Stimulation of synthesis of immunoglobulin (Ig) in vitro by Con A and anti-Ig in cultures of rabbit lymphoid cells has been analyzed qualitatively using an assay that measures the incorporation of [3H]leucine into newly synthesized proteins, followed by the specific absorption of tritiated immunoglobulin by staphylococcal protein A. Whereas Con A stimulates Ig production by spleen cells only if T lymphocytes are present, anti-immunoglobulin serum enhances Ig synthesis in the absence of T lymphocytes. In contrast, neither Con A nor anti-immunoglobulin serum stimulates peripheral blood lymphocytes to produce enhanced levels of Ig. It is concluded that both Con A and anti-immunoglobulin serum do not activate resting B cells but drive differentiation of B cells which are already synthesizing Ig. Anti-Ig acts directly whereas stimulation of B-cell Ig synthesis by Con A occurs indirectly through stimulation of T cells.  相似文献

Only 30% of commercial heparin reacts with antithrombin III (ATIII). This study shows that the interaction is pH dependent: 100% of the heparin binds to ATIII at pH 3.0, 30% at physiological pH. Binding of ATIII, platelet factor 4, and protamine to heparin was studied using a new rocket precipitin electrophoresis procedure, adapted from the Laurell rocket immunoelectrophoresis procedure. Protamine is incorporated into agarose gel, and heparin mixtures with protamine, ATIII, or platelet factor 4 electrophoresed into the gel from a series of wells. The residual free heparin is precipitated by the protamine in a rocket-shaped arc, the height of which is proportional to the amount of free heparin. No antibody is employed. This procedure is useful for quantitation of heparin and for studying the binding of heparin to proteins.  相似文献

In four early Celtic settlements in the central Neckar region of Baden-Württemberg (Germany), 250 archaeobotanical samples were collected and studied for their plant macrofossils.Hordeum vulgare L. (hulled barley) andTrilicum spella L. (spelt wheat) could be recognized as the main crops, as well asPanicum miliaceum L. (millet) in some places. Legumes andLinum usitatissimum L. (flax) were important crops. Typical field weeds dominated the wild plant remains, but some present-day grassland taxa were abundantly and frequently found. The analysis of the data set of finds of the cereal remains, field weeds and grassland plants was done by Correspondence Analysis, but no significant differences between the various groups were found. Samples dominated by remains of grassland taxa, which could indicate that there were hay meadows, were absent. Consideration of plant macrofossil results as well as archaeological, osteological and pollen analytical data shows that there is likely to have been a land management system of arable fields and grassland. The densely settled fertile central Neckar region could therefore have been intensively utilised, using all available resources. Permanent grasslands in the form of pasture on river floodplains and on dry soils were certainly present. Hay meadows, on the other hand, were not identified. Cultivated plants such asFicus carica L. (fig) andVicia ervilia (L.) Willd. (bitter vetch) as well as some field weeds suggest connections with the Mediterranean lands.  相似文献

Standard population genetic analyses are implemented for a series of precontact and contact period samples from central and northern Florida to investigate changes in genetic variability and population affinity coincident with the establishment of Spanish missions during the 17th century. Estimates of F(ST) based on odontometric data indicate limited heterogeneity for the Apalachee samples, suggestive of some degree of within-group endogamy for this ethnic group prior to contact. This corresponds well with ethnohistoric reconstructions indicating that Apalachee were populous, partially linguistically isolated from its neighbors, and involved in persistent cycles of warfare with neighboring groups. Estimates of extralocal gene flow for the Apalachee samples indicate limited initial changes in the mating structure of these populations. After 1650, however, extralocal gene flow increases, consistent with evidence for dramatic population movements throughout northern Florida and increased Spanish presence in the province, particularly at the mission of San Luis. Inclusion of non-Apalachee outgroups does not increase estimates of genetic heterogeneity, as was expected based on ethnohistoric data. The pattern of genetic distances suggests a biological division between north and south Florida population groups, consistent with archaeological and ethnohistoric data, and similarly indicates some distinction between precontact and postcontact local groups. Differential extralocal gene flow experienced by pre-1650 Apalachee and Timucua populations suggests localized mission experience. The Apalachee, with large, dense populations, experienced limited initial changes in genetic diversity or mating structure. However, after 1650 they were apparently involved in a much more expansive mating network that may have included Spaniards and immigrant Native American groups to the region. These results are in contrast to the mission experience of the Guale Indians of the Georgia coast.  相似文献

 In transmission electron microscopy studies, lanthanum ions have been used as electron-opaque tracers to delineate the apoplastic pathways for ion transport in barley (Hordeum vulgare L.) roots. To localize La3+ on the subcellular level, e.g. in cell walls and on the surface of membranes, electron-energy-loss spectroscopy and electron-spectroscopic imaging were used. Seminal and nodal roots were exposed for 30 min to 1 mM LaCl3 and 10 mM LaCl3, respectively. In seminal roots, possessing no exodermis, La3+ diffusion through the apoplast was stopped by the Casparian bands of the endodermis. In nodal roots with an exodermis, however, La3+ diffusion through the cortical apoplast had already stopped at the tight junctions of the exodermal cell walls resembling the Casparian bands of the endodermis. Therefore, we conclude that in some specialized roots such as the nodal roots of barley, the physiological role of the endodermis is largely performed by the exodermis. High-resolution magic-angle spinning (MAS) 1H nuclear magnetic resonance (NMR) spectroscopic and pattern recognition (PR) based methods have been applied to studies on the acute biochemical effects of La(NO3)3 on rats. Male Wistar rats were treated with various doses of La(NO3)3 (2, 10, and 50 mg/kg body weight), and MAS 1H NMR spectra of their intact liver and kidney tissues were analyzed using principal components analysis to extract metabolic information. The biochemical effects of La(NO3)3 were characterized by the increase of triglyceride and bile acid and the decrease of glycogen in liver tissue, together with a slight elevation of triglyceride level in kidney tissue. The target lesion of La(NO3)3 to liver was found by MAS NMR-PR methods. This study illustrated the power of the combination of MAS 1H NMR and pattern recognition for the analysis of biochemical effects of rare earths.  相似文献

Tropical rainforests have experienced episodes of severe heat and drought in recent decades, and climate models project a warmer and potentially drier tropical climate over this century. However, likely responses of tropical rainforests are poorly understood due to a lack of frequent long‐term measurements of forest structure and dynamics. We analyzed a 12‐year record (1999–2010) of 47 817 annual measurements of canopy height to characterize the response of an old‐growth Neotropical rainforest to the severe heat and drought associated with the 1997–1998 El Niño. Well‐drained soils on slopes and plateaus experienced a threefold increase in the fraction of the landscape in gaps (≤2 m) and a reduction in the fraction in high canopy (>15 m) causing distributions of canopy height to depart from equilibrium for a period of 2–3 years. In contrast, forests on low‐lying alluvial terraces remained in equilibrium and were nearly half as likely to experience upper canopy (>15 m) disturbance over the 12 years of observation. Variation in forest response across topographic positions suggests that tropical rainforests are more sensitive to moisture deficits than high temperature and that topography likely structures landscape‐level variation in the severity of drought impacts.  相似文献

Tropical arid to semi‐arid ecosystems are nearly as diverse as more humid forests and occupy large parts of the tropics. In comparison, however, they are vastly understudied. For instance, fog precipitation alone supports a unique vegetation formation, locally termed lomas, on coastal mountains in the Peruvian desert. To effectively protect these highly endemic and threatened ecosystems, we must increase our understanding of their diversity patterns in relation to environmental factors. Consequently, we recorded all vascular species from 100 random 4 × 4 m plots on the fog‐exposed southern slope of the mountain Mongón. We used topographic and remotely sensed covariates in statistical models to generate spatial predictions of alpha diversity and plant species' distribution probabilities. Altitude was the most important predictor in all models and may represent fog moisture levels. Other significant covariates in the models most likely refer also to water availability but on a finer spatial scale. Additionally, model‐based clustering revealed five altitudinal vegetation zones. This study contributes to a better spatial understanding of the biodiversity and spatial arrangement of vegetation belts of the largely unknown but highly unique lomas formations. Furthermore, mapping species richness and plant species' distributions could support a long‐needed lomas strategic conservation scheme.  相似文献
