Redox Fluctuations Frame Microbial Community Impacts
on N-cycling Rates in a Humid Tropical Forest Soil 总被引:1,自引:0,他引:1
Fluctuating soil redox regimes may facilitate the co-occurrence of microbial nitrogen transformations with significantly different sensitivities to soil oxygen availability. In an upland humid tropical forest, we explored the impact of fluctuating redox regimes on gross nitrogen cycling rates and microbial community composition. Our results suggest that the rapidly fluctuating redox conditions that characterize these upland soils allow anoxic and oxic N processing to co-occur. Gross nitrogen mineralization was insensitive to soil redox fluctuations. In contrast, nitrifiers in this soil were directly affected by low redox periods, yet retained some activity even after 3–6 weeks of anoxia. Dissimilatory nitrate reduction to ammonium (DNRA) was less sensitive to oxygen exposure than expected, indicating that the organisms mediating this reductive process were also tolerant of unfavorable (oxic) conditions. Denitrification was a stronger sink for NO3− in consistently anoxic soils than in variable redox soils. Microbial biomass and community composition were maintained with redox fluctuation, but biomass decreased and composition changed under static oxic and anoxic soil regimes. Bacterial community structure was significantly correlated with rates of nitrification, denitrification and DNRA, suggesting that redox-control of soil microbial community structure was an important determinant of soil N-cycling rates. Specific nitrogen cycling functional groups in this environment (such as nitrifiers, DNRA organisms, and denitrifiers) appear to have adapted to nutrient resources that are spatially and temporally variable. Temporary pools are numerous in coastal plains of Atlantic Morocco and have a rich and diverse flora. These habitats are increasingly
under pressure by man impact through grazing by domestic livestock and the development of annual crops in their catchments.
The objectives of this work were to evaluate the species richness and the species composition of the vegetation of a sample
of 30 pools in this region, in order to assess the structure of the vegetation within pools and to evaluate the role of environmental
and anthropogenic factors in their richness and species composition. The results highlighted the species richness of the pools
with 300 species found among which annual species were heavily dominant. The structure of the vegetation varied within pools
according to a topographical/flooding gradient and between pools in relation with the land use in the surrounding areas. Three
vegetation belts were recognized from the centre to the periphery of the pool. The total species richness per pool was not
found significantly related to environmental or anthropogenic factors. The species characteristics of the pools were found
mostly in the centre and their number affected by hydrological factors and land use in the pool and in the surrounding areas.
The species characteristics of woodland habitats and of agriculture crops were found mostly in the peripheral zone. Temporary
pools maintain specific communities of both aquatic and amphibious species, probably because of the selection induced by water
level variations. The peripheral zone, although often dominated by terrestrial species, is clearly interpreted as part of
the pool. It contains amphibious species highly characteristic of the pool for which the irregular flooding is a key environmental

The free-radical state of K562 human erythroleukemia cells changes during the development of resistance to doxorubicin, an antitumor antibiotic with prooxidant action widely used in oncology. It was found that the level of superoxide anion in the resistant cells decreased. The addition of doxorubicin to the culture medium induced a much smaller increase in O 2 ? generation in the resistant cells than in the sensitive cells. Again, the semiquinone-type EPR signal with a g-factor of 2.006 considerably decreased in the resistant cells grown without doxorubicin as compared with the sensitive cells under the same conditions. The EPR study has shown that the level of paramagnetic nitrosyl complexes of nonheme iron in the resistant cells decreased, which indicates that the content of free nonheme iron declined in development of drug resistance. In addition, we have found with the use of RT-PCR that the level of mRNA of the transferrin receptor decreased in the resistant cells. The data suggest that the suppression of free-radical processes during the development of resistance of K562 cells to doxorubicin is a coordinated redox-dependent adaptive response. 相似文献

The sodium-selective amiloride-sensitive epithelial sodium channel (ENaC) mediates electrogenic sodium re-absorption in tight epithelia. ENaC expression at the plasma membrane requires regulated transport, processing, and macromolecular assembly of subunit proteins in a defined and highly compartmentalized manner. Ras-related Rab GTPases monitor these processes in a highly regulated sequence of events. In order to evaluate the role of Rab proteins in ENaC function, Rab4 wild-type (WT), the GTPase-deficient mutant Rab4Q67L, and the dominant negative GDP-locked mutant Rab4S22N were over-expressed in the colon cancer cell line, HT-29 and amiloride-sensitive currents were recorded. Rab4 over-expression inhibited amiloride-sensitive currents. The effect was reversed by introducing Rab4-neutralizing antibody and Rab4 specific SiRNA. The GDP-locked Rab4 mutant inhibited, while GTPase-deficient mutant moderately stimulated amiloride-sensitive currents. Active status of Rab4 was confirmed by GTP overlay assay, while its expression was verified by Western blotting. Immunoprecipitation and pull-down assay suggest protein-protein interaction between Rab4 and ENaC. In addition, the functional modulation coincides with concomitant changes in ENaC expression at the cell surface and in intracellular pool. We propose that Rab4 is a critical element that regulates ENaC function by mechanisms that include GTP-GDP status, recycling, and expression level. Our observations imply that channel expression in apical membranes of epithelial cell system incorporates RabGTPase as an essential determinant of channel function and adds an exciting paradigm to ENaC therapeutics. 相似文献

In a recent issue, Rausch and Morgan (2005) analyzed the role of selfing in the autopolyploidization process theoretically based on the formulation of Felber (1991), although they did not explicitly model a pollen pool. This paper reformulates the male contribution to outcrossing in terms of gamete frequencies within a pollen pool. Abstract: Sodium has many fundamental physiological functions in animals but is rare in boreal ecosystems where moose (Alces alces) thrive. In Québec (Canada), sodium is readily available in aquatic vegetation and in salt pools that form along highways. We do not know if moose are adopting specific behaviors to access sodium sources or if they simply use the sodium sources they encounter during their movements. We tested the hypothesis that moose modify both space and habitat use to gather sodium from salt pools. We expected moose to use salt pools mostly in spring and early summer, when needs are greatest and before aquatic vegetation has fully developed. We fitted 47 moose with Global Positioning System telemetry collars and collected data for 2 to 36 months between 2003 and 2006. We rarely located moose at salt pools (0.12% among the 95,007 locations collected). As we expected, use of salt pools was highest in late spring and in early summer, and we observed a time lag between peak use of salt pools compared to use of lakes and waterways, indicating moose fulfilled their sodium requirements in salt pools before aquatic vegetation was available. Moose selected salt pools over lakes and waterways when these 2 sodium sources were present in their home range and moved rapidly over large distances to reach them. Our results were consistent with moose using salt pools when they are likely to be sodium deficient. Salt pools were less accessible, required long-distance movements, and were located in habitually avoided areas along highways. Elimination of roadside salt pools should be considered among strategies to reduce cervid-vehicle collision risks in boreal environments. 相似文献

Changes in the nature of the ecological resources exploited by a species can lead to the evolution of novel suites of behaviours. We identified a case in which the transition from large pool use to the use of very small breeding pools in neotropical poison frogs (family Dendrobatidae) is associated with the evolution of a suite of behaviours, including biparental care (from uniparental care) and social monogamy (from promiscuity). We manipulated breeding pool size in order to demonstrate experimentally that breeding habitat selection strategy has evolved in concert with changes in parental care and mating system. We also manipulated intra- and interspecific larval interactions to demonstrate that larval adaptation to the use of very small pools for breeding affected the evolution of larval competition and cannibalism. Our results illustrate the intimate connection between breeding pool ecology, parental care and mating strategies in Peruvian poison frogs. 相似文献

Archaea have been detected recently as a major and often dominant component of the microbial communities performing ammonia oxidation in terrestrial and marine environments. In a molecular survey of archaeal ammonia monooxygenase (AMO) genes in terrestrial hot springs of Iceland and Kamchatka, the amoA gene encoding the alpha-subunit of AMO was detected in a total of 14 hot springs out of the 22 investigated. Most of these amoA-positive hot springs had temperatures between 82 and 97 degrees C and pH range between 2.5 and 7. In phylogenetic analyses, these amoA genes formed three independent lineages within the known sequence clusters of marine or soil origin. Furthermore, in situ gross nitrification rates in Icelandic hot springs were estimated by the pool dilution technique directly on site. At temperatures above 80 degrees C, between 56 and 159 mumol NO(3)(-) L(-1) mud per day was produced. Furthermore, addition of ammonium to the hot spring samples before incubation yielded a more than twofold higher potential nitrification rate, indicating that the process was limited by ammonia supply. Our data provide evidence for an active role of archaea in nitrification of hot springs in a wide range of pH values and at a high temperature. 相似文献

Inventories of soil C pools are still lacking from tropical sites. Our objective was to assess total C and N concentrations in the different mineral soil fractions down to 50 cm depth in relation to selected physical and chemical properties of 5 ecosystems at La Flor Sustainable Center in Guanacaste, Costa Rica. The ecosystems studied were a derived savanna with scattered trees, a gallery forest, an abandoned Mango indigofera L. plantation, a Citrus sp. plantation, and a Saccharum officinarum L. (sugarcane) plantation. Significant differences were found for the main fixed factor ecosystem for all variables analyzed (ANOVA). The TSC concentration was significantly higher in the sugarcane plantation compared to the rest of land use systems. The TSC concentration decreased significantly with increase in depth in all ecosystems and ranged from 20.3–38.3 to 4.3–20.9 g kg−1 in the 0–10 and 40–50 cm depth, respectively. In all cases, the clay + silt fraction (<50 μm) contained the highest C concentration. N concentration (0–10 cm depth) at La Flor ranged from 0.32 to 0.19%, and decreased in the order sugarcane > Curatella savanna > Mango and Citrus plantations > gallery forest. A principal component analysis (PCA) performed with all variables studied showed that the ordination of land uses (ecosystems) in the factorial plane defined by the first two axes was significant (Monte Carlo permutation test, P < 0.0001). The highest TSC pool down to 50 cm depth was obtained in the sugarcane plantation (160 Mg C ha−1) while less C was found in the rest of ecosystems, i.e. from 66 (gallery forest) to 80 Mg C ha−1 (Curatella savanna). The TSC concentration obtained in the sugarcane plot is likely the result of the incorporation of surface residues into the soil that would have otherwise been lost through burning, which is the current practice in the region. Further studies on C stabilization in the clay fraction are thus needed to test the hypothesis of soil C enrichment due to residue management. Finally, trade-offs are to be considered for both preservation of the fragile dTf and the productivity of derived land uses that increases soil C at the same time. 相似文献
