Sera from 287 sheep were screened for cytotoxic antibodies against sheep lymphocytes. Forty four antisera were selected which provisionally define 13 lymphocyte antigens. The frequency of these antigens was studied in 305 sheep from 8 flocks of different breeds. Family studies confirm that inheritance of sheep lymphocyte antigens is controlled by the autosomal codominant genes of at least 2 linked loci.  相似文献

A horse family in which a recombination occurred in the chromosome region coding for the serological specificities of the ELA complex and those of the A blood group system of a mare was further analysed by mixed lymphocyte reaction (MLR) and Southern blot hybridization. This family consisted of a stallion, a mare and five full sibs. The stallion and the mare were heterozygous for internationally recognized ELA specificities while only the mare was heterozygous for the A blood group system. MLR between all members of the family confirmed that the stallion possessed two different ELA haplotypes and suggested that recombination in the mare occurred outside the segment delimited by the ELA-A locus and the MLR region. DNA samples from all individuals were investigated by Southern blot analysis using three restriction enzymes (EcoRI, HindIII or TaqI), three human HLA probes (one of class I cDNA and two of class II probes), one cDNA (DR beta) and one genomic (DQ alpha). Class I and class II restriction fragments of the mare segregated in accordance to the ELA specificities and thus clearly confirming that the crossing-over did not occur between the ELA-A gene and the class I, class II region nor between DR beta and DQ alpha subsets. The A blood group genetic determinants would thus be situated outside the ELA region defined by class I and class II genes.  相似文献

For chemical synthesis of a gene coding for human necrosis factor α (TNF-α), DNA sequence predicted by the amino acid sequence of human TNF molecule was prepared. Codons were chosen according to the codon usage in Escherichia coli (E. coli). The 490 hp gene was assembled by enzymic ligation of 42 oligonucleotides and was cloned into vector (]KK223-3) for high expression of active TNF-α in E. coli. With use of site-directed mutagenesis on this DNA, five different muteins of TNF-α were synthesized. TNF-M1 and TNF-M4 have deletions of His-73 and Gln-102, respectively. These deletions didn't cause loss of the cytotoxic activity against L929 cells. TNF-M5, which has a substitution of Asp-10 to Arg, had the similar cytotoxic activity to that of TNF-α. The cytotoxic spectra against several tumor cells were not changed by this substitution. TNF-M3 has an amino acid substitution of Glu-116 to His which occupies this position in human TNF-β. This substitution didn't change the cytotoxicity. In addition, evidence was presented that the change of the carboxyl terminal residue doesn't always influence the cytotoxic activity of TNF-α. Many different muteins were also isolated by random mutagenesis with hydroxylamine-HCl. One of the muteins, which carries a mutation of His-15 to Tyr, lost the cytotoxic activity almost completely.  相似文献

Summary. We tested 53 cattle with ocular squamous cell carcinoma (cancer-eye) and 53 paired, matched controls for 25 class I antigens of the bovine major histocompatibility system. The most common antigen was W5 which was present in 40% of the animals with cancer-eye and 36% of the controls. There were no statistically significant differences in BoLA antigen frequency between cattle with and cattle without cancer-eye.  相似文献

Hydrogen peroxide (H2O2) is a reactive oxygen species that signals between cells, and H2O2 signaling is essential for diverse cellular processes, including stress response, defense against pathogens, and the regulation of programmed cell death in plants. Although plasma membrane intrinsic proteins (PIPs) have been known to transport H2O2 across cell membranes, the permeability of each family member of PIPs toward H2O2 has not yet been determined in most plant species. In a recent study, we showed that certain isoforms of Arabidopsis thaliana AtPIPs, including AtPIP2;2, AtPIP2;4, AtPIP2;5, and AtPIP2;7, are permeable for H2O2 in yeast cells. Since the expression of PIPs is differently modulated in Arabidopsis by abiotic stress or H2O2 treatment, it is important to investigate the integrated regulation of aquaporin expression and their physiological significance in H2O2 transport and plant response to diverse abiotic stresses.  相似文献

BALB/c mice are sensitive to Leishmaniamajor infection, while C57BL/6 mice are resistant and able to mount an effective immune response against the parasite. Since the secreted antigens of L. major suppress the proliferation of BALB/c mice lymphocytes in vitro, we analyzed their effects on the immune system of resistant C57BL/6 mice. Secreted antigens were semi-purified and two fractions with immunosuppressive activity were isolated. 15 μg/ml of fraction could suppress 60% of lymphocyte proliferation and prevent the stimulated lymphocytes entering from G1 phase into the S phase of the cell cycle. These fractions decreased the production of IFN-γ, increased IL-4 level in the lymphocyte culture and down-regulated the nitric oxide production by activated macrophages. These results may suggest that L. major parasite by secreting immunosuppressive factors could down-regulate the immune system of both sensitive and resistant mice for own survival advantage.  相似文献

We investigated the effect of copper (Cu) on circadian rhythms in red seabream, Pagrus major, under various concentrations of Cu (10, 20, 30 and 40 μg/L). To examine variability in circadian rhythms, we measured changes in the period 2 (Per2), cryptochrome 1 (Cry1), serotonin and arylalkylamine N-acetyltransferase (AANAT2) proteins. We found that circadian rhythm-related plasma proteins were significantly lower in a high-Cu environment (30–40 μg/L) than in low-Cu concentration (0, 10, or 20 μg/L). Our results indicate that environmental Cu at concentrations greater than 30 μg/L can have deleterious effects on fish circadian rhythms.  相似文献

While anthropogenic land‐use changes threaten wildlife globally, some species take advantage of such changes and disperse into urban areas. The wildlife in urban areas often promotes conflicts with humans, notably when the animals are associated with the spread of zoonotic diseases. In Israel, current urban invasion of rock hyraxes (Procavia capensis) draws public attention, since the species is a reservoir host of cutaneous leishmaniasis, a serious skin disease. The rock hyrax, however, has seldom been studied in densely populated areas, and the drivers for its urban expansion, as well as its abilities to live and spread in core urban areas, are relatively unknown. Here, we explore the rock hyrax expansion to urban areas process by examining the availability, characteristics and use of shelter along an urban gradient. Our findings suggest that a series of factors determines shelter availability and quality for the rock hyrax, which facilitates its dispersion across the urban gradient. We found that rock hyraxes from the Judean Desert expand to the peri‐urban region of Jerusalem by colonizing new rocky shelters formed as by‐products of urban development. With their populations reaching extreme densities in this area and saturating the available shelters, there is some spill over to the adjacent core urban areas where they colonize littered sites, which are made available due to the local socio‐economic conditions and cultural norms of waste disposal and illegal placement of temporary structures. Our work emphasizes the significance of the urban gradient approach for studying the mechanisms promoting wildlife expansion to cities. Our findings suggest that changes in shelter availability and quality due to urban development, and cultural norms promote shifts of the hyrax population by pushing from the already established areas and pulling into new environment across the urban gradient.  相似文献

Doubling the atmospheric CO2 concentration from 350 to 700 μ1 1?1 increased the relative growth rate (RGR) of hydroponically grown Urtica dioica L. and Plantago major ssp. pleiosperma Pilger only for the first 10–14 days. Previous experiments with P. major led to the conclusion that RGR did not respond in proportion to the rate of photosynthesis. The present paper is focussed on the analysis of the impact of changes in leaf morphology, dry matter partitioning, dry matter chemical composition and ontogenetic drift on this discrepancy. Soon after the start of the treatment, carbohydrate concentrations were higher at elevated CO2: a reaction that was largely due to starch accumulation. An increase in the percentage of leaf dry matter and decreases in the specific leaf area (SLA) and the shoot nitrogen concentration were correlated with an increase in the total nonstructural carbohydrate concentration (TNC). A combination of accumulation of soluble sugars and starch and ontogenetic drift explains the decrease in SLA at the elevated CO2 level. A similar ontogenetic effect of elevated CO2 was observed on the specific root length (SRL). Other variables such as shoot nitrogen concentration and percentage leaf dry matter were not affected by correction of data for TNC levels. The net diurnal fluctuation of the carbohydrate pool in P. major was equal for both CO2 concentrations, indicating that the growth response to elevated CO2 may be ruled by variables other than photosynthesis, as for instance sink strength. Elevated CO2 did not greatly influence the partitioning of nitrogen between soluble and insoluble, reduced N and nitrate, nor the allocation of dry matter between leaf. stem and root. The finding that the root to shoot ratio (R/S) was not affected by elevated CO2 implies that, in order to maintain a balanced activity between roots and shoot, no shift in partitioning of dry matter upon doubling of the atmospheric CO2 concentration is required. Our data on R/S are in good agreement with the response of R/S to high CO2 predicted by models based on such a theorem.  相似文献
