Transitions in growth irradiance level from 92 to 7 Em-2 s-1 and vice versa caused changes in the pigment contents and photosynthesis of Oscillatoria agardhii. The changes in chlorophyll a and C-phycocyanin contents during the transition from high to low irradiance (HL) were reflected in photosynthetic parameters. In the LH transition light utilization efficiencies of the cells changed faster than pigment contents. This appeared to be related to the lowering of light utilization efficiencies of photosynthesis. As a possible explanation it was hypothesized that excess photosynthate production led to feed back inhibition of photosynthesis. Time-scales of changes in the maximal rate of O2 evolution were discussed as changes in the number of reaction centers of photosystem II in relation to photosynthetic electron transport. Parameters that were subject to change during irradiance transitions obeyed first order kinetics, but hysteresis occurred when comparing HL with LH transients. ABSTRACT.
  • 1 Patterns of vertical habitat use of ten species of cockroaches are examined. Three assemblages of cockroaches are recognized on the basis of morphology, foraging behaviour, foraging heights, and overlaps and breadths of vertical distributions.
  • 2 Three apterous and brachypterous species occur near the ground and comprise one assemblage. They feed mainly on material in the leaf-litter.
  • 3 Species that perch higher either migrate into the leaf-litter on a diel basis and feed on both leaf-litter and epiphyllic materials, or some are strictly arboreal and forage on algae, liverworts, lichens, spores, pollen and trichomes on the surfaces of leaves.
  • 4 Trophic and behavioural correlations with perch height are described and the functions of perching are examined.
  • 5 We conclude that studies of interactions among species are confounded by our lack of understanding of stage- and sex-specific interactions of coexisting species. A simplistic ‘species’approach to such interactions is inadequate because it does not recognize intraspecific variation.

The mating activity of mating-type plus gametes of Chlamydomonas eugametos depends on light. Cells lost their ability to agglutinate with mating-type minus gametes after a dark period of 30 min. They regained their agglutinability after 10 min exposure to light. Other mating reactions, such as tipping and flagellar tip activation, were not dependent upon light. Since cycloheximide and tunicamycin did not affect the light-induced activation of flagellar agglutinability, no protein synthesis or glycosylation is involved in this process. Equal amounts of biologically active agglutination factor could be extracted from cells placed either in light or in darkness. A minor portion of the active material was found to be located on the flagellar surface of illuminated cells. No active material was found on the flagellar surface of dark-exposed cells, whereas their cell bodies contained the same amount of active material as the cell bodies of illuminated cells. Since a light-induced flow of agglutination factors from the cell body to the flagella could not be detected and dark-exposed cells could be slightly activated by amputation or fixation by glutaraldehyde, we propose that light affects flagellar agglutinability by an in-situ modification of the agglutination factor on the flagella. Phyllodia of the Crassulacean acid metabolism (CAM) plant Kalanchoë tubiflora were allowed to fix 13CO2 in light and darkness during phase IV of the diurnal CAM cycle, and during prolongation of the regular light period. After 13CO2 fixation in darkness, only singly labelled [13C]malate molecules were found. Fixation of 13CO2 under illumination, however, produced singly labelled malate as well as malate molecules which carried label in two, three or four carbon atoms. When the irradiance during 13CO2 fixation was increased, the proportion of singly labelled malate decreased in favour of plurally labelled malate. The irradiance, however, did not change either the ratio of labelled to unlabelled malate molecules found in the tissue after the 13CO2 application, or the magnitude of malate accumulation during the treatment with label. The ability of the tissue to store malate and the labelling pattern changed throughout the duration of the prolonged light period. The results indicate that malate synthesis by CAM plants in light can proceed via a pathway containing two carboxylation steps, namely ribulose-1,5-bisphosphate-carboxylase/oxygenase (EC 4.1.1.39) and phosphoenolpyruvate carboxylase (EC 4.1.1.31) which operate in series and share common intermediates. Glucose-6-phosphate dehydrogenase (D-glucose-6-phosphate: NADP oxidoreductase, EC 1.1.1.49) has been purified to electrophoretic homogeneity from pea chloroplasts. The enzyme, which has a Stokes radius of 52 Å, is a tetramer made up of four 56000 Da monomers. The pH optimum is around 8.2. The enzyme is absolutely specific for NADP. The apparent Km(NADP) is 2.4 ± 0.1 μM. NADPH inhibition of the enzyme is competitive with respect to NADP (mean Ki, 18 ± 5 μM) and is mixed (Kp >Km, Vmax >Vp) with respect to glucose 6-phosphate (mean crossover point, 0.5 ± 0.1 mM). The apparent Km(glucose 6-phosphate) is 0.37 ± 0.01 mM. The purified enzyme is inactivated in the light in the presence of dilute stroma and washed thylakoids, and by dithiothreitol. Enzyme which has been partially inactivated by treatment with dithiothreitol can be further inactivated in the light in the presence of dilute stroma and washed thylakoids and reactivated in the dark, but only to the extent of the reverse of light inactivation. Dithiothreitol-inactivated enzyme is not reactivated further by addition of crude stroma or oxidized thioredoxin. Dithiothreitol-dependent inactivation of the enzyme follows pseudo-first-order kinetics and shows rate saturation. The enzyme which has been partially inactivated by treatment with dithiothreitol does not differ from the untreated control with respect to thermal and tryptic inactivation. However, enzyme which has been partially light inactivated shows different thermal and tryptic inactivation patterns as compared to the dark control. These observations suggest that the changes in the enzyme brought about by light modulation are not necessarily identical with those brought about by dithiothreitol inactivation.  相似文献

The hypothesis that chloroplasts having different light-saturated rates of photosynthesis will have different proportions of the intrinsic thylakoid complexes engaged in light-harvesting and electron transport (Anderson, J.M. (1982) Mol. Cell. Biochem. 46, 161–172) has been tested. Peas were grown in light regimes which varied in light intensity, quality and time of irradiance, and ranged from sunlight through red to blue-enriched light of very low radiation. The electron-transport capacity at saturating light of Photosystem I and Photosystem II of chloroplasts isolated from light-adapted peas was 2-fold and 5–6-fold lower, respectively, in the lowest radiation compared to sunlight. There was a marked increase in the amount of total chlorophyll associated with the main chlorophyll ab-proteins (LHCP1, LHCP2 and LHCP3) and a 2-fold decrease in the core reaction centre complex of Photosystem II (CP a) as the radiation decreased; the LHCP1–3CP a ratio changed from 3.5 to 9.0. The amount of chlorophyll associated with Photosystem I varied from 34% in sunlight to 27% in the lowest radiation, but the antenna size of Photosystem I was not markedly different; there was a 2-fold decrease in the amount of cytochrome f on a chlorophyll basis, which partly accounted for the decreased electron-transport capacity of Photosystem I. Since the increases or decreases in the levels of each of the components correlated with decreasing radiation, it is clear that the light-adaptation of both light-harvesting and electron-transport components is indeed closely co-ordinated.  相似文献

Light Enhances the Turnover of Phosphatidylinositol in Rat Retinas   总被引:4,自引:1,他引:3  
Light stimulation of isolated rat retinas is shown to enhance the turnover of phosphatidylinositol (PI) as demonstrated by a light-dependent increase in [3H]inositol incorporation and concurrent hydrolysis of existing PI. Studies with rat retinas incubated with [3H]inositol and then microdissected at the level of the outer plexiform layer into photoreceptor cell and inner retina layers indicated that the light-enhanced incorporation of [3H]inositol was associated with the photoreceptor cell layer. The rate of PI hydrolysis in retinas prelabeled in vivo with [3H]inositol was higher in light than in dark incubations and was higher in the photoreceptor cell layer than within the inner retina. Within the photoreceptor cell layer, PI turnover involved 2%/min of the total PI contentin dark and 6–8%/min in light. In contrast to what has been reported for stimulus-enhanced turnover of PI in some tissues, this light-enhanced turnover of PI in the retina was not associated with detectable reductions in PI content. Parallel studies of sodium (22Na) uptake demonstrated that the photoreceptor cells remained functional during these incubations as they retained the capacity to restrict the entry of 22Na in light but not in dark.  相似文献

Summary The response of the cotton plant (Gossypium hirsutum L.) to 9.0 and 12.5 cm intra- and 75.0 and 96.5 cm inter-row spacing was studied under irrigation with saline water (5.5 dS/m, SAR 18). In general, the dry weight per plant matter, the leaf area, number of bolls and flowers, and yield per plant were all affected significantly by intra-row spacing but, not by inter-row spacing. However, on a unit area basis, they were affected only by the inter-row spacing. This indicates that competition between plants exists when intra-row spacing is reduced, whereas no significant competition occurs due to a decrease in the inter-row spacing. Although the effect of intra-row spacing on yield for a unit area was found to be not significant, the effect of inter-row spacing was significant. Yield in plots with the conventional spacing (96.5 cm between rows and 12.5 cm between plants in the row) was 4863 kg/ha, whereas the yield in plots with 75 cm between rows was about 23% higher (5974 kg/ha). Summary Changes in human costal cartilage with increasing age (2–81 years) have been studied in the optical and electron microscope using routine and histochemical techniques.Concurrent with increasing age, chondrocytes undergo degeneration which is characterized initially by the accumulation of lipidic material within cells and, subsequently, by the formation of a halo around degenerating chondrocytes. The halo material is composed of electron dense bodies, amorphous material, and collagen fibrils. Both electron dense bodies and the amorphous material are of cellular origin and they have similar histochemical responses.Using histochemical techniques in the optical and in the electron microscope, it has been shown that chondroitin sulfate decreases with increasing age, while a hyaluronidase resistant material (presumably keratan sulfate) increases, initially in the central zone, and subsequently in the peripheral zones. Hyaluronidase resistant material is minute or absent in the central zone of aged cartilage.The genesis of collagen fibrils progresses from thin unbanded collagen-like fibrils in the pericellular lacunae of chondrocytes in young specimens to thick fibrils (sometimes in excess of 0.5 ) with a period of 640 Å in ageing cartilage. Aggregation of collagen fibrils seems to be related at least initially to the preponderance of matrix granules and beaded filaments which have been shown to originate intracellularly in vacuoles formed in degenerating mitochondria. Both of these structures contain glycosaminoglycans and, with increasing age, glycosaminoglycans decrease while collagen fibrils aggregate. In old age, the amorphous material, and possibly the content of disrupting electron dense bodies, seem to give origin to some collagen fibrils. Summary The number of Golgi cells per unit volume was determined in different regions of the cerebellar cortex of man and of ten other mammals. Despite the general belief in the uniform architecture of the cerebellar cortex, regional differences in the distribution of Golgi cells were found. In the inferior parts of the vermis, the number of Golgi cells per unit volume is twice that in the corresponding hemispheres. In addition, there are differences between the anterior and inferior parts of the vermis. These differences are a feature of the cytoarchitecture of the cerebellum in man and all the investigated mammals. The ratio of Purkinje cells to Golgi cells was also determined and found to differ in different species. In man, this ratio is 11.5, while in the monkey and cat it is almost 11.9 and in the rat 13.3. These differences in the ratio of Purkinje cells to Golgi cells are discussed from the point of view of cerebellar evolution.Supported by the Deutsche Forschungsgemeinschaft.  相似文献
