Late Embryogenesis Abundant (LEA) proteins are commonly found in plants and other organisms capable of undergoing severe and reversible dehydration, a phenomenon termed “anhydrobiosis”. Here, we have produced a tagged version for three different LEA proteins: pTag-RAB17-GFP-N, Zea mays dehydrin-1dhn, expressed in the nucleo-cytoplasm; pTag-WCOR410-RFP, Tricum aestivum cold acclimation protein WCOR410, binds to cellular membranes, and pTag-LEA-BFP, Artemia franciscana LEA protein group 3 that targets the mitochondria. Sheep fibroblasts transfected with single or all three LEA proteins were subjected to air drying under controlled conditions. After rehydration, cell viability and functionality of the membrane/mitochondria were assessed. After 4 h of air drying, cells from the un-transfected control group were almost completely nonviable (1% cell alive), while cells expressing LEA proteins showed high viability (more than 30%), with the highest viability (58%) observed in fibroblasts expressing all three LEA proteins. Growth rate was markedly compromised in control cells, while LEA-expressing cells proliferated at a rate comparable to non-air-dried cells. Plasmalemma, cytoskeleton and mitochondria appeared unaffected in LEA-expressing cells, confirming the protection conferred by LEA proteins on these organelles during dehydration stress. This is likely to be an effective strategy when aiming to confer desiccation tolerance to mammalian cells.  相似文献

Elasmosauridae were cosmopolitan Late Cretaceous plesiosaurs with conspicuous morphological diversity. Within this group, vertebral morphology is a criterion for estimating relative age in plesiosaur. On the other hand, the microstructure of plesiosaur bone is considered as indicative of ontogenetic stage. However, knowledge about ontogenetic tissue transformation in different elements of the skeleton is poorly known. Resorption and remodelling of skeletal tissues are required for development and growth, mechanical adaptation, repair and mineral homeostasis of the vertebrate skeleton. This contribution analyses different postcranial elements of a Late Cretaceous elasmosaurid from Patagonia. Characterisation of bone microstructure indicates the presence of compact bone inner organisation in an adult derived plesiosaur from the Cretaceous and that the distribution of bone specialisations depicts conspicuous variations within a single skeleton depending on the skeletal element considered. Bone compactness or degree of remodelling in elasmosaurids is not necessarily correlated with the ontogenetic age of the animal or to costal versus pelagic lifestyles. The available data are still scarce, but we propose a topic of discussion: perhaps the degree of remodelling and compactness also may be related to the activity level and increased mechanical load in different skeletal elements.  相似文献

Paleobiotic assemblages from the Deccan infra- and intertrappean beds are reviewed in great detail. Three distinct paleoenvironments (fluvio-lacustrine/terrestrial, brackish water and marine) have been identified within the infra- and intertrappean biotic assemblages of peninsular India. Recently, marine incursions have been recorded in a few of the Deccan intertrappean beds exposed in central and south-eastern India. The intertrappean beds have yielded marine planktic foraminiferans and freshwater/brackish water ostracods. The affinities of the paleobiotas are commonly considered to show a mixed pattern resulting from the addition of Gondwanan and Laurasian elements to endemic Indian taxa. During the last four decades, various biogeographic models (southern and northern connections) have been proposed to explain the presence of anomalous biogeographic biota in the Late Cretaceous of India. Based on the recovered fauna and flora assemblages, the Cretaceous–Paleogene boundary has been marked and a Late Cretaceous to Early Paleocene age has been assigned to these Deccan volcano-sedimentary sequences.  相似文献

In the latest Maastrichtian, the European hadrosauroid fauna was more diverse than those of North America and Asia. The European record of hadrosaurid dentaries is an example of this diversity, and most of the sites with mandibular remains are located in the Ibero-Armorican Realm. Within the Iberian Peninsula, most of the remains are located in the Tremp Basin (South Central Pyrenees). Two of the three valid hadrosaurid taxa defined in this basin are from the Blasi sites (Arén, Huesca province): Arenysaurus ardevoli (Blasi-3) and Blasisaurus canudoi (Blasi-1). A new locality in Blasi (Blasi 3.4) has provided a new dentary from an indeterminate euhadrosaurid. This dentary presents some characters intermediate between Arenysaurus and Blasisaurus, some characters similar to Pararhabdodon isonensis (from the nearby province of Lleida), and some characters of its own. Nevertheless, due to its fragmentary character, without dentition or its edentulous anterior part, it cannot be determined above the level of Euhadrosauria. It thus represents a fourth Iberian euhadrosaurian taxon in the Ibero-Armorican Realm, different from Arenysaurus, Blasisaurus and Pararhabdodon, increasing the diversity of hadrosauroids in this realm at the very end of the Cretaceous.  相似文献

The brachiopod Superfamily Spiriferoidea diversified greatly and was widely distributed in the late Palaeozoic (Carboniferous–Permian), and yet its phylogeny has been seldom investigated with analytical methods. This is reflected in the current flux of very different classification schemes for this superfamily. This paper provides the first attempt to investigate the phylogenetic relationships of spiriferoid brachiopods through both cladistic and Bayesian analyses involving 24 discrete and continuous characters. The continuous characters, from morphometric data, have been separately discretized using the gap weighting method, and the ‘as such’ option in TNT. Our results highlight the potential significance of continuous characters in reconstructing and elucidating phylogenies, as much as qualitative characters. Building on the outcomes of the analyses, we also briefly evaluate existing classification schemes of Spiriferoidea. We found that none of the existing classifications fully reflect the phylogeny properly; major families within the superfamily, such as Spiriferidae, Choristitidae, and Trigonotretidae, turned out to be polyphyletic. Although this study is considered preliminary, due to the selection of and restriction to certain taxa, combined with the use of a relatively small number of characters, it nevertheless demonstrates that potentially the true phylogenetic relationships of spiriferoid taxa sharply contrast with any of the existing classification schemes. This highlights the need to develop an alternative scheme that takes into account a more comprehensive range of phylogenetic variables.  相似文献

Plectorthis is a commonly reported Late Ordovician brachiopod genus from Laurentia and several other tectonic plates. Examination of shells from the Cincinnati Arch type area shows that species previously assigned to the genus have a considerable range of variability, such as ribbing style, shell outline, and musculature. The type species, Orthis plicatella Hall, 1847 is characterized by generally simple costae, with double-rowed aditicules on each rib, and a cordate ventral muscle field. Some species previously attributed to Plectorthis have a subtriangular ventral muscle field and hence a closer affinity to the plaesiomyid genus Austinella. Orthis fissicosta is here demoted to a subspecies of Plectorthis plicatella based on its similar ribbing style to some specimens of the type species. In Laurentia, Plectorthis first appeared in the Darriwilian–Sandbian boundary interval, achieved a relatively high species diversity and wide paleogeographic distribution in pericratonic seas during the early Katian; the subequatorial species attained a notably larger shell than those in the higher paleotropics. The genus did not invade the intracratonic basins of Laurentia and became extinct by the late Katian.  相似文献
