The stomach contents were analyzed monthly for each year-class to elucidate the foraging pattern of bluegills in a small vegetated lake by the frequency occurrence and the points methods. Seasonal dietary changes of the year-classes were considered comparing the monthly fluctuations in abundance of major prey organisms. Though these bluegills are dietary generalists and opportunists like those in North America, their foraging pattern was characterized by a relatively clearer dietary shift during ontogeny and a wider food niche including piscivorous than those of bluegills with congeners in their home land. Therefore this finding provides evidence of the ecological release caused by the absence of congeners.  相似文献

Ischemia-reperfusion (I/R) injury is a complex process involving the generation and release of inflammatory cytokines, and the accumulation and infiltration of neutrophils and macrophages, which disturbs the microcirculatory hemodynamics. Nonetheless, ischemic preconditioning (IPC) is known to produce immediate tolerance to subsequent prolonged I/R insults, although its underlying mechanism largely remains unknown. Our study investigated the role of the IB--NF-B-TNF- (tumor necrosis factor-) pathway in IPC's ability to ameliorate I/R-induced microcirculatory disturbances in rat cremaster muscle flaps. Male Sprague-Dawley rats were randomized (n=8 per group) into 3 groups: a sham-operated control group, an I/R group (4 h of pudic epigastric artery ischemia followed by 2 h of reperfusion), and an IPC+I/R group (3 cycles of 10 min of ischemia followed by 10 min reperfusion before I/R). Intravital microscopy was used to observe leukocyte/endothelial cell interactions and quantify functional capillaries in cremaster muscles. I/R markedly increased the number of rolling, adhering, and migrating leukocytes. It was also observed that I/R significantly increased TNF- expression in these injured tissues. On the other hand, IPC prevented I/R-induced increases in leukocyte rolling, adhesion, and transmigration. Moreover, TNF- protein production and its mRNA expression were downregulated in the IPC group. Finally, I/R-induced IB- phosphorylation and NF-B (p65) nuclear translocation were both suppressed by IPC. These results indicated that IPC attenuated NF-B activation and subsequently reduced TNF- expression, which resulted in the amelioration of microcirculatory disturbances in I/R-injured cremaster muscles.  相似文献

The solution conformation of a designed tetradecapeptide Boc‐Val‐Ala‐Leu‐Dpg‐Val‐Ala‐Leu‐Val‐Ala‐Leu‐Dpg‐Val‐Ala‐Leu‐OMe (Dpg‐14) containing two di‐n‐propyl glycine (Dpg) residues has been investigated by 1H NMR and circular dichroism in organic solvents. The peptide aggregates formed at a concentration of 3 mM in the apolar solvent CDCl3 were broken by the addition of 12% v/v of the more polar solvent DMSO‐d6. Successive NiH Ni+1H NOEs observed over the entire length of the sequence in this solvent mixture together with the observation of several characteristic medium‐range NOEs support a major population of continuous helical conformations for Dpg‐14. Majority of the observed coupling constants ( ) also support ? values in the helical conformation. Circular dichroism spectra recorded in methanol and propan‐2‐ol give further support in favor of helical conformation for Dpg‐14 and the stability of the helix at higher temperature. The development and the possible mechanism of the chitosan-induced resistance to viral infection were investigated in potato plants. The plants were sprayed with a solution of chitosans (1 mg/ml) with the mol wt of 3, 36, and 120 kD. After 1, 2, 3, or 4 days, the treated leaves were cut off and mechanically infected with the potato virus X (PVX). The disks cut out from the inoculated leaves were used for determining virus accumulation, callose content, and ribonuclease and -1,3-glucanase activities. In another set of experiments, the plants were infected with PVX within 1, 4, or 8 days after chitosan treatment, and the number of systemically infected plants was determined. It was found that, a day after treatment, the plants acquired a resistance to viral infection. The disks from the chitosan-treated leaves, as compared to the control, accumulated less amount of virus. The chitosan treatment also significantly decreased the number of systemically infected plants as compared to the control. After 2–3 days, the resistance disappeared or even gave way to an increased susceptibility to the infection; subsequently, the resistance increased again. The extent of the resistance correlated with the callose content and the level of ribonuclease activity observed on the infection day. The resistance towards the infection with PVX is probably mediated by the callose and ribonuclease induction. The cultivation of test-tube potato plants from the cuttings previously infected with PVX on the chitosan-containing nutrient medium did not eradicate the viral infection from the plants.  相似文献

Antibody Z13e1 is a relatively broadly neutralizing anti-human immunodeficiency virus type 1 antibody that recognizes the membrane-proximal external region (MPER) of the human immunodeficiency virus type 1 envelope glycoprotein gp41. Based on the crystal structure of an MPER epitope peptide in complex with Z13e1 Fab, we identified an unrelated protein, interleukin (IL)-22, with a surface-exposed region that is structurally homologous in its backbone to the gp41 Z13e1 epitope. By grafting the gp41 Z13e1 epitope sequence onto the structurally homologous region in IL-22, we engineered a novel protein (Z13-IL22-2) that contains the MPER epitope sequence for use as a potential immunogen and as a reagent for the detection of Z13e1-like antibodies. The Z13-IL22-2 protein binds Fab Z13e1 with a Kd of 73 nM. The crystal structure of Z13-IL22-2 in complex with Fab Z13e1 shows that the epitope region is faithfully replicated in the Fab-bound scaffold protein; however, isothermal calorimetry studies indicate that Fab binding to Z13-IL22-2 is not a lock-and-key event, leaving open the question of whether conformational changes upon binding occur in the Fab, in Z13-IL-22, or in both.  相似文献

With the advent of molecular phylogenies the assessment of community assembly processes has become a central topic in community ecology. These processes have focused almost exclusively on habitat filtering and competitive exclusion. Recent evidence, however, indicates that facilitation has been important in preserving biodiversity over evolutionary time, with recent lineages conserving the regeneration niches of older, distant lineages. Here we test whether, if facilitation among distant-related species has preserved the regeneration niche of plant lineages, this has increased the phylogenetic diversity of communities. By analyzing a large worldwide database of species, we showed that the regeneration niches were strongly conserved across evolutionary history. Likewise, a phylogenetic supertree of all species of three communities driven by facilitation showed that nurse species facilitated distantly related species and increased phylogenetic diversity.  相似文献

基于CRISPR/Cas9n double nick技术构建人DNAH2(Homo sapiens dynein,axonemal,heavy chain 2)基因敲除的U2OS稳定细胞株,旨在研究DNAH2基因的生物学功能。首先设计并合成A、B两个sg RNA(Single guide RNA)以及各自的互补链,退火连接形成DNAH2 sg RNA-A、B双链,再分别与带有BbsⅠ粘性末端的p X462线性载体相连,形成p X462-DNAH2-A、p X462-DNAH2-B重组真核表达质粒。质粒共转染至U2OS细胞后,加入嘌呤霉素,以有限稀释法获得阳性单克隆细胞株,再以蛋白印迹实验检测DNAH2蛋白的表达,最后通过PCR-基因测序技术分析突变特点。结果显示A、B sg RNA双链成功插入p X462载体,U2OS-DNAH2-KO单克隆细胞株中DNAH2蛋白不表达,DNAH2基因发生移码突变,从而证实利用CRISPR/Cas9n double nick系统成功构建人DNAH2基因敲除的U2OS稳定细胞株,为研究DNAH2基因提供有利工具。  相似文献

Trienoic fatty acids, namely -linolenic acid and hexadecatrienoic acid, present in leaf lipids are produced by -3 fatty acid desaturases located in the endoplasmic reticulum and plastid membranes. The changes in the level of trienoic fatty acids during leaf maturation were investigated in wild-type plants of Arabidopsis thaliana (L.) Heynh. and in the fad7 mutant deficient in the activity of a plastid -3 desaturase. The levels of trienoic fatty acids increased in 26 °C- and 15 °C-grown wild-type plants with maturation of leaves. The increase in trienoic fatty acids was mainly due to galactolipids enriched in plastid membranes. In addition, the relative levels of trienoic fatty acids in major glycerolipids, including phospholipids enriched in the endoplasmic reticulum membranes, also increased with leaf maturation. By contrast, when the fad7 mutant was grown at 26 °C, the relative levels of trienoic fatty acids in individual lipids decreased with leaf maturation. The decreases in the levels of trienoic fatty acids, however, were alleviated when the fad7 mutant was grown at 15 °C. This study was undertaken in order to understand thebehaviour of airborne pollen grains, namely therelationship between their concentrations andconcomitant meteorological parameters, at differentaltitudes (ground level, 200, 400 and 600 m) of thelower atmosphere and its daily variations over onesite (the experimental farm of the University ofLeón). The experimental design involved a tetheredballoon (2.25 m3), an ADAS remote meteorologicalstation and an original radio controlled three headedpollen and spore sampler (called GABIS) using theRotorod design. Each head sampled a differentaltitude, while two control samples were taken atground level. Twenty-one takeoffs could be realized inthe 5 weeks period between end of May and end of June1997. Sampling was done early in the morning and sampling time at each altitudewas of 15 minutes. Results show that 45 differenttypes of pollen grains could be collected at this timeof the year and that significant variations could beobserved in the behaviour of the pollen cloud on a dayto day basis, probably because of differentmeteorological situations. Contrary to what isgenerally believed, pollen was in most cases moreabundant at higher altitude – on average by 30% ascompared to ground level – making evident anaerobiological layer of transport at about 500 m aboveground. This was especially the case for trees (Quercus and Castanea). The atmosphere MixingRatio was the most explicative factor at 200 m, whiletemperature dominated significance analysis at both400 and 600 m.  相似文献
