Among different cancer immunotherapy approaches, bispecific antibodies (BsAbs) are of great interest due to their ability to recruit immune cells to kill tumor cells directly. Various BsAbs against Her2 tumor cells have been proposed with potent cytotoxic activities. However, most of these formats require extensive processing to obtain heterodimeric bispecific antibodies. In this study, we describe a bispecific antibody, BiHC (bispecific Her2-CD3 antibody), constructed with a single-domain anti-Her2 and a single-chain Fv (variable fragment) of anti-CD3 in an IgG-like format. In contrast to most IgG-like BsAbs, the two arms in BiHC have different molecular weights, making it easier to separate hetero- or homodimers. BiHC can be expressed in Escherichia coli and purified via Protein A affinity chromatography. The purified BiHC can recruit T cells and induce specific cytotoxicity of Her2-expressing tumor cells in vitro. The BiHC can also efficiently inhibit the tumor growth in vivo. Thus, BiHC is a promising candidate for the treatment of Her2-positive cancers.  相似文献

文章基于线粒体控制区基因序列和微卫星标记比较秦岭细鳞鲑(Brachymystax tsinlingensis Li)、黑龙江流域的尖吻细鳞鲑(Brachymystax lenok Pallas)和钝吻细鳞鲑(Brachymystax tumensis Mori)的分子遗传差异,为澄清其分类地位争议提供分子证据。结果表明:(1)扩增217个样本的mtDNA D-loop区序列,共获得45个单倍型,类群间无共享单倍型;基于单倍型构建的系统进化树显示三个细鳞鲑类群呈独立的支系;(2)基于14个呈多态性位点的遗传分化结果表明,秦岭细鳞鲑与尖吻或钝吻细鳞鲑之间的遗传距离均大于尖吻细鳞鲑和钝吻细鳞鲑之间的遗传距离;(3)基于线粒体D-loop和多态性微卫星位点计算出的遗传分化系数(FST)都远高于0.25表明三个类群间的遗传分化程度极高。这些结果表明,秦岭细鳞鲑与黑龙江流域细鳞鲑之间遗传分化程度高,结合前期发现秦岭细鳞鲑与黑龙江细鳞鲑类群有明显形态分化的研究结果及它们之间地理隔离已久的现状,研究初步判定秦岭细鳞鲑为独立物种,并建议以Brachymystax tsinlinge...  相似文献

对湖南省的油樟(Cinnamomum longepaniculatum (Gamble)N.Cho ex H.W.Li)资源作了调查。用色谱、色谱/质谱联用方法鉴定了油樟叶精油的43个化学成分,依主含成分不同可将其划分为6个不同的化学类型。其中,主含甲基丁香酚、龙脑、樟脑、芳樟醇和倍半萜烯的5个化学类型为首次在油樟中发现。  相似文献

The kinetics of the light-driven Cl? uptake pump of Synechococcus R-2 (PCC 7942) were investigated. The kinetics of Cl? uptake were measured in BG-11 medium (pHo, 7·5; [K+]o, 0·35 mol m?3; [Na+]o, 18 mol m?3; [Cl?]o, 0·508 mol m?3) or modified media based on the above. Net36Cl? fluxes (?Cl?o,i) followed Michaelis-Menten kinetics and were stimulated by Na+ [18 mol m?3 Na+ BG-11 ?Cl?max= 3·29±0·60 (49) nmol m?2 s?1 versus Na+-free BG-11 ?Cl?max= 1·02±0·13 (54) nmol m?2 s?1] but the Km was not significantly different in the presence or absence of Na+ at pHo 10; the Km was lower, but not affected by the presence or absence of Na+ [Km = 22·3±3·54 (20) mmol m?3]. Na+ is a non-competitive activator of net ?Cl?o,i. High [K+]o (18 mol m?3) did not stimulate net ?Cl?o,i or change the Km in Na+-free medium. High [K+]o (18 mol m?3) added to Na+ BG-11 medium decreased net ?Cl?o,i [18 mol m?3K+ BG-11; ?Cl?max= 2·50±0·32 (20) nmol m?2 s?1 versus BG-11 medium; ?Cl?max= 3·35±0·56 (20) nmol m?2 s?1] but did not affect the Km 55·8±8·100 (40) mmol m?3]. Na+-stimulation of net ?Cl?o,i followed Michaelis-Menten kinetics up to 2–5 mol m?3 [Na+]o but higher concentrations were inhibitory. The Km for Na+-stimulation of net ?Cl?o,i [K1/2(Na+)] was different at 47 mmol m?3 [Cl?]o (K1/2[Na+] = 123±27 (37) mmol m?3]. Li+ was only about one-third as effective as Na+ in stimulating Cl? uptake but the activation constant was similar [K1/2(Li+) = 88±46 (16) mmol m?3]. Br? was a competitive inhibitor of Cl? uptake. The inhibition constant (Ki) was not significantly different in the presence and absence of Na+. The overall Ki was 297±23 (45) mmol m?3. The discrimination ratio of Cl? over Br? (δCl?/δBr?) was 6·38±0·92 (df = 147). Synechococcus has a single Na+-stimulated Cl? pump because the Km of the Cl? transporter and its discrimination between Cl? and Br? are not significantly different in the presence and absence of Na+. The lithium dendrite, inducing short circuit and breaking solid electrolyte interphase (SEI) films, is deleterious to the stability of Li metal batteries due to the uncontrollable occurrence of miscellaneous stresses. In contrast to conventional suppression routes, herein a strategy is proposed via controlling SEI film broken regions to minimize releasing stress in terms of weaving lithium pits. Inspired by the principle of zippers, zipper‐like SEI films enable offering ordered pattern on the surface of Li anode via mechanical rolling. For the available cells, net‐like sewing/breaking patterns alternatively occur in Li plating/stripping. In the same electrolyte, a stable and dendrite‐free Li homogeneous growth is achieved.  相似文献

Although the rechargeable lithium–oxygen (Li–O2) batteries have extremely high theoretical specific energy, the practical application of these batteries is still limited by the instability of their carbon‐based air‐electrode, Li metal anode, and electrodes, toward reduced oxygen species. Here a simple one‐step in situ electrochemical precharging strategy is demonstrated to generate thin protective films on both carbon nanotubes (CNTs), air‐electrodes and Li metal anodes simultaneously under an inert atmosphere. Li–O2 cells after such pretreatment demonstrate significantly extended cycle life of 110 and 180 cycles under the capacity‐limited protocol of 1000 mA h g?1 and 500 mA h g?1, respectively, which is far more than those without pretreatment. The thin‐films formed from decomposition of electrolyte during in situ electrochemical precharging processes in an inert environment, can protect both CNTs air‐electrode and Li metal anode prior to conventional Li–O2 discharge/charge cycling, where reactive reduced oxygen species are formed. This work provides a new approach for protection of carbon‐based air‐electrodes and Li metal anodes in practical Li–O2 batteries, and may also be applied to other battery systems.  相似文献
