Lipophilic non-electrolyte spin labels greatly accelerate the fusion of unilamellar vesicles of dipalmitoylphosphatidylcholine when the system is maintained below the lipid phase transition. Differential scanning calorimetry and centrifugation measurements show that the transformed vesicles are large and probably unilamellar. Differential scanning calorimetry and fluorescence depolarization measurements were also carried out on mixtures of labeled dipalmitoylphosphatidylcholine vesicles and of vesicles composed of pure dimyristoylphosphatidylcholine. A mixing of the membrane components is observed when the vesicles are incubated above the transition temperature of the two constituent lipids. However, the process does not involve a real fusion of the entire vesicles. An exchange of lipid and label monomers between the two lipid phases seems to occur. These observations are discussed in view of the molecular organization of the spin label within the dipalmitoylphosphatidylcholine matrix below and above the lipid transition temperature.  相似文献

Several marine algae of the class Pelagophyceae produce the unusual marine sterol 24‐propylidenecholesterol, mainly as the (24E)‐isomer. The (24Z)‐isomer had previously been considered as a specific biomarker for Aureococcus anophagefferens, the ‘brown tide’ alga of the Northeast coast of the USA. To test this hypothesis and to generate chemotaxonomic information, the sterol compositions of 42 strains of pelagophyte algae including 17 strains of Aureococcus anophagefferens were determined by GC analysis. A more comprehensive sterol analysis by HPLC and 1H‐NMR was obtained for 17 selected pelagophyte strains. All strains analyzed contained 24‐propylidenecholesterol. In all strains belonging to the order Sarcinochrysidales, this sterol was found only as the (E)‐isomer, while all strains in the order Pelagomonadales contained the (Z)‐isomer, either alone or together with the (E)‐isomer. The occurrence of Δ22 and 24α‐sterols was limited to the Sarcinochrysidales. The first occurrence of Δ22‐24‐propylcholesterol in an alga, CCMP 1410, was reported. Traces of the rare sterol 26,26‐dimethyl‐24‐methylenecholesterol were detected in Aureococcus anophagefferens, and the (25R)‐configuration was proposed, based on biosynthetic considerations. Traces of a novel sterol, 24‐propylidenecholesta‐5,25‐dien‐3β‐ol, were detected in several species.  相似文献

Lipid bilayers of diphytanoyl lecithin (DPhL) in which a cyanobacterial toxin, microcystin-LR (MC-LR) was incorporated, were found to be a convenient model of natural mechanosensitive membranes. The effects of pressure difference, leading to lateral membrane tension, on artificial membranes formed on the tips of glass micropipettes were investigated using patch-clamp methodology. Emplacement of MC-LR from the bathing solution was enhanced by transmembrane voltage and/or pressure difference. MC-LR pores could be recorded over a wide voltage range, their opening probability being first increased and then reduced at high membrane potential. The pores exhibited several open pore conductance levels, the higher conductance states being more probable at greater lateral tensions. Ion gradient experiments established that the MC-LR pores are cation selective, but discriminate only weakly between K and Na. These results suggest that a lipid liquid crystal matrix containing monomers of multimeric pore-forming molecules could be used as a mechanical sensor and molecular switch. Offprint requests to: P. Abstract : The passage of either unesterified docosahexaenoic acid (DHA) or lysophosphatidylcholine-containing DHA (lysoPC-DHA) through an in vitro model of the blood-brain barrier was investigated. The model was constituted by a brain capillary endothelial cell monolayer set over the medium of an astrocyte culture. Cells were incubated for 4 h with a medium devoid of serum, then the endothelial cell medium was replaced by the same medium containing labeled DHA or lysoPC-DHA and incubations were performed for 2 h. DHA uptake by cells and its transfer to the lower medium (astrocyte medium when they were present) were measured. When the lower medium from preincubation and astrocytes were maintained during incubation, the passage of lysoPC-DHA was higher than that of unesterified DHA. The passage of both forms decreased when astrocytes were removed. The preference for lysoPC-DHA was not seen when the lower medium from preincubation was replaced by fresh medium, and was reversed when albumin was added to the lower medium. A preferential lysoPC-DHA passage also occurred after 2 h with brain endothelial cells cultured without astrocytes but not with aortic endothelial cells cultured and incubated under the same conditions. Altogether, these results suggest that the blood-brain barrier cells released components favoring the DHA transfer and exhibit a preference for lysoPC-DHA.  相似文献

Abstract The peptidoglycan layer of bacterial cell walls is biosynthesised using a lipid carrier undecaprenyl phosphate to assemble and transport the MurNAc(GlcNAc)-pentapeptide precursor. Similar lipid-linked cycles are involved in the biosynthesis of other bacterial exopolysaccharides and eukaryotic asparagine-linked glycoproteins, the latter involving the structurally related dolichyl phosphate as a lipid carrier. Recent protein sequence data and common inhibitors of the bacterial and eukaryotic systems have revealed functional similarities between the two systems. Biological and physical studies on the lipid carriers themselves have provided clues to their role in oligosaccharide translocation, but have not revealed significant differences in function between undecaprenyl phosphate and dolichyl phosphate. The presence of dolichyl phosphate and a family of saturated isoprenoid lipids in Archaebacteria suggests a possible evolutionary link between the two systems.  相似文献

Liacarus subterraneus is a large, soil-dwelling oribatid mite species that possesses a conspicuously shiny, clean and not wettable cuticular surface. The exocrine cuticular chemistry of this species was investigated by means of gas chromatography–mass spectrometry. Besides a fraction of hydrocarbons and a terpene, hexane extracts of whole mite bodies exhibited free carboxylic acids and their glycerides as main components. The compounds were arranged in three distinct extract profiles. Based on data from individual extracts, (1) the majority (more than 3/4) of specimens showed large amounts of 1,2-dioctanoyl-glycerol (and three other related esters) but no (or only traces of) free carboxylic acids. (2) In about 1/8 of extracts, free acids (mainly octanoic (caprylic) acid) and glycerides were detected. This second type of profile highly varied with respect to the relative abundance of acids and esters. (3) The third profile (in about 7% of specimens) exclusively exhibited free acids and no (or only traces of) glycerides. In addition, a few extracts exhibited no components at all. The extract compounds most likely originate from the lipid layer of the cerotegument of L. subterraneus. The cuticle of individuals that possessed extractable cerotegumental compounds (profile I, II, III) exhibited strong water repellent properties, while the cuticle of individuals that possessed no components in their extract did not. After hexane extraction, water repellent properties got lost. The distinct extract profiles detected most likely portray the stepwise generation of an anti-wetting, exocrine surface lipid layer of glycerides: If this layer is lost, fatty acids may be discharged again (profile III) and may subsequently esterify (profile II) to larger and more stable esters (diacyl-glycerols), eventually building up the “raincoat” (mainly profile I) of L. subterraneus.  相似文献

We studied the technogenic impact of a diamond mining company on the content of metabolites in the needles of Larix gmelinii (Rupr.) Kuzen. The samples of larch needles were collected in the Arctic zone in the restricted area where the diamond mining company works. The studied sites are located in Anabarsky district of Yakutia (Russia). An activity of the diamond mining company did not have a significant impact on the characteristics of the forest stand. But at the same time, near the sorting and enriching plants of the diamond mining company, the concentrations of Si, Ca, Fe, Mn, Al, Na, Sr, Ba, Zn, Pb, Ni, V were higher in the needles of L. gmelinii than those in the control zone. The activity of the diamond mining industry resulted in: intensification of the lipid peroxidation process, growth of a content of resin acids and amino acids, activation of cellular respiration processes, thickening of the cell wall, and a decline in the concentration of unsaturated fatty acids and antioxidants. We have concluded that some observable biochemical changes occur in the trees as a resulting effect of adaptive processes to the technogenic load despite the absence of significant changes in the forest stand.  相似文献

The constituents of the soluble cuticular lipids (SCL) of the leaf blades of Citrus aurantium L. were identified by gas chromatography-mass spectrometry and quantified. Major components were 1-alkanols (C24 to C40), n-alkyl esters (C36 to C56), n-alkanoic acids (C28 to C34), n-alkanes (C22 to C40) and triterpenones, while n-alkanals (C29 to C38), sterols, and alkyl benzenes (molecular weights 260, 274 and 288) made minor contributions. Leaf age and side significantly affected the quantitative composition of SCL. Increased day temperature during the development of leaves led to decreased amounts per unit area of n-alkanes, 1-alkanols, n-alkanoic acids and n-alkyl esters while increased night temperatures resulted in increased amounts of n-alkanes n-alkanoic acids and 1-alkanols. Relative humidity had no effect on the amounts or composition of SCL. The permeability of cuticular membranes to water (described in part I of this paper) and the composition of SCL were not related.
