The diversity of abundant mRNA sequences in various parts of 4-d etiolated pea seedlings (Pisum sativum L. var. Rondo CB) was compared by a cell-free translation of the mRNAs in the presence of [35S]methionine and by an analysis of the products by two-dimensional electrofocussing/ electrophoresis (2D separation). The various parts of the seedlings were also examined for the pattern of protein synthesis in vivo. Proteins were labeled by injection of [35S]methionine into the cotyledons, followed by 2D separation of the products. Over 95% of the abundant mRNA sequences and newly synthesized abundant polypeptides were shared by all parts of etiolated seedlings, including the cotyledons. However, a few distinct differences were observed when comparing mRNAs of roots and shoots; the most prominent among these were a group of six abundant mRNA sequences found exclusively in shoots. Only about 30% of the polypeptides synthesized on isolated RNA could be traced in equivalent positions on the gels as the polypeptides synthesized in vivo. Analysis of total RNA from light-grown pea seedlings showed the appearance of some twenty-five translation products not found with total RNA from etiolated seedlings, while about nine other translation products disappeared. At least ten of the light-induced RNA sequences were also present after growth in low-intensity red light (>600 nm) and are therefore thought to be controlled by the phytochrome system. Comparison of 11-d light-grown pea plants with 4-d light-grown seedlings did not reveal additional translatable RNA sequences, indicating that the major morphogenetic changes that occur after 4 d are not accompanied by significant changes in the pattern of abundant RNA sequences.  相似文献

During the greening of etiolated rice leaves, total glutamine synthetase activity increases about twofold, and after 48 h the level of activity usually observed in green leaves is obtained. A density-labeling experiment with deuterium demonstrates that the increase in enzyme activity is due to a synthesis of the enzyme. The enhanced activity obtained upon greening is the result of two different phenomena: there is a fivefold increase of chloroplastic glutamine synthetase content accompanied by a concommitant decrease (twofold) of the cytosolic glutamine synthetase. The increase of chloroplastic glutamine synthetase (GS2) is only inhibited by cycloheximide and not by lincomycin. This result indicates a cytosolic synthesis of GS2. The synthesis of GS2 was confirmed by a quantification of the protein by an immunochemical method. A mutant of Halobacterium halobium which contains halorhodopsin was isolated from strain S9. An absorbance change at 380 nm caused by steady orange light illumination (λ ?530 nm) was observed. This change depended upon the intensity of the actinic light. The bleached envelope vesicles and vesicles derived from nicotine-grown cells showed a small or no absorbance change at 380 nm, suggesting that the change stemmed from the photochemical intermediate of halorhodopsin (referred to as P-380). When blue light was superimposed on orange background illumination, the membrane potential (Δψ) of the envelope vesicles decreased. Δψ was determined from the tetraphenylphosphonium cation (TPP+) distribution by means of a TPP+ electrode. When blue light intensity was increased, both Δψ and the amount of P-380 were decreased. An equation was derived which showed that Δψ is proportional to the concentration of P-380 formed by illumination under the assumption that the ionic composition is not significantly changed upon illumination. This equation was checked experimentally from the following three points: The blue light effect, the relationship between Δψ and light intensity, and the effect of gramicidin. The data obtained accorded well with the theoretical relationship.  相似文献

Myelin basic protein caused rapid aggregation of vesicles containing acidic phospholipids. Aggregation could be reversed by trypsin digestion of the myelin basic protein. Aggregated vesicles containing gel phase phospholipids or vesicles containing greater than 15 mol% lysolecithin underwent fusion. The extent of fusion was measured by irreversible changes in the light-scattering intensities or diffusion coefficients of the vesicles. Fusion was also measured by the fluorescence quenching which occurred when vesicles containing a covalently bound fluorophore, N-4-nitrobenzo-2-oxa-1,3-diazole, were fused with vesicles containing the covalently bound spin label, 4,4-dimethyl-oxazolidine-N-oxyl. The kinetics of fusion were first order in phospholipid and had half-times of 0.5–5 min depending on lysolecithin composition. This protein-enhanced membrane fusion may provide a valuable model system for studying some types of biological membrane fusions.  相似文献

A plethora of compounds stimulate protective mechanisms in plants against microbial pathogens and abiotic stresses. Some defense activators are synthetic compounds and trigger responses only in certain protective pathways, such as activation of defenses under regulation by the plant regulator, salicylic acid (SA). This review discusses the potential of naturally occurring plant metabolites as primers for defense responses in the plant. The production of the metabolites, hexanoic acid and melatonin, in plants means they are consumed when plants are eaten as foods. Both metabolites prime stronger and more rapid activation of plant defense upon subsequent stress. Because these metabolites trigger protective measures in the plant they can be considered as “vaccines” to promote plant vigor. Hexanoic acid and melatonin instigate systemic changes in plant metabolism associated with both of the major defense pathways, those regulated by SA- and jasmonic acid (JA). These two pathways are well studied because of their induction by different microbial triggers: necrosis-causing microbial pathogens induce the SA pathway whereas colonization by beneficial microbes stimulates the JA pathway. The plant’s responses to the two metabolites, however, are not identical with a major difference being a characterized growth response with melatonin but not hexanoic acid. As primers for plant defense, hexanoic acid and melatonin have the potential to be successfully integrated into vaccination-like strategies to protect plants against diseases and abiotic stresses that do not involve man-made chemicals.  相似文献

This article reviews the organization of the forebrain nuclei of the avian song system. Particular emphasis is placed on recent physiologic recordings from awake behaving adult birds while they sing, call, and listen to broadcasts of acoustic stimuli. The neurons in the descending motor pathway (HVc and RA) are organized in a hierarchical arrangement of temporal units of song production, with HVc neurons representing syllables and RA neurons representing notes. The nuclei Uva and NIf, which are afferent to HVc, may help organize syllables into larger units of vocalization. HVc and RA are also active during production of all calls. The patterns of activity associated with calls differ between learned calls and those that are innately specified, and give insight into the interactions between the forebrain and midbrain during calling, as well as into the evolutionary origins of the song system. Neurons in Area X, the first part of the anterior forebrain pathway leading from HVc to RA, are also active during singing. Many HVc neurons are also auditory, exhibiting selectivity for learned acoustic parameters of the individual bird's own song (BOS). Similar auditory responses are also observed in RA and Area X in anesthetized birds. In contrast to HVc, however, auditory responses in RA are very weak or absent in awake birds under our experimental paradigm, but are uncovered when birds are anesthetized. Thus, the roles of both pathways beyond HVc in adult birds is under review. In particular, theories hypothesizing a role for the descending motor pathway (RA and below) in adult song perception do not appear to obtain. The data also suggest that the anterior forebrain pathway has a greater motor role than previously considered. We suggest that a major role of the anterior forebrain pathway is to resolve the timing mismatch between motor program readout and sensory feedback, thereby facilitating motor programming during birdsong learning. Pathways afferent to HVc may participate more in sensory acquisition and sensorimotor learning during song development than is commonly assumed. © 1997 John Wiley & Sons, Inc. While investigations into shoot responses to elevated atmospheric CO2 are extensive, few studies have focused on how an elevated atmospheric CO2 environment might impact root functions such as water uptake and transport. Knowledge of functional root responses may be particularly important in ecosystems where water is limiting if predictions about global climate change are true. In this study we investigated the effect of elevated CO2 on the root hydraulic conductivity (Lp) of a C3 perennial, Larrea tridentata, and a C3 annual, Helianthus annuus. The plants were grown in a glasshouse under ambient (360 μmol mol–1) and elevated (700 μmol mol–1) CO2. The Lp through intact root systems was measured using a hydrostatic pressure-induced flow system. Leaf gas exchange was also determined for both species and leaf water potential (ψleaf) was determined in L. tridentata. The Lp of L. tridentata roots was unchanged by an elevated CO2 growth environment. Stomatal conductance (gs) and transpiration (E) decreased and photosynthetic rate (Anet) and Ψleaf increased in L. tridentata. There were no changes in biomass, leaf area, stem diameter or root : shoot (R : S) ratio for L. tridentata. In H. annuus, elevated CO2 induced a nearly two-fold decrease in root Lp. There was no effect of growth under elevated CO2 on Anet, gs, E, above- and below-ground dry mass, R : S ratio, leaf area, root length or stem diameter in this species. The results demonstrate that rising atmospheric CO2 can impact water uptake and transport in roots in a species-specific manner. Possible mechanisms for the observed decrease in root Lp in H. annuus under elevated CO2 are currently under investigation and may relate to either axial or radial components of root Lp.  相似文献

根据对巴山松针叶群体的水平和垂直变化特征及趋势进行的研究,得到如下结果:(1)单株木的针叶总数和各龄针叶总数随胸径增加而增多,随密度增加而减少;当年生针叶数量所占比例随径阶增加而增大,2年生针叶比例与此相反,而1年生和3年生针叶比例基本保持稳定。(2)单株木的径阶愈小,针叶平均寿命亦愈长;就针叶平均生存期限而言,劣势木最短,优势木次之,而平均木最长。(3)各龄级针叶群体的数量一般在树冠中部所占比例最大,下部所占比例次之,上部比例最小。在林分中处于不同地位的林木表现形式略有差异,与树冠内光照的分布有关。(4)针叶的内禀增长率随着径阶的增大、树高的增加及相对光照强度的增高而增加;树冠中针叶的平均寿命随树高增加而减小。另外,对产生上述结果的原因也进行了部分探讨。  相似文献

本文报道了两种土壤水分条件下羊草明呼吸速率与光照和温度的关系,以及暗呼吸速率与温度的关系。结果表明,羊草的明呼吸速率与光强呈非线性函数关系。在低光强下,明呼吸速率随光强升高而有较快的增加;随着光强的增高,其增加速度减慢。在温度低于羊草光合的高温补偿点的条件下,明呼吸速率在一定温度范围内随温度升高而增大;当温度达到一定限度时,有一个下降阶段,而后又回升,羊草的暗呼吸速率随温度增加而升高,且在一定限度内,其升高速度随温度增高而加快。当土壤干旱时,明呼吸速率显著降低,而暗呼吸速率仅略有减小。  相似文献

 We studied photosynthetic acclimation of eastern hemlock [Tsuga canadensis (L.) Carr.] seedlings in the first month after sudden exposure of shade-grown seedlings to full sunlight. In a greenhouse experiment, seedlings were grown under full sun or 80% shade, and after 7 months, a sample of the shaded trees was transferred to full sun in the greenhouse. Photosynthetic responses of shaded, transferred, and sun trees were followed over the course of 26 days to track short to medium-term acclimation responses. A partial acclimation of photosynthesis at high light occurred in pre-existing (formed in the previous environment) and new foliage of transferred seedlings. This was associated with non-stomatal limitations to photosynthesis. Pre-existing foliage of transferred plants had a prolonged reduction in the ratio of variable to maximal fluorescence, and a limited capacity to adjust photochemical quenching or photosystem II quantum yield in the light to increasing light intensity compared to sun foliage, and apparently had some difficulty sustaining non-photochemical quenching. Seedling survival was only 58% among transferred seedlings, compared to 80% and 100% in the shade or sun groups, respectively. Photosystem II quantum yield in the light, and photochemical and non-photochemical quenching were similar between newly formed foliage of transferred and sun plants. These findings indicate that eastern hemlock depends strongly on the production of new foliage for photosynthetic adjustments to high light, and that development of photosynthetic competence may be a gradual process that occurs over successive foliar production cycles.
