目的对循环水养殖系统水质指标和刺参幼参的生长代谢进行分析。方法在水温(16±1)℃的循环养殖系统中投喂不同的微生态制剂。结果30d实验结果表明:循环养殖系统具有一定自净能力,微生态制剂能够显著延长循环养殖系统的自净周期。其中“EM菌剂”在控制溶解氧含量等方面具有优势。结论复合微生态制剂“海微净水剂”和“hEM菌剂”在降解氨氮、亚硝酸盐、磷酸盐和化学耗氧量等方面效果较单一微生态制剂“EM菌剂”效果好,并且刺参的生理活动也较强。此外“NEM菌剂”能够显著提高刺参幼参的生长速度。  相似文献

Proanthocyanidin (PA) and anthocyanin accumulation and location in developing leaves, flowers, and seeds of the legumes Medicago sativa, Lotus japonicus, Lotus uliginosus, Hedysarum sulfurescens, and Robinia pseudacacia were investigated by quantitative measurements and by histological analysis after staining with 1% vanillin/HCl, butanol/HCl, or 50% HCl. M. sativa leaves and flowers, L. japonicus leaves, and R. pseudacacia flowers do not contain PAs, but seeds of all investigated species contain PAs. Anthocyanins are absent in the seed coats of all five species and in leaves of L. japonicus. PA content generally increases as a function of development in leaves, but declines in flowers. With the exception of H. sulfurescens, flower PAs are synthesized in the parenchyma cells of the standard petal, while anthocyanins are located in the neighboring epidermal cells. Leucocyanidin reductase (LCR) catalyzes the conversion of 2,3-trans-3,4-cis-leucocyanidin to (+)-catechin and is the first enzyme in the PA-specific pathway. LCR activity was only detected in PA-containing tissues and generally declined during tissue development.  相似文献

为了解不同细菌刺激后仿刺参(Apostichopus japonicus)体腔液中免疫因子的应答变化,分别用灿烂弧菌(Vibrio splendidus)、哈维氏弧菌(V.harveyi)、假交替单胞菌(Pseudoalteromonas nigrifacien)、溶壁微球菌(Micrococcus lysodeikticus)和停乳链球菌(Streptococcus dysgadysgalactiae)注射刺激仿刺参,然后分别采用对硝基苯基磷酸酯(p NPP)底物法、氯化硝基四氮唑蓝(NBT)法、溶壁微球菌粉法和多巴络合物生成法对体腔液上清中的酸性磷酸酶(ACP)与碱性磷酸酶(AKP)、超氧化物歧化酶(SOD)、溶菌酶(LYZ)和酚氧化酶(PO)的活力进行了测定。结果显示,灿烂弧菌刺激后,酸性磷酸酶和碱性磷酸酶活力显著升高,而超氧化物歧化酶、溶菌酶和酚氧化酶活力显著降低;哈维氏弧菌刺激后,酸性磷酸酶、超氧化物歧化酶、溶菌酶和酚氧化酶活力显著升高,碱性磷酸酶活力变化不规律;假交替单胞菌刺激后,酸性磷酸酶、溶菌酶和酚氧化酶活力显著升高,超氧化物歧化酶活力先升高后降低,碱性磷酸酶活力变化不规律;溶壁微球菌刺激后,酸性磷酸酶和酚氧化酶活力显著升高,超氧化物歧化酶活力先升高后降低,溶菌酶活力先升高后降低,而后在72 h恢复至对照水平,碱性磷酸酶活力变化不规律;停乳链球菌刺激后,除碱性磷酸酶活力在4 h有所下降外,其余免疫相关酶活力均显著升高。研究结果表明,酚氧化酶是仿刺参非特异性免疫系统中最敏感、高效的免疫因子之一;革兰氏阳性细菌与革兰氏阴性细菌之间在诱导仿刺参免疫因子应答变化上无明显规律性差异;溶壁微球菌诱导溶菌酶的应答变化与灿烂弧菌、哈维氏弧菌、假交替单胞菌和停乳链球菌存在明显差异,溶菌酶可能是仿刺参清除入侵溶壁微球菌的主要免疫因子;灿烂弧菌诱导仿刺参免疫因子应答变化显著不同于其他4株细菌,显示出本研究选取的5个免疫指标在预警灿烂弧菌病害上具有潜在应用价值;停乳链球菌在仿刺参中具有作为免疫增强剂的潜在应用价值。  相似文献

Previous studies have demonstrated that petal shape and size in legume flowers are determined by two separate mechanisms, dorsoventral (DV) and organ internal (IN) asymmetric mechanisms, respectively. However, little is known about the molecular mechanisms controlling petal development in legumes. To address this question, we investigated petal development along the floral DV axis in Lotus japonicus with respect to cell and developmental biology by comparing wild‐type legumes to mutants. Based on morphological markers, the entire course of petal development, from initiation to maturity, was grouped to define 3 phases or 13 stages. In terms of epidermal micromorphology from adaxial surface, mature petals were divided into several distinct domains, and characteristic epidermal cells of each petal differentiated at stage 9, while epidermal cells of all domains were observed until stage 12. TCP and MIXTA‐like genes were found to be differentially expressed in various domains of petals at stages 9 and 12. Our results suggest that DV and IN mechanisms interplay at different stages of petal development, and their interaction at the cellular and molecular level guides the elaboration of domains within petals to achieve their ideal shape, and further suggest that TCP genes determine petal identity along the DV axis by regulating MIXTA‐like gene expression.  相似文献

A new triterpenoid saponin containing five sugars was isolated from the ethanolic root extract of Dipsacus japonicus Miq., and its structure was established as 3-O-[β-D-glucopyranosyl(1→4)][α-L-rhamnopyranosyl(1→3)]-β-D-glucopyranosyl(1→3)-α-L-rhamnopyranosyl(1→2)-α-L-arabinopyranosyl-oleanoic acid. The sites of glycosylation and the sequence of sugars in the glycoside can be determined unambiguously and total assignment of severely overlapping proton resonance of sugar residues were achieved by a combined use of the 1D-SEMDY and NOE difference spectroscopy in rotating frame techniques, without having recourse to chemical degration or modification. The results showed that these new NMR techniques are very effective and convenient for the structure determination and spectral assignment of this class of compounds.  相似文献

In this study, we report the first dataset of phosphoproteins of the seeds of a model plant, Lotus japonicus. This dataset might be useful in studying the regulatory mechanisms of seed germination in legume plants. By proteomic analysis of seeds following water absorption, we identified a total of 721 phosphopeptides derived from 343 phosphoproteins in cotyledons, and 931 phosphopeptides from 473 phosphoproteins in hypocotyls. Kinase‐specific prediction analyses revealed that different kinases were activated in cotyledons and hypocotyls. In particular, many peptides containing ATM‐kinase target motifs, X‐X‐pS/pT‐Q‐X‐X, were detected in cotyledons. Moreover, by real‐time RT‐PCR analysis, we found that expression of a homolog of ATM kinase is upregulated specifically in cotyledons, suggesting that this ATM‐kinase homolog plays a significant role in cell proliferation in the cotyledons of L. japonicus seeds. Growing rats fed a protein deficient diet showed an increase in acid and alkaline protease activity and a decrease in protease inhibitor activity. Similar results were found after two and four day fasting periods. Animals on diets of 10% and 20% casein showed parallel protease and protease inhibitor activity when compared to each other at similar body weight periods.  相似文献

The native rock pool mosquito, Aedes atropalpus (Coquillett), and the invasive Aedes japonicus (Theobald) have been found in many types of artificial and natural containers throughout North America. Little is known about the ecology of these two species in habitats where they co‐occur, although multiple investigators have reported the decline of the native species concurrent with the introduction and spread of the invasive species. Here we report the results of riverine rock pool collections (n=503) in the southern Appalachian Mountains between 2009‐2015. Surface water temperatures strongly predicted the presence of each species across a broad range of observed temperatures (11‐39.8° C). For every unit of increase in temperature (°C) the odds of collecting Ae. atropalpus larvae increased by 0.34 while the odds of collecting Ae. japonicus larvae decreased by 0.28. No Ae. japonicus larvae or pupae were collected at temperatures greater than 36° C; however, immature Ae. atropalpus were found in rock pools with temperatures up to 39.8° C. In contrast, Ae. japonicus were highly abundant in cooler rock pools (<17° C) where Ae. atropalpus were infrequent or absent. Our findings suggest that in spite of the successful invasion by Ae. japonicus, Ae. atropalpus remains well established in the southern Appalachian Mountains. Given the strong correlation of temperature with the presence of the two species and the contrasting absence of each species at observed temperature extremes, the role of thermal conditions should be carefully explored in the context of other ecological factors likely influencing the range and abundance of these mosquitoes.  相似文献

This study focused on three species of enemies, the parasitoid wasp Lysiphlebus japonicus Ashmead (Hymenoptera: Aphidiidae), the ladybird Scymnus posticalis Sicard (Coleoptera: Coccinellidae) and the predatory gall midge Aphidoletes aphidimyza (Rondani) (Diptera: Cecidomyiidae), all of which are able to exploit aphids attended by ants. I experimentally evaluated the effects of prey aphid species on the abundance of each of the three enemy species in ant‐attended aphid colonies on citrus. The aphids compared were Aphis gossypii Glover versus Aphis spiraecola Patch in late spring, and Toxoptera citricidus (Kirkaldy) versus A. spiraecola in late summer (all, Hemiptera: Aphididae). Colonies of the three aphid species were attended by the ant Pristomyrmex punctatus Smith (Hymenoptera: Formicidae). The initial number of attending ants per individual aphid did not differ significantly between the colonies of the two aphid species compared in each season. Between A. gossypii and A. spiraecola, there was no significant difference in the number of mummies formed by the parasitoid or foraging larvae of each of the two predators per aphid colony. A significant difference was detected between T. citricidus and A. spiraecola for each of the three enemy species, with a far greater number of L. japonicus mummies in T. citricidus colonies and distinctly more larvae of each of the two predators in A. spiraecola colonies. Thus, the abundance of each of the three enemy species in ant‐attended aphid colonies was significantly influenced by the species of the prey aphids, with the three enemies showing different responses to the three aphid species.  相似文献
