The copy frequency distribution of a transposable element family in a Drosophila melanogaster natural population is generally characterised by the values of the Charlesworths' model parameters α and β (Charlesworth & Charlesworth, 1983). The estimation of these parameters is made using the observed distribution of the occupied sites in a population sample. Several results have been interpreted as due either to the influence of stochastic factors or to deterministic factors (transposition, excision, selection…). The accuracy of this method was tested by estimations performed on samples from simulated populations. The results show that with the sample size usually used for natural population studies, the confidence intervals are too large to reasonably deduce either the element copy number distribution or the values of transposition and excision rate and selective coefficients. 相似文献

The first 12 NH2-terminal amino acids of the Pseudomonas putida putidaredoxin reductase were shown to be Met-Asn-Ala-Asn-Asp-Asn-Val-Val-Ile-Val-Gly-Thr. Comparison of these data with the DNA sequence of the BamHI-HindIII 197-base fragment derived from the PstI 2.2-kb fragment obtained from the P. putida plasmid showed that the putidaredoxin reductase gene was downstream from the cytochrome P-450 gene and the intergenic region had the 24-nucleotide sequence TAAACACATGGGAGTGCGTGCTAA. The Shine-Dalgarno sequence GGAG was detected in this region. The initiating triplet for the reductase gene was GTG, which normally codes for valine, but in the initiating codon position codes for methionine. From the amino acid sequence and X-ray data comparisons with other flavoproteins, what appears to be the AMP binding region of the FAD can be recognized in the NH2-terminal portion of the reductase involving residues 5–35.This article was presented during the proceedings of the International Conference on Macromolecular Structure and Function, held at the National Defence Medical College, Tokorozawa, Japan, December 1985. 相似文献

The use of fossil fuel is predicted to cause an increase of the atmospheric CO2 concentration, which will affect the global pattern of temperature and precipitation. It is therefore essential to incorporate effects of temperature and water supply on carbon partitioning of plants to predict effects of elevated [CO2] on growth and yield of Triticum aestivum. Although earlier papers have emphasized that elevated [CO2] favours investment of biomass in roots relative to that in leaves, it has now become clear that these are indirect effects, due to the more rapid depletion of nutrients in the root environment as a consequence of enhanced growth. Broadly generalized, the effect of temperature on biomass allocation in the vegetative stage is that the relative investment of biomass in roots is lowest at a certain optimum temperature and increases at both higher and lower temperatures. This is found not only when the temperature of the entire plant is varied, but also when only root temperature is changed whilst shoot temperature is kept constant. Effects of temperature on the allocation pattern can be explained largely by the effect of root temperature on the roots' capacity to transport water. Effects of a shortage in water supply on carbon partitioning are unambiguous: roots receive relatively more carbon. The pattern of biomass allocation in the vegetative stage and variation in water-use efficiency are prime factors determining a plant's potential for early growth and yield in different environments. In a comparison of a range of T. aestivum cultivars, a high water-use efficiency at the plant level correlates positively with a large investment in both leaf and root biomass, a low stomatal conductance and a large investment in photosynthetic capacity. We also present evidence that a lower investment of biomass in roots is not only associated with lower respiratory costs for root growth, but also with lower specific costs for ion uptake. We suggest the combination of a number of traits in future wheat cultivars, i.e. a high investment of biomass in leaves, which have a low stomatal conductance and a high photosynthetic capacity, and a low investment of biomass in roots, which have low respiratory costs. Such cultivars are considered highly appropriate in a future world, especially in the dryer regions. Although variation for the desired traits already exists among wheat cultivars, it is much larger among wild Aegilops species, which can readily be crossed with T. aestivum. Such wild relatives may be exploited to develop new wheat cultivars well-adapted to changed climatic conditions. 相似文献

The heart is one of the least regenerative organs in the body, and highly vulnerable to the increasing incidence of cardiovascular diseases in an aging world population. Cell-based approaches aimed at cardiac repair have recently caused great public excitement. But clinical trials of patients’ own skeletal myoblasts or bone marrow cells for transplantation have been disappointing. Human embryonic stem cells (hESCs) form bona fide cardiomyocytes in vitro which are readily generated in mass culture and are being tested in animal models of heart damage. The early results, while encouraging, underscore that much remains to be done. Proteolysis of the hydroxylase component of soluble methane monooxygenase (MMO) with trypsin yielded a protein which retained 50% activity in a standard MMO assay. In an H2O2-driven assay, in which H2O2 replaced two of the protein components, NADH and O2 used in the standard assay, the proteolysed hydroxylase retained full activity for ethane, propane and propene, but had a 2–3 fold increase with methane as substrate. Several crosslinking reagents have been tested for their ability to stabilise the proteolysed form of the hydroxylase. Using polyoxyethylene bis(imidazolyl carbonyl) (Mr 3350) as the crosslinking agent, increased thermostability of the hydroxylase was observed. Activated methoxypolyethylene glycol (Mr 5000) was used to modify the hydroxylase which was now soluble in organic solvents as well as water and could be activated by H2O2. The glycol-modified hydroxylase functioned well in organic solvents in the catalysis of propene oxidation. 相似文献

Water-insoluble compounds can be substrates for enzymatic reactions when lipases are immobilized properly and suitable organic solvents are used. In this review, three type of lipase immobilization method and their application to the asymmetric syntheses of complex molecules are described. Lipases immobilized with Celite or synthetic prepolymers such as urethane prepolymer and photo-crosslinkable resin prepolymer have been applied for the kinetic resolution of many kinds of water-insoluble substrate.
Phospholipid-lipase aggregates with ether linkages are novel and have been found to function effectively as immobilized lipases in asymmetric hydrolysis or esterification reactions in water-saturated organic solvent. In order to investigate gene expression changes associated with cytotoxicity, we used cDNA arrays to monitor the expression
of over 5,000 genes in response to toxic stress in the HepG2 liver cell line. Cells were treated with cytotoxic doses of acetaminophen,
caffeine or thioacetamide for nine time points ranging from 1 to 24 h. Samples of mRNA from each time point were used to prepare
radiolabeled cDNA, which was hybridized to nylon-membrane-based cDNA arrays. High-stringency washes were applied to reduce
cross-hybridization. Analysis of spot intensities revealed that each compound led to approximately 150-250 gene expression
changes that were sustained over at least three adjacent time points. The affected genes could be classified into clusters
based on their temporal patterns of differential expression. A common set of 44 genes showed similar expression changes in
response to all three compounds. Of these changes, 90% could be confirmed by quantitative RT-PCR analysis. The results indicate
that detailed array-based time-course studies, coupled with a sensitive and highly specific confirmation assay, provide a
powerful means of identifying cytotoxicity-associated gene expression changes.

The melanogenic actions of the melanocortins are mediated by the melanocortin‐1 receptor (MC1R). MC1R is a member of the G‐protein‐coupled receptors (GPCR) superfamily expressed in cutaneous and hair follicle melanocytes. Activation of MC1R by adrenocorticotrophin or α‐melanocyte stimulating hormone is positively coupled to the cAMP signaling pathway and leads to a stimulation of melanogenesis and a switch from the synthesis of pheomelanins to the production of eumelanic pigments. The functional behavior of the MC1R agrees with emerging concepts in GPCR signaling including dimerization, coupling to more than one signaling pathway and a high agonist‐independent constitutive activity accounting for inverse agonism phenomena. In addition, MC1R displays unique properties such as an unusually high number of natural variants often associated with clearly visible phenotypes and the occurrence of endogenous peptide antagonists. Therefore MC1R is an ideal model to study GPCR function. Here we review our current knowledge of MC1R structure and function, with emphasis on information gathered from the analysis of natural variants. We also discuss recent data on the regulation of MC1R function by paracrine and endocrine factors and by external stimuli such as ultraviolet light. 相似文献
