The supplementation of conjugated linoleic acids (CLA) to the rations of dairy cows represents an opportunity to reduce the content of milk fat. Therefore, CLA have the potential beneficial effect of reducing energy requirements of the early lactating cow. The present study aimed at the examination of long-term and post-treatment effects of dietary CLA intake on performance, variables of energy metabolism-like plasma levels of non esterified fatty acids (NEFA) and ß-hydroxybutyrate (BHB), and fatty acid profile in milk fat. Forty-six pregnant German Holstein cows were assigned to one of three dietary treatments: (1) 100 g/d of control fat supplement (CON), (2) 50 g/d of control fat supplement and 50 g/d of CLA supplement (CLA-1) and (3) 100 g/d of CLA supplement (CLA-2). The lipid-encapsulated CLA supplement consisted of approximately 10% of trans-10, cis-12 CLA and cis-9, trans-11 CLA each. The experiment started 1 d after calving and continued for about 38 weeks, divided into a supplementation (26 weeks) and a depletion period (12 weeks). Over the first 7 weeks of treatment, 11 and 16% reductions in dry matter intake compared to control were observed for the cows fed CLA-1 and CLA-2 supplements respectively. Consequently, the calculated energy balance for these two CLA groups was lower compared to the control. Plasma levels of NEFA and BHB remained unaffected. Later in lactation the highest CLA supplementation resulted in a reduction of milk fat content of 0.7%. However, no reduction in milk fat yield, and accordingly no milk fat depression (MFD), could be shown. The trans-10, cis-12 CLA in milk fat increased with increasing dietary CLA supplementation in a dose-dependent manner. The proportion of C16 in milk fat was decreased by the highest CLA supplementation. With the exception of an increase in plasma glucose level in the CLA-2 group, no post-treatment effects were observed. Overall, under the conditions of the present study no improvement in the calculated energy balance by CLA supplementation could be shown for the entire evaluation period. 相似文献

β, β‐carotene‐9′, 10′‐dioxygenase (BCO2) plays a role in cleaving β‐carotene eccentrically, and may be involved in the control of adipose and milk colour in cattle. The bovine BCO2 gene was sequenced as a potential candidate gene for a beef fat colour QTL on chromosome (BTA) 15. A single nucleotide base change located in exon 3 causes the substitution of a stop codon (encoded by the A allele) for tryptophan80 (encoded by the G allele) (c. 240G>A, p.Trp80stop, referred to herein as SNP W80X). Association analysis showed significant differences in subcutaneous fat colour and beta‐carotene concentration amongst cattle with different BCO2 genotypes. Animals with the BCO2 AA genotype had more yellow beef fat and a higher beta‐carotene concentration in adipose tissues than those with the GA or GG genotype. QTL mapping analysis with the BCO2 SNP W80X fitted as a fixed effect confirmed that this SNP is likely to represent the quantitative trait nucleotide (QTN) for the fat colour‐related traits on BTA 15. Moreover, animals with the AA genotype had yellower milk colour and a higher concentration of beta‐carotene in the milk. 相似文献

The aim of this study was to analyze milk protein composition in purebred and crossbred dairy cattle and estimate the effects of individual sources of variation on the investigated traits. Milk samples were collected from 505 cows from three commercial farms located in Northern Italy, some of which had originated from crossbreeding programs, although most were purebred Holsteins (HO). The basic crossbreeding scheme was a three-breed rotational system using Swedish Red (SR) semen on HO cows (SR×HO), Montbeliarde (MO) semen on SR×HO cows (MO×(SR×HO)) and HO semen again on MO×(SR×HO) cows. A smaller number of purebred HO from each of the herds were mated inverting the breed order (MO×HO and SR×(MO×HO)) or using Brown Swiss (BS) bulls (BS×HO) then MO bulls (MO×(BS×HO)). Milk samples were analyzed by reverse-phase HPLC to obtain protein fraction amounts (g/l) and proportions (% of total true protein). Traits were analyzed using a linear model, which included the fixed effects of herd-test-day (HTD), parity, days in milk and breed combination. Results showed that milk protein fractions were influenced by HTD, stage of lactation, parity and breed combination. The increase in protein concentration during lactation was due in particular to β-casein (β-CN), αS1-CN and β-lactoglobulin (β-LG). The higher protein content of primiparous milk was mainly due to higher concentrations of all casein fractions. The milk from crossbred cows had higher contents and proportions of κ-CN and α-lactalbumin (α-LA), lower proportions of β-LG and greater proportion of caseins/smaller in whey proteins on milk true protein than purebred HO. The three-way crossbreds differed from two-way crossbreds only in having greater proportions of α-LA in their milk. Of the three-way crossbreds, the SR sired cows yielded milk with a smaller content and proportion of β-LG than the MO sired cows, and, consequently, a higher proportion of caseins than whey proteins. Results from this study support the feasibility of using crossbreeding programs to alter milk protein profiles with the aim of improving milk quality and cheese-making properties. 相似文献

The content, composition and variation of vitamin compounds in goat milk have been little studied. An experimental design was based on 28 commercial farms, selected considering the main feeding system (based on main forage and especially pasture access), goat breed (Alpine vs Saanen) and reproductive management (seasonal reproduction), in the main French goat milk production area. Each farm received two visits (spring and autumn) that included a survey on milk production conditions and bulk milk sampling. Milk vitamins (A, E, B2, B6, B9, B12) and carotenoid concentrations plus colour indices were evaluated. A stepwise approach determined the variables of milk production conditions that significantly altered milk indicators. The main forage in the diet was the major factor altering goat milk vitamin and carotenoid concentrations and colour indices. Bulk milk from goats eating fresh grass as forage was richer in α-tocopherol (+64%), pyridoxal (+35%) and total vitamin B6 (+31%), and b* index (characterising milk yellowness in the CIELAB colour space) was also higher (+12%) than in milk from goats eating conserved forages. In milk from goats eating fresh grass, concentrations of pyridoxamine, lutein and total carotenoids were higher than in milk of goats fed corn silage (+24, +118 and +101%, respectively), and retinol and α-tocopherol concentrations were higher than in milk of goats fed partially dehydrated grass (+45 and +55%). Vitamin B2 concentration was higher in milk of goats eating fresh grass than in milk of goats fed hay or corn silage as forage (+10%). However, bulk milk when goats had access to fresh grass was significantly poorer in vitamin B12 than when fed corn silage (?46%) and in γ-tocopherol (?31%) than when fed conserved forage. Alpine goats produced milk with higher vitamin B2 and folate concentrations than Saanen goats (+18 and +14%, respectively). Additionally, the milk colour index that discriminates milks based on their yellow pigment contents was 7% higher in milk from Alpine than Saanen herds, but milk from Saanen goats was richer in lutein (+46%). Goat milks were richer in vitamins B2 and B12 and folates, but poorer in vitamin B6 in autumn than in spring (+12, +133, +15 and ?13%, respectively). Elf5 is an epithelial-specific ETS factor. Embryos with a null mutation in the Elf5 gene died before embryonic day 7.5, indicating that Elf5 is essential during mouse embryogenesis. Elf5 is also required for proliferation and differentiation of mouse mammary alveolar epithelial cells during pregnancy and lactation. The loss of one functional allele led to complete developmental arrest of the mammary gland in pregnant Elf5 heterozygous mice. A quantitative mRNA expression study and Western blot analysis revealed that decreased expression of Elf5 correlated with the downregulation of milk proteins in Elf5(+/-) mammary glands. Mammary gland transplants into Rag(-/-) mice demonstrated that Elf5(+/-) mammary alveolar buds failed to develop in an Elf5(+/+) mammary fat pad during pregnancy, demonstrating an epithelial cell autonomous defect. Elf5 expression was reduced in Prolactin receptor (Prlr) heterozygous mammary glands, which phenocopy Elf5(+/-) glands, suggesting that Elf5 and Prlr are in the same pathway. Our data demonstrate that Elf5 is essential for developmental processes in the embryo and in the mammary gland during pregnancy. 相似文献

Until the turn of the century, farmers in West Africa considered cotton to be the ‘white gold’ for their livelihoods. Large fluctuations in cotton prices have led farmers to innovate into other business including dairy. Yet the productivity of cows fed traditional diets is very poor, especially during the long dry season. This study combines earlier published results of farmer participatory experiments with simulation modelling to evaluate the lifetime productivity of cows under varying feeding strategies and the resulting economic performance at farm level. We compared the profitability of cotton production to the innovation of dairy. The results show that milk production of the West African Méré breed could be expanded if cows are supplemented and kept stall-fed during the dry season. This option seems to be profitable for better-off farmers, but whether dairy will replace (some of) the role of cotton as the white gold for these smallholder farmers will depend on the cross price elasticity of cotton and milk. Farmers may (partly) replace cotton production for fodder production to produce milk if the price of cotton remains poor (below US$0.35/kg) and the milk price relatively strong (higher than US$0.38/kg). Price ratios need to remain stable over several seasons given the investments required for a change in production strategy. Furthermore, farmers will only seize the opportunity to engage in dairy if marketing infrastructure and milk markets are further developed. 相似文献

In this study a novel method to determine iodine concentrations in human breast milk was developed and validated. The iodine was analyzed by inductively coupled plasma mass spectrometry (ICPMS) following tetramethylammonium hydroxide (TMAH) extraction at 90 °C in disposable polypropylene tubes. While similar approaches have been used previously, this method adopted a shorter extraction time (1 h vs. 3 h) and used antimony (Sb) as the internal standard, which exhibited greater stability in breast milk and milk powder matrices compared to tellurium (Te). Method validation included: defining iodine linearity up to 200 μg L−1; confirming recovery of iodine from NIST 1549 milk powder. A recovery of 94–98% was also achieved for the NIST 1549 milk powder and human breast milk samples spiked with sodium iodide and thyroxine (T4) solutions. The method quantitation limit (MQL) for human breast milk was 1.6 μg L−1. The intra-assay and inter-assay coefficient of variation for the breast milk samples and NIST powder were <1% and <3.5%, respectively. NIST 1549 milk powder, human breast milk samples and calibration standards spiked with the internal standard were all stable for at least 2.5 months after extraction. The results of the validation process confirmed that this newly developed method provides greater accuracy and precision in the assessment of iodine concentrations in human breast milk than previous methods and therefore offers a more reliable approach for assessing iodine concentrations in human breast milk. 相似文献

目的探讨岩藻糖基化人乳低聚糖在新生儿无乳链球菌肺炎中的治疗作用。方法收集本院2015年7月至2017年2月痰培养阳性新生儿无乳链球菌、大肠埃希菌和肺炎克雷伯菌感染性肺炎病例,分析不同喂养方式、不同病原类别肺炎在住院时间、炎症、并发症等指标差异,探讨岩藻糖基化人乳低聚糖对无乳链球菌感染的独特治疗作用;使用含有不同浓度岩藻糖基化人乳低聚糖的培养基对无乳链球菌进行培养,研究岩藻糖基化人乳低聚糖抑制无乳链球菌的最佳浓度;分别使用岩藻糖基化人乳低聚糖或酸性人乳低聚糖加入培养基中对无乳链球菌、大肠埃希菌、肺炎克雷伯菌进行培养,进一步探讨岩藻糖基化人乳低聚糖对无乳链球菌的独特抑制作用。结果无乳链球菌肺炎患儿,母乳喂养组的住院时间较人工喂养组住院时间减少[(8.35±0.77)d vs(10.91±0.54)d,t=1.557 676,P<0.05)],PCT值较人工喂养组低[(2.38±0.63)ng/mL vs(8.69±2.23)ng/mL,t=1.419 964,P<0.05],白细胞计数较人工喂养组低[(13.28±1.08)×10^9/L vs(16.16±0.98)×10^9/L,t=1.878 447,P<0.05],脑膜炎发生率较人工喂养组低(5%vs 35%,χ^2=5.601 353,P<0.05),呼吸机使用率较人工喂养组低(10.0%vs 36.4%,χ^2=4.005 042,P<0.05);大肠埃希菌肺炎患儿、肺炎克雷伯菌肺炎患儿,母乳喂养组与人工喂养组比较,住院时间稍减少,PCT值、白细胞计数、脑膜炎发生率和呼吸机使用率均有所降低,差异无统计学意义;细菌培养实验发现岩藻糖基化人乳低聚糖在2.5 mg/L浓度的时候可以明显抑制无乳链球菌的生长,对大肠埃希菌和肺炎克雷伯菌无明显抑制作用,酸性人乳低聚糖对3种细菌均无抑制作用。结论岩藻糖基化人乳低聚糖可以明显抑制无乳链球菌的生长,可能成为未来无乳链球菌感染性肺炎治疗中新的靶点,值得深入研究。 相似文献

Exposure to cow's milk constitutes one of the most common causes of food allergy. In addition, exposure to soy proteins has become relevant in a restricted proportion of milk allergic pediatric patients treated with soy formulae as a dairy substitute, because of the cross‐allergenicity described between soy and milk proteins. We have previously identified several cross‐reactive allergens between milk and soy that may explain this intolerance. The purpose of the present work was to identify epitopes in the purified αS1‐casein and the recombinant soy allergen Gly m 5.0101 (Gly m 5) using an α‐casein‐specific monoclonal antibody (1D5 mAb) through two different approaches for epitope mapping, to understand cross‐reactivity between milk and soy. The 1D5 mAb was immobilized onto magnetic beads, incubated with the peptide mixture previously obtained by enzymatic digestion of the allergens, and the captured peptides were identified by MALDI‐TOF MS analysis. On a second approach, the peptide mixture was resolved by RP‐HPLC and immunodominant peptides were identified by dot blot with the mAb. Finally, recognized peptides were sequenced by MALDI‐TOF MS. This novel MS based approach led us to identify and characterize four peptides on α‐casein and three peptides on Gly m 5 with a common core motif. Information obtained from these cross‐reactive epitopes allows us to gain valuable insight into the molecular mechanisms of cross‐reactivity, to further develop new and more effective vaccines for food allergy. 相似文献
