Autoimmune hepatitis is a rare but life threatening autoimmune disease of the liver of unknown etiology1,2. In the past many attempts have been made to generate an animal model that reflects the characteristics of the human disease 3-5. However, in various models the induction of disease was rather complex and often hepatitis was only transient3-5. Therefore, we have developed a straightforward mouse model that uses the major human autoantigen in type 2 autoimmune hepatitis (AIH-2), namely hCYP2D6, as a trigger6. Type 1 liver-kidney microsomal antibodies (LKM-1) antibodies recognizing hCYP2D6 are the hallmark of AIH-27,8. Delivery of hCYP2D6 into wildtype FVB or C57BL/6 mice was by an Adenovirus construct (Ad-2D6) that ensures a direct delivery of the triggering antigen to the liver. Thus, the ensuing local inflammation generates a fertile field9 for the subsequent development of autoimmunity. A combination of intravenous and intraperitoneal injection of Ad-2D6 is the most effective route to induce a long-lasting autoimmune damage to the liver (section 1). Here we provide a detailed protocol on how autoimmune liver disease is induced in the CYP2D6 model and how the different aspects of liver damage can be assessed. First, the serum levels of markers indicating hepatocyte destruction, such as aminotransferases, as well as the titers of hCYP2D6 antibodies are determined by sampling blood retroorbitaly (section 2). Second, the hCYP2D6-specific T cell response is characterized by collecting lymphocytes from the spleen and the liver. In order to obtain pure liver lymphocytes, the livers are perfused by PBS via the portal vein (section 3), digested in collagen and purified over a Percoll gradient (section 4). The frequency of hCYP2D6-specific T cells is analyzed by stimulation with hCYP2D6 peptides and identification of IFNγ-producing cells by flow cytometry (section 5). Third, cellular infiltration and fibrosis is determined by immunohistochemistry of liver sections (section 6). Such analysis regimen has to be conducted at several times after initiation of the disease in order to prove the chronic nature of the model. The magnitude of the immune response characterized by the frequency and activity of hCYP2D6-specific T and/or B cells and the degree of the liver damage and fibrosis have to be assessed for a subsequent evaluation of possible treatments to prevent, delay or abrogate the autodestructive process of the liver.  相似文献

Conclusion Unless the basis of reactivity can be strictly elucidated the biological significance of molecular mimicry must be held in question and the attention given to fulfilling the criteria outlined in table 2 cannot be overstressed. Nevertheless, the evidence so far is intriguing in implicating some form of role for retroviruses or bacteria in certain autoimmune diseases. The reader would be well advised to keep an open mind and await with critical interest more stringent data from those laboratories with the resources, patience and dedication to pursue this line of research.  相似文献

Tree cavities are a critical resource for many animals, especially as nesting sites for birds. Patterns of cavity distribution in temperate forests are well studied, yet little is known of cavities in tropical forests, despite a hypothesized decrease in cavity availability with decreasing latitude. We studied cavity density and distribution in a wet lowland tropical forest in Costa Rica and compared our results with estimates from forests around the world. Cavities at our site were common, occurred frequently in living trees, and were often formed by damage or decay rather than by woodpeckers. Most cavities had small openings, and woodpecker-created cavities were nonrandomly oriented. Contrary to prediction, cavity density appears to increase from the poles to the tropics. We suggest potential mechanisms to explain these patterns.  相似文献

The Collection and Sale of Wild Mushrooms in a Community of Tlaxcala, Mexico. In this paper, we characterize the harvest and sale of wild mushrooms in Javier Mina (Tlaxcala state, Mexico) based on observations made during visits to that community during the rainy seasons of 1995 and 2001. While there, we visited the forests with various families of mushroom gatherers and also carried out 16 visits to a local mushroom broker to obtain information on the buying and selling of the mushrooms, including a list of the prices per kilo of the species that are sold commercially. Boletus cf. pinophilus, Lyophyllum cf. decastes, Hebeloma aff. mesophaeum and the Amanita caesarea complex were the highest–priced species, and were those collected in the greatest amounts. Our observations indicate that the gathering of mushrooms, a traditional activity among the people of Javier Mina, contributes, through the sale of the mushrooms, to the income of the community during the rainy season.
Recolección y venta de hongos silvestres en una comunidad de Tlaxcala, México. Recolección y venta de hongos silvestres en una comunidad de Tlaxcala, México. El objetivo de este trabajo es describir el proceso de recolección y venta de hongos silvestres en Javier Mina, Tlaxcala. Para describir estas actividades se realizaron visitas a los bosques con varias familias recolectoras de hongos durante las épocas de lluvias de 1995 y 2001. Además, se realizaron 16 visitas a un distribuidor local para registrar información con respecto a la compra y venta de los hongos. Se obtuvo un listado de los precios por kilo de los hongos que se comercializan. Boletus cf. pinophilus, Lyophyllum cf. decastes, Hebeloma aff. mesophaeum y Amanita caesarea complex fueron los hongos recolectados en mayor cantidad durante este estudio, mismos que tuvieron los precios más altos de compra–venta. Se concluye que la recolección de hongos es una actividad tradicional para la gente de Javier Mina; sin embargo, la venta de éstos, contribuye a la obtención de ingresos durante la temporada de lluvias.

Palynological assemblages are reported for the first time for the La Irene Formation, southwestern Santa Cruz Province, Argentina. This unit represents the sedimentation during a regressive period from transitional or delta-plain to fluvial environments. Palynological assemblages are scarce and moderately well preserved. They mainly consist of continental elements (wood remains, cuticles, spores and pollen grains) with scarse marine palynomorphs (dinoflagellate cysts). The spore-pollen assemblages are dominated by fern spores, followed by angiosperm and gymnosperm pollen grains. Bryophyte spores and fungal remains are also present. Among ferns, Cyatheaceae and spores of Laevigatosporites, of uncertain affinity, are dominant. Of the angiosperm pollen, those of Chloranthaceae (Clavatipollenites sp.) and Arecaceae (Arecipites spp., Longapertites sp., Spinizonocolpites hialinus Archangelsky and Zamaloa) are the most abundant. Pollen of Liliaceae (Liliacidites spp.), Proteaceae (Proteacidites sp., Peninsulapollis gillii (Cookson) Dettmann and Jarzen, Retidiporites camachoii Archangelsky) and Ericaceae (Ericipites scabratus Harris) are also present. Gymnosperm pollen is represented by Podocarpaceae (Podocarpidites spp.) and Ephedraceae (Equisetosporites sp.). These palynological suites would represent a fern-angiosperm dominated coastal vegetation, developed under warm and at least locally humid climatic conditions. La Irene Formation is considered Maastrichtian in age based on stratigraphic evidence, which is, in turn consistent with the ages suggested by the species ranges and the similarities observed with others previously described assemblages. This is the southernmost record of Spinizonocolpites, similar to the extant mangrove palm Nypa.  相似文献

Crown dimensions and occlusal surface wear rate and wear plane were evaluated using paired first and second mandibular molars from a sample of 84 Early Agricultural period (1600 B.C.-A.D. 200) skeletons from northwest Mexico. Although this period represents a major shift in subsistence strategies in the Sonoran Desert, from food-foraging to agriculture, archaeological and dental pathology studies have identified this period as one of relative dietary stability. It was therefore predicted that very little variation in occlusal wear would have occurred between the early phase (San Pedro: 1600-800 B.C.) and late phase (Cienega: 800 B.C.-A.D. 200). Comparison of crown diameters identified some phenotypic differences between sexes but not between archaeological phases. Molar occlusal surfaces were then divided into four quadrants, and wear scores recorded for each quadrant. Principle axis analysis was performed between total wear scores of paired, adjacent first and second mandibular molars to assess rate and occlusal wear plane over time. The analysis demonstrated that both wear rate and wear plane increased from the early to the late phase of the Early Agricultural period. These results indicate that although diet may have indeed remained stable during this period in the Sonoran Desert increases in the rate of wear and wear plane may reflect changes in food-processing techniques. It is suggested that more intensive processing of agricultural products during the Cienega phase simultaneously softened the diet to create more tooth-contact wear and introduced more grit to cause faster and more angled wear on the molar occlusal surfaces.  相似文献

研究了Cd2+毒害对低等水生植物槐叶苹的生理影响及外源La、Ca的缓解效应。结果表明,Cd2+污 染时,槐叶苹叶片出现褪绿、叶缘拳卷并伴有烂斑和根状叶断离现象。随着Cd2+浓度的升高,叶片的叶绿素 含量、SOD、CAT、POD活性先升后降,但各指标达到最大值时的Cd2+浓度各不相同,以POD对应的最高。可 溶性蛋白含量及丙二醛(MDA)产量分别呈现出下降和上升趋势,且可溶性蛋白含量受Cd2+的影响更大。外 源La和Ca均能缓解Cd2+对槐叶苹叶片叶绿素含量、可溶性蛋白含量的降低和MDA的积累,对保护酶系统 的影响存在较大差异。  相似文献

Cerium (III), lanthanum (III) and neodymium (III) complexes with 3,3'-benzylidenebis[4-hydroxycoumarin] were synthesized in view of their application as cytotoxic agents. The complexes were characterized by different physicochemical methods: elemental analysis, mass spectrometry, 1H NMR, 13C NMR and IR spectroscopy. The spectra of the complexes were interpreted on the basis of comparison with the spectrum of the free ligand. The vibrational analysis showed that in the complexes the ligand coordinated to the metal ion through both deprotonated hydroxyl groups; however, participation of the carbonyl groups in the coordination to the metal ion was also suggested. The evaluation of the cytotoxic activity of the novel lanthanide complexes on HL-60 myeloid cells revealed that they are potent cytotoxic agents. The cerium complex was found to exhibit superior activity in comparison to the lanthanum and neodymium coordination compounds, the latter being the least active. Our data give us reason to conclude that the newly synthesized lanthanide complexes should be submitted to further more detailed pharmacological and toxicological evaluation.  相似文献

Abstract. Questions: Does distance decay exist in an old‐growth neotropical forest? Is this distance decay stronger than expected due to environmental heterogeneity alone? At what spatial scales are distance decay manifested? Location: La Selva Biological Station, Costa Rica, Central America. Methods: An index of distance decay is applied appropriate for small quadrats (the probability of encountering a conspecific tree) to a grid of 1170 0.01‐ha plots. A null model is provided that accounts for environmental heterogeneity. Results: Significant, but weak, distance decay is found. After correcting for known patterns of environmental heterogeneity, the distance decay almost disappears, except for fine spatial scales. Conclusions: These results are inconsistent with models that predict distance decay at all spatial scales. However, biological processes leading to distance decay may be more relevant and detectable at scales broader than this study. Research utilizing objectively‐located samples over much broader scales is necessary to evaluate the generality and magnitude of distance decay.  相似文献

Northern terrestrial ecosystems have shown global warming‐induced advances in start, delays in end, and thus increased lengths of growing season and gross photosynthesis in recent decades. The tradeoffs between seasonal dynamics of two opposing fluxes, CO2 uptake through photosynthesis and release through respiration, determine the influence of the terrestrial ecosystem on the atmospheric CO2 and 13C/12C seasonality. Here, we use four CO2 observation stations in the Northern Hemisphere, namely Alert, La Jolla, Point Barrow, and Mauna Loa Observatory, to determine how changes in vegetation productivity and phenology, respiration, and air temperature affect both the atmospheric CO2 and 13C/12C seasonality. Since the 1960s, the only significant long‐term trend of CO2 and 13C/12C seasonality was observed at the northern most station, Alert, where the spring CO2 drawdown dates advanced by 0.65 ± 0.55 days yr?1, contributing to a nonsignificant increase in length of the CO2 uptake period (0.74 ± 0.67 days yr?1). For Point Barrow station, vegetation phenology changes in well‐watered ecosystems such as the Canadian and western Siberian wetlands contributed the most to 13C/12C seasonality while the CO2 seasonality was primarily linked to nontree vegetation. Our results indicate significant increase in the Northern Hemisphere soil respiration. This means, increased respiration of 13C depleted plant materials cancels out the 12C gain from enhanced vegetation activities during the start and end of growing season. These findings suggest therefore that parallel warming‐induced increases both in photosynthesis and respiration contribute to the long‐term stability of CO2 and 13C/12C seasonality under changing climate and vegetation activity. The summer photosynthesis and the soil respiration in the dormant seasons have become more vigorous which lead to increased peak‐to‐through CO2 amplitude. As the relative magnitude of the increased photosynthesis in summer months is more than the increased respiration in dormant months, we have the increased overall carbon uptake rates in the northern ecosystems.  相似文献
