着床期小鼠子宫内膜Le~y糖蛋白的动态变化

为了解子宫内膜Ｌｅ~ｙ糖蛋白在胚泡着床期间的变化,以及与胚泡表面阶段特异性Ｌｅ~ｙ抗原出现的关系,应用对Ｌｅ~ｙ寡糖特异的ＡＨ－６单抗为探针,通过ＳＤＳ－ＰＡＧＥ和Ｗｅｓｔｅｒｎｂｌｏｔ免疫酶标染色,观察了小鼠子宫内膜Ｌｅ~ｙ糖蛋白在着床期的动态变化和分布特点。结果表明：（１）未孕及着床前后的小鼠子宫内膜均含Ｌｅ~ｙ糖蛋白Ｍｒ５０～２００ｋＤ）,未孕内膜的含量明显高于着床期间的样品;（２）在着床日（Ｄ４）样品可见微量１６ｋＤ的Ｌｅ~ｙ糖蛋白条带;（３）未孕至Ｄ４的子宫内膜均有Ｌｅ~ｙ糖蛋白分泌至宫腔,但呈渐减趋势,未孕样品Ｌｅ~ｙ糖蛋白主要为分泌型,而Ｄ４样品Ｌｅ~ｙ糖蛋白主要存在于子宫内膜,宫腔液含量很少;结果提示子宫内膜Ｌｅ~ｙ糖蛋白在着床期间具有含量、组成和分布上的动态变化,这些变化可能与胚泡着床以及胚泡在宫腔内其表面Ｌｅ~ｙ糖抗原转为阳性密切有关。     

Specificity of DC-SIGN for mannose- and fucose-containing glycans

The dendritic cell specific C-type lectin dendritic cell specific ICAM-3 grabbing non-integrin (DC-SIGN) binds to "self" glycan ligands found on human cells and to "foreign" glycans of bacterial or parasitic pathogens. Here, we investigated the binding properties of DC-SIGN to a large array of potential ligands in a glycan array format. Our data indicate that DC-SIGN binds with K(d)<2muM to a neoglycoconjugate in which Galbeta1-4(Fucalpha1-3)GlcNAc (Le(x)) trisaccharides are expressed multivalently. A lower selective binding was observed to oligomannose-type N-glycans, diantennary N-glycans expressing Le(x) and GalNAcbeta1-4(Fucalpha1-3)GlcNAc (LacdiNAc-fucose), whereas no binding was observed to N-glycans expressing core-fucose linked either alpha1-6 or alpha1-3 to the Asn-linked GlcNAc of N-glycans. These results demonstrate that DC-SIGN is selective in its recognition of specific types of fucosylated glycans and subsets of oligomannose- and complex-type N-glycans.     

Extended Holstein small polaron model for charge transfer in dry DNA

In this paper, the charge transfer problem in dry DNA was investigated by employing an extended Holstein small polaron model with external potential traps being involved in consideration. The ground state energy and the probability amplitude of polaron in various DNA chains with different external trap potentials were obtained by variational method with the trial function being taken in coherent state form. The stability of transfered charges in various circumstances was discussed accordingly.     

Les sclérobiontes des huîtres du Cénomanien supérieur du Mans (Sarthe,France)

Oysters of the Marnes à Pycnodonte biauriculata Formation, Upper Cenomanian (Guerangeri biozone) were significantly sampled in building works within the town of Le Mans (Sarthe, France). They are colonized by episkeletobionts (encrusting organisms) and endoskeletobionts (boring and bioeroding organisms). First ones include bivalves, polychaetes, foraminiferans and bryozoans. Second ones are represented by about fifteen ichnotaxa, the producers of which, when they are known, belong to ten taxa. The oysters, some of which have a large fixation surface, are allochtonous or at least parautochtonous. The settlement of sclerobionts began when the oysters were alive (Entobia isp.) and continued post-mortem on isolated valves, transported in their final depositional environment (Gnathichnus pentax). The significance of the coexistence of both Entobia and Gnathichnus ichnocoenosis is discussed.     

Paranthodon africanus (broom) a stegosaurian dinosaur from the Lower Cretaceous of South Africa

The holotype of Paranthodon africanus (Broom) from the Kirkwood Formation (Lower Cretaceous) of the Algoa Basin, Cape Province, South Africa consists of a partial skull, the bones of which are very similar to those of Stegosaurus. Both sides of the maxillary tooth crowns have a bulbous cingulum and a very prominent central vertical ridge above the large apical denticle that is bordered anteriorly and posteriorly by four to seven smaller denticles. Diagnostically stegosaurian material is also known from the Lower Cretaceous of England (Craterosaurus) and China (Wuerhosaurus) and the Upper Cretaceous of India (Dravidosaurus).     

高效液相法测定盐酸乐卡地平片含量

使用高效液相色谱法测定乐卡地平片含量,流动相为乙腈-0.O1 mol/L乙酸铵溶液—三乙胺(650:350:1)(pH 6.0).结果显示盐酸乐卡地平在浓度为2.05～404.0μg/mL范围内具有良好的线性关系,盐酸乐卡地平片剂的平均标示量含量为100.3％,符合要求.研究表明HPLC色谱法对乐卡地平片剂进行含量测定...     

胚泡表面Le^Y寡糖对小鼠胚泡表皮生长因子及其受体表达和分泌的影响

在胚胎与子宫内膜间识别、粘附及胚胎植入过程中 ,阶段特异表达的寡糖抗原LeY 起重要作用。经与特异性单克隆抗体AH6预保温 ,中和胚泡表面LeY 寡糖后 ,通过RT PCR和免疫印迹方法 ,观察了LeY 寡糖对着床前小鼠胚泡表皮生长因子及其受体的表达和分泌的影响。结果显示 ,在体外培养中 ,经AH6封闭胚泡表面的LeY寡糖后 1.5h ,胚泡EGF的转录及分泌明显受到抑制 (P <0 .0 1) ,并且这种抑制作用持续到 6h以上 ;而EGF R表达及分泌仅有轻微降低的趋势。结果提示 ,EGF表达和分泌的降低可能是胚泡表面LeY 寡糖调节胚泡自身发育及随后的着床过程的多种途径之一     

Identification and estimation of the proportion of limiting cell types involved in the phytohemagglutinin response by limiting cell dose-response analysis

Fluoresceinated peanut agglutinin (PNA-FITC) and a monoclonal anti-T-cell antibodies were used to identify human thymocyte subpopulations. The phenotypes of PNA+ and PNA? thymocytes were studied using two different techniques: agglutination with PNA and double-labeling immunofluorescence. The mitogen-responsive PNA- subset was shown to include early thymocytes bearing the OKT9 antigen as well as lymphocytes with the OKT3 phenotype. Nearly all PNA+ thymocytes were found to bind simultaneously OKT4, OKT6, and OKT8 antibodies, whereas about 30% of them weakly bind the OKT3 antibody. These data suggest the existence of several intermediate stages of intrathymic differentiation, including a subset simultaneously bearing the OKT6- and OKT3-defined antigens.
