自20世纪六十年代末结肠镜检査术问世以来,无论是其操作手法还是器械改革均得到飞速发展,并广泛应用于临床,使直视病灶、实现活检及治疗成为可能。虽然,结肠镜检查为结直肠疾病的诊断及治疗带来了巨大的帮助并成为结直肠癌诊断的金标准,但其局限性也日渐明显。作为一种侵入性操作,结肠镜检查无法避免的腹部疼痛、难以预知的并发症、无法保证的成功率以及可能存在的漏诊率限制了其临床应用不仅在一定程度上降低了患者的依从性,增加了操作的困难度,且使一部分具有操作指征的患者错失早期诊断及治疗的最佳时机。本文简要阐述了传统注气式结肠镜检查法的一般特点并总结了其应用现状,以期为今后传统注气式结肠镜检查法的合理化应用提供一定的参考依据。 相似文献

Microbial fuel cells (MFCs) were used to monitor metabolism changes in Clostridium acetobutylicum fermentations. When MFCs were inoculated with C. acetobutylicum, they generated a unique voltage output pattern where two distinct voltage peaks occurred over a weeklong period. This result was markedly different to previously studied organisms which usually generate one sustained voltage peak. Analysis of the fermentation products indicated that the dual voltage peaks correlated with glucose metabolism. The first voltage peak correlated with acidogenic metabolism (acetate and butyrate production) and the second peak with solventogenic metabolism (acetone and butanol production). This demonstrates that MFCs can be applied as a novel tool to monitor the shift from acid production to solvent production in C. acetobutylicum. 相似文献

An outward current that appeared to activate instantaneously in response to depolarising voltage pulses at low sampling frequencies
predominated in the plasma membrane of ground-parenchyma protoplasts derived from coats of developing Phaseolus vulgaris L. (cv. Redland Pioneer) seeds. However, the outward current showed time-dependent activation when higher sampling frequencies
were used to measure the current. Activation of the current was best described as a double-exponential time course with the
fast and slow time constants being 1 and 20 ms, respectively. The current also exhibited a rapid deactivation that followed
a double-exponential time course with time constants of approximately 2 and 30 ms, respectively. “Tail-current” analysis allowed
us to show that this current exhibited a low selectivity between K+ and Cl− (P
K:Cl=1.8). Such a fast-activating current may account for some of the reports of time-independent, instantaneous currents that
have been observed in plasma membranes of plant cells digitised at low sampling frequencies. Therefore, when “instantaneous”
currents appear it is advisable to characterise these currents using higher sampling frequencies with correspondingly higher
filtering frequency cut-offs.

A study of the benthic invertebrate community inhabiting a small, foothill trout stream in the Rocky Mountains of Idaho was conducted over a two-year period. Monthly Hess samples and short-term experiments using substratum-filled trays were used to describe the spatial dispersion of the benthos and to examine the response of invertebrate populations to substratum and current. A method was devised for measuring available surface area which involved coating individual stones with latex and measuring the area of the print resulting from inking the impression left on the latex mold.The dispersion of all populations was clumped throughout the year. Alteration of the cross-sectional pattern of current velocity and stream bed composition changed the pattern of distribution but not the extent of clumping. Collections made in areas of depositing and eroding substrata revealed a more diverse fauna in the latter. Most groups of organisms found in the riffle were scarcer in the pools or absent from them. The pool fauna contained no important additions over those found in the riffles.After a year's study of invertebrate populations in an otherwise undisturbed riffle, the substratum was altered and the flow made more uniform; an increase in the abundance of most of the benthic invertebrates followed. No single factor was responsible for the increase, but the change in substratum size and degree of compaction accounted for most of the change. Interpretation of the results was aided by findings from experiments using substratum-filled trays.Two series of stream experiments using the trays were conducted: one to test the relative importance of current and substratum and the other to test the effect of particle size on the distribution of the benthic fauna. In the first series, placement of trays of stones in a pool resulted in an increase in numbers of some but not all of the invertebrates over numbers usually occurring in the pool. Trays filled with stones and placed in a riffle supported fewer animals than found on the adjacent stream bed but more than in the pool. Variations are attributed to differences in current velocity and amounts of imported organic and inorganic debris. Three different relationships of population numbers to current velocity were found for different members of the community (direct, indirect, and parabolic) over the range of 10 to 60 cm/sec. The second series of experiments consisted of two sets of trays filled with stones of medium or large pebbles, respectively. Nine taxa, as well as all of the combined taxa, showed a preference for trays of small stones over the natural stream bed. A few taxa were noticeably more abundant on the small substratum than on the large but most of the fauna showed only slight increases in numbers or remained constant on the two substrata. Only three taxa showed a direct relation of numbers to total surface area presented by the stones.Number and kinds of organisms found in trays filled with a uniform size of substratum did not correspond to those taken in Hess samples from the natural stream bed. This has important implications in terms of currently recommended pollution monitoring techniques. However, it is suggested that if the substratum composition of the trays more nearly matched that of the stream, the correspondence would be much better. The results of the present study also throw considerable doubt on the adequacy of generalizations derived from earlier studies of responses to substratum size and suggest several reasons for reevaluating current ideas regarding the influence of substratum on invertebrate distribution. 相似文献

New records of Scaphodinium mirabile (Dinophyceae), an unnoticed dinoflagellate in the Pacific Ocean
Previous records of dinoflagellate Scaphodinium mirabile Margalef (Leptodiscaceae, Noctilucales) were restricted to the Mediterranean‐Black Sea and Eastern Atlantic Ocean. Nine and 34 specimens were observed in the upper 100 m layer in May and July, respectively, in a cross‐section in the vicinity of the Kuroshio Current (NW Pacific Ocean). Nearly all the Lugol‐fixed specimens appeared folded over themselves, an appearance that differs from the view reported in the scarce literature available. 相似文献

Eastern boundary current systems are among the most productive and lucrative ecosystems on Earth because they benefit from upwelling currents. Upwelling currents subsidize the base of the coastal food web by bringing deep, cold and nutrient‐rich water to the surface. As upwelling is driven by large‐scale atmospheric patterns, global climate change has the potential to affect a wide range of significant ecological processes through changes in water chemistry, water temperature, and the transport processes that influence species dispersal and recruitment. We examined long‐term trends in the frequency, duration, and strength of continuous upwelling events for the Oregon and California regions of the California Current System in the eastern Pacific Ocean. We then associated event‐scale upwelling with up to 21 years of barnacle and mussel recruitment, and water temperature data measured at rocky intertidal field sites along the Oregon coast. Our analyses suggest that upwelling events are changing in ways that are consistent with climate change predictions: upwelling events are becoming less frequent, stronger, and longer in duration. In addition, upwelling events have a quasi‐instantaneous and cumulative effect on rocky intertidal water temperatures, with longer events leading to colder temperatures. Longer, more persistent upwelling events were negatively associated with barnacle recruitment but positively associated with mussel recruitment. However, since barnacles facilitate mussel recruitment by providing attachment sites, increased upwelling persistence could have indirect negative impacts on mussel populations. Overall, our results indicate that changes in coastal upwelling that are consistent with climate change predictions are altering the tempo and the mode of environmental forcing in near‐shore ecosystems, with potentially severe and discontinuous ramifications for ecosystem structure and functioning. 相似文献

Quasi-synoptic surveys along the east coast of Australia between 28 and 34°S show that the heterotrophic dinoflagellate, Noctiluca scintillans, occurs along this entire stretch of the coast. Areas of relatively high abundance of Noctiluca were observed downstream of regions predisposed to current-induced upwellings as a consequence of alongshore topographic variations. High-resolution temporal and spatial sampling of upwelling events showed that Noctiluca was abundant (up to 28 cells l?1) within mature upwelled waters. A high proportion (>80%) of fed Noctiluca cells (cells with prey in their vacuoles) was observed in the mature upwelled waters indicating that the observed increase in abundance of Noctiluca was associated with increased feeding activity. The absolute abundance of Noctiluca in upwelled waters was, however, found to vary from one upwelling location to another and between seasons. In particular, highest abundances of Noctiluca were recorded south of 31.5°S, where the East Australian Current (EAC) characteristically separates from the coast. The high abundances partly arise from southward advection and retention of the Noctiluca cells, and partly from upwelling inshore of the separated EAC driven by cross-shelf boundary layer fluxes. The temperature of the EAC was also found to influence absolute abundances. Surface water temperatures during our summer cruise were anomalously high due to a strong La Niña phase, and up to 4°C warmer than during our spring cruise. We found that the warmer surface water temperatures were associated with relatively lower average abundances of Noctiluca in the near shore zone. 相似文献

Estimation of population firing rates and current source densities from laminar electrode recordings
This model study investigates the validity of methods used to interpret linear (laminar) multielectrode recordings. In computer
experiments extracellular potentials from a synaptically activated population of about 1,000 pyramidal neurons are calculated
using biologically realistic compartmental neuron models combined with electrostatic forward modeling. The somas of the pyramidal
neurons are located in a 0.4 mm high and wide columnar cylinder, mimicking a stimulus-evoked layer-5 population in a neocortical
column. Current-source density (CSD) analysis of the low-frequency part (<500 Hz) of the calculated potentials (local field
potentials, LFP) based on the ‘inverse’ CSD method is, in contrast to the ‘standard’ CSD method, seen to give excellent estimates
of the true underlying CSD. The high-frequency part (>750 Hz) of the potentials (multi-unit activity, MUA) is found to scale
approximately as the population firing rate to the power 3/4 and to give excellent estimates of the underlying population
firing rate for trial-averaged data. The MUA signal is found to decay much more sharply outside the columnar populations than

Human antibody-forming cells were demonstrated by a plaque in agar technique following in vitro stimulation with either pokeweed mitogen or Cowan I strain of protein A-positive Staphylococcus aureus bacteria. We evaluated the effects on this antibody formation caused by the addition of cells which had been stimulated with PH A or Con A. Both Con A and PHA cells harvested after 3 days showed strong inhibition of pokeweed-induced plaque formation. The majority of the suppression could be accounted for by a blast fraction separated on 1g sedimentation gradients from the Con A or PHA cultures. Small cells from such cultures showed inhibition of PFC when added at high ratios (1:2), but this suppressive activity diluted out much more rapidly than that of the blast cells. No helper activity was noted with either small cells or blasts. Our studies indicate a T-cell blast as the suppressive fraction in Con A- or PHA-stimulated human lymphoid cells. While this T-cell suppression applies to T-dependent responses such as antibody stimulation with pokeweed mitogen, it does not have a substantial effect on Cowan I-induced plaque-forming responses. The finding that Cowan I-induced plaques could not be inhibited by Con A or PHA blasts indicates the T independence of this response. 相似文献
