The spin trap 5,5-dimethyl-l-pyrroline-l-oxide (DMPO) has been applied to monitor the generation of free oxy-radicals in samples derived from isolated hearts and heart cells. · OH was trapped in the effluent of isolated hearts in the early phase of reperfusion following an ischemia time of only 10min. Radical detection was possible even when the cardioactive DMPO was added to the effluent after draining off the heart, demonstrating that the short-lived · OH was generated by components released from the affected heart. These results support the hypothesis that radicals are of relevance for reperfusion injury.

By omitting antioxidants commonly used for incubation media of cultured cells, it was possible for the first time to demonstrate the formation of · OH in the incubation solution of cardiac cells.  相似文献

The relation between the effects of glucagon on oxygen consumption and glycolysis in livers from rats under different metabolic conditions was examined. Respiration of substrate-free perfused livers with different glycolytic fluxes, induced by changes in the pattern of food intake, responds differently to the infusion of 1 nM glucagon. The increases in oxygen uptake caused by 1 nM glucagon correlate reasonably well with the absolute decreases in glycolysis. The degree of inhibition of glycolysis is approximately constant (58 per cent) for all metabolic conditions. When no recovery of glycolysis occurs upon cessation of glucagon infusion, the same happens with oxygen consumption, which remains stimulated. It is concluded that in livers with no appreciable biosynthetic activities, the action of glucagon on respiration and glycolysis may be interpreted in terms of an interaction of interpreted in terms of an interaction of cytosolic and mitochondrial ATP generating processes.  相似文献

Attempts to increase European biodiversity by restoring rivers and floodplains are based on inadequate data on natural systems. This is particularly the case for NW European rivers because all catchments have been impacted by agriculture and river engineering. If river restoration is to have an ecological, as opposed to `cosmetic' design basis then baseline models are required. However, this poses three questions; (a) what is the natural river‐floodplain state, (b) how can it be defined and modelled and (c) can this state be recreated today? The first two questions can only be addressed by using palaeohydrological and palaeoecological data. A second and equally vital consideration is the stability/instability of any restored system to change in external forcing factors (e.g. climate) and in this context it may not be realistic to expect baseline models to provide equilibrium solutions but instead to define process‐form domains. Over the last two decades evidence has accumulated that the natural state of lowland rivers in much of NW Europe was multi rather than single thread‐braided, anastomosing or anabranching. Until recently our knowledge of floodplain palaeoecology was generally derived from pollen diagrams, which have source‐area of problems and lack of taxonomic specificity. The precision and breadth of palaeoecological reconstruction (including richness and structure) has been greatly increased by the use of multiple palaeo‐indicators including macrofossils, diatoms and beetles. The dynamics of small to medium sized, low‐energy, predeforestation floodplains were dominated by disturbance (windthrow, beavers, etc.) and large woody debris. In order to compare the hydrogeomorphological basis of floodplain ecology, both temporally and spatially, a simple index of fluvial complexity is presented. Palaeoecological and geomorphological investigations have the potential to provide in‐depth models of the natural range of channel conditions and sensitivity to external change that can be used to provide a scientific basis for floodplain restoration. There is also the possibility that floodplain‐channel restoration may be a valuable tool in the mitigation of future geomorphological change forced by climatic instability.  相似文献

The estuary of the River Kyrönjoki was echo sounded and, on the basis of these results the delta was delimited and areas of erosion and accumulation were separated. Samples of the sediment surface (0–2 cm) from 65 sites in the estuary were analysed for organic content, C, N, P, Ca, Fe, Mn, Pb, Cu, Zn, Cd and Hg. On the basis of the chemical analyses the delta was divided into two parts according to the sedimentary conditions: (I) In the inner part, where acid fresh water of c. pH 5.5 from the river is predominant, the conditions were reducing. The Fe/Mn ratio of the sediment was high. The heavy metal content was usually low. The Hg content in the organic compounds of the sediment was, due to the acidity, higher than in the outer part. (II) In the outer part of the delta, where neutral brackish water of pH c. 7.0 dominates, the sedimentary conditions were mostly oxidizing. The Fe/Mn ratio was very low. The heavy metal and organic content was mostly high in the accumulation areas. This is because the fine-grained allochthonous material, which has a high heavy metal and organic content, flocculates and precipitates when it reaches the brackish water of the Gulf of Bothnia, and the salinity and the pH are higher.  相似文献

Sub-optimal conditions in terms of maintenance ration and/or low temperature resulted in otolith bands, with distinct microstructural features, in response to each particular condition. Check(s) were not only produced corresponding to the transition from optimal to the sub-optimal conditions but also from sub-optimal to the optimal ones. Daily increment deposition, although faint and narrow, persisted in the otoliths of fish when they were kept at low temperatures and fed on maximum food rations. Observable increments were produced only in the first few days of the period when fish were fed on maintenance rations at the optimal temperature. There were only a few checks formed in the otoliths of fish under the conditions of low temperature and maintenance rations. Restraining fish growth in each of the above sub-optimal conditions boosted the growth of the fish and their otoliths when optimal conditions resumed.  相似文献

Evolutionary comparative methods taking into account the potential effects of relatedness reveal that, among 202 species of animal-dispersed tropical woody angiosperms from Peru, large seeds may be an adaptation to: (1) dispersal by mammals rather than by birds and (2) greater plant height. Using the most powerful techniques currently available, appropriate comparisons were made between related species or species groups at all taxonomic levels, thus allowing use of all species in the data set. Even so, the results show no evidence of adaptation (i.e., a functional relationship) of seed mass to successional syndrome or to growth form (trees or vines). In previous cross-species analyses, seed mass has been linked to both of these ecological variables. The lack of relationship between seed mass and habitat use or growth form may be due to targeting the wrong trait to predict these two variables, or because seed mass is subject to exaptation, i.e., a particular seed mass has arisen for other reasons, but allows invasion into a habitat and currently serves to maintain that species in the habitat. Differentiation between these two explanations for the lack of correlation between seed mass and successional syndrome or growth form would entail species-by-species examination of the function of seed mass under natural conditions. The effect of relatedness on the ecological correlates of seed mass and plant height is determined for each of three taxonomic schemes. It is concluded that the three schemes do not differ greatly in the results obtained.  相似文献

Isoprene emission from plants accounts for nearly half of all non‐methane hydrocarbons entering the atmosphere. Light and temperature regulate the instantaneous rate of isoprene emission but there is increasing evidence that they also affect the capacity for isoprene emission (i.e. the rate measured under standard conditions). We tested the rate of acclimation of the capacity for isoprene emission following step changes in growth conditions. Acclimation to new growth temperatures was very rapid, with most of the change occurring within a few hours and complete adjustment occurring within a day. Acclimation to new light levels was more complicated. Following a switch from low‐light growth conditions to standard assay conditions (30 °C and 1000 µmol photons m?2 s?1), there was a rapid (5–10 min) and a slightly slower (10–50 min) acclimation of the capacity for isoprene emission. After accounting for these short‐term changes, there was also a small, long‐term (4–6 d) acclimation of the isoprene emission capacity to the light level of growth conditions. We found no effect of growth conditions on the coefficients used to describe the instantaneous light and temperature response of isoprene emission. Therefore, current models of isoprene emission will only need to be altered to account for changes in the capacity for isoprene emission.  相似文献

Evolution is assumed to begin in a very particular compartmentalized location with periodic conditions. A highly diversified world is the driving force for the continuous increase in complexity by colonizing increasingly less favourable regions. Modeling the “origin-of-life” a Darwinian cyclic process is simulated (multiplication with sporadic errors followed by a construction and selection). Starting from a RNA-world (R-strands of R1 and R2 monomers building Hairpin-Assembler devices) and introducing another kind of monomers (A1 and A2 which interlink to the Hairpin-Assembler devices such that they become bound and form an A-oligomer) it is shown that a simple translation apparatus evolves producing enzymes (specific sequences of A1 and A2 monomers given by the sequences of R1 and R2 monomers on the assembler-strands). Later on D-strands are introduced, which are not capable of participating in the synthesis of A-oligomers. These D-strands become carriers of the genetic information and induce the formation of increasingly complex entities of functionally interplaying components.  相似文献

ABSTRACT

This review concerns the current knowledge of melatonin and alcohol-related disorders. Chronobiological effects of ethanol are related to melatonin suppression and in relation to inflammation, stress, free radical scavenging, autophagy and cancer risk. It is postulated that both alcohol- and inflammation-induced production of reactive oxygen species (ROS) alters cell membrane properties leading to tissue dysfunction and, subsequent further ROS production. Lysosomal enzymes are often used to assess the relationships between intensified inflammation states caused by alcohol abuse and oxidative stress as well as level of tissue damage estimated by the increased release of cellular enzymes into the extracellular space. Studies have established a link between alcoholism and desynchronosis (circadian disruption). Desynchronosis results from the disorganization of the body’s circadian time structure and is an aspect of the pathology of chronic alcohol intoxication. The inflammatory conditions and the activity of lysosomal enzymes in acute alcohol poisoning or chronic alcohol-dependent diseases are in most cases interrelated. Inflammation can increase the activity of lysosomal enzymes, which can be regarded as a marker of lysosomal dysfunction and abnormal cellular integrity. Studies show alcohol toxicity is modulated by the melatonin (Mel) circadian rhythm. This hormone, produced by the pineal gland, is the main regulator of 24 h (sleep-wake cycle) and seasonal biorhythms. Mel exhibits antioxidant properties and may be useful in the prevention of oxidative stress reactions known to be responsible for alcohol-related diseases. Naturally produced Mel and exogenous sources in food can act in free radical reactions and activate the endogenous defense system. Mel plays an important role in the normalization of the post-stress state by its influence on neurotransmitter systems and the synchronization of circadian rhythms. Acting simultaneously on the neuroendocrine and immune systems, Mel optimizes homeostasis and provides protection against stress.

Chlorella vulgaris MSU 01 strain isolated from the sediment of the pond is able to produce molecular hydrogen in a clean way. To relate the dynamic coupling between the cultural conditions and biological responses, an original lab scale set up has been developed for hydrogen production. Different sources like mannitol, glucose, alanine, citric acid, aspartic acid, l-alanine, l-cysteine, sodium succinate and sodium pyruvate were used for algal media optimization. Corn stalk, from 1 to 5 g/L was tested for the effective algal growth and hydrogen production. The cell concentration of 1.6-19 g/L dry cell weight (DCW) was found at the 10th day. The kinetic parameters involved in the hydrogen production at 4 g/L corn stalk using the algal inoculum (50 mL) in the bioreactor volume (500 mL) was found to be with the hydrogen production potential (Ps) of 7.784 mL and production yield of (Pr) 5.534 mL respectively. The growth profile of the algal biomass at the above mentioned condition expressed the logistic model with R2 0.9988. The final pH of the broth was increased from 7.0 to 8.5-8.7. The anaerobic fermentation by C. vulgaris MSU 01 strain involved in the conversion process of complex carbon source has increased the H2 evolution rate and higher butyrate concentration in the fermentate.  相似文献
