. This low P-induced enhancement of low-Ca tolerance may be related to the lower Ca requirement to maintain PM and cell wall structures in roots of rice with fewer phospholipids and pectins.  相似文献

The violaxanthin cycle describes the reversible conversion of violaxanthin to zeaxanthin via the intermediate antheraxanthin. This light-dependent xanthophyll conversion is essential for the adaptation of plants and algae to different light conditions and allows a reversible switch of photosynthetic light-harvesting complexes between a light-harvesting state under low light and a dissipative state under high light. The photoprotective functions of zeaxanthin have been intensively studied during the last decade, but much less attention has been directed to the mechanism and regulation of xanthophyll conversion. In this review, an overview is given on recent progress in the understanding of the role of (i) xanthophyll binding by antenna proteins and of (ii) the lipid properties of the thylakoid membrane in the regulation of xanthophyll conversion. The consequences of these findings for the mechanism and regulation of xanthophyll conversion in the thylakoid membrane will be discussed.  相似文献
