Significantly larger numbers of Toxoplasma gondii cysts were detected in the brains of RAG1?/?NOS2?/? than RAG1?/? mice following infection. In contrast, the cyst numbers markedly decreased in a same manner in both strains of mice after receiving CD8+ immune T cells. Thus, NOS2-mediated innate immunity is important for inhibiting formation of cysts in the brain but not required for the T cell-initiated cyst removal, which is associated with phagocyte accumulation. Treatment with chloroquine, an inhibitor of endolysosomal acidification, partially but significantly inhibited the T cell-mediated cyst removal, suggesting that phagosome–lysosome fusion could be involved in the T. gondii cyst elimination.  相似文献

Resistance to leaf spot and melting out caused by Drechslera poae was evaluated in eight cultivars and seven breeding lines of Poa pratensis under greenhouse conditions. The results obtained in this study were compared with levels of resistance scored after natural infection under field conditions during 1994–1997 in sun and shade lawn maintenance, as described before. It was found that resistance assessed in the greenhouse as leaf spots in the three‐leaf seedling stage 10–14 days after the first inoculation (assessment I) and resistance assessed as severity of melting out on the crown 56 days after the first inoculation (assessment IV) showed the highest repeatability with the scores obtained in the field under sun and shade conditions during 1994–1997.  相似文献

A panel of five plant lectins with different binding specificities was used to determine if plant lectins could bind specifically to membrane-associated glycolipids. Ricinis communis and wheat germ agglutinins both bound specifically to mixed brain gangliosides and globoside I from human erythrocytes. Wheat germ agglutinin also bound to ganglioside GM1 and human erythrocyte ceramide trihexoside, but not to ceramide dihexoside, mono-, or digalactosyl diglycerides. Concanavalin A bound to liposomes with or without glycolipid substituents, and this binding was partially inhibited by α-methyl mannoside. This study indicates that lectins can specifically recognize and bind to certain glycolipids in membranes.  相似文献

Plants of Poa pratensis cv. Holt initiate inflorescence primordia when exposed to short days (SD) and low temperature, but require a secondary induction by at least 4 long days (LD) for further inflorescence development and stem elongation. Single or double applications of 10 µg per plant of gibberellins A1, A3, A5 and 16,17‐dihydro A5 (DHGA5) induced inflorescence development in a high proportion of plants in SD, but only if the plants were detillered to a single stem. Exposure to 2 LD cycles did not cause heading and flowering alone but enhanced the effect of exogenous gibberellins (GAs), bringing flowering to 100%. GA5 and DHGA5 were less effective than GA1 and GA3 in SD, especially with double applications, but were more effective than GA1 and GA3 when given together with 2 LD. The GAs had differential effects on vegetative growth and flowering, GA5 and DHGA5 causing much less leaf and stem growth than the other two GAs. Marginal induction, whether by LD or GA application, resulted in a high proportion of spikelets with viviparous proliferation. Thus, whereas GAs are inhibitory to the primary induction by SD, they can replace secondary induction by LD when vegetative growth is limited.  相似文献

Four hundred and forty-four ecotypes of Kentucky bluegrass ( Poa pratensis L.) collected from Poland were screened for rust resistance. A field experiment was established at Hof Steimke, DSV (Deutschen Saatveredelung), Germany. The ecotypes originated from the Polish Gene Bank, IHAR – Botanical Garden, Bydgoszcz. Sixty-two (13.9%) ecotypes that scored on average more than 5 were included in a group with high resistance. Ecotypes scored on average less than 5 were either included in a susceptible group (average scores 4.0–4.9) or a very susceptible group (average scores 2.0–3.9). There were 181 ecotypes in the susceptible group (40.8%) and 201 (45.3%) in the very susceptible group. Thirty-three (7.4%) ecotypes were more resistant and 29 (6.5%) ecotypes had resistance on the level of the most resistant control variety Ottos (score of resistance 5.3). Eight ecotypes (1.8%) with the highest level of resistance (score 7 or higher) were identified as especially promising sources of rust resistance. One ecotype (141858) showed reaction type 9 in both years and another one (141903) showed reaction type 9 in one year and reaction type 8 in another year (mean 8.5). Six ecotypes were scored on average 7 or 7.5. For 17 different habitats from which the tested ecotypes were collected only those ecotypes originating from 10 habitats (meadow, peat meadow, forest meadow, pasture, field, lakeside, waterside, ditch, roadside and gravel pit) showed high levels of resistance to rust. The value of newly identified resistant ecotypes for control of rusts on Kentucky bluegrass is discussed.  相似文献

Forest openings, also known as glades, arise through a variety of mechanisms including disturbance (fire and blow downs) and local variation in soil or bedrock geology. They are common in many forest types and are often dominated by locally rare herbaceous species. Prescribed burning is increasingly used as a management approach for maintaining glades although little is known about the effects of fire on these habitats. Of particular concern is the potential for nutrient loss during and after fire because glades are often characterized by nutrient‐poor soils. We quantitated nutrient losses through combustion and leaching for glade and adjacent forest habitats subjected to a prescribed burn. Our findings suggest that spring burns do not result in appreciable loss of nutrient capital from glades in comparison with those observed in the surrounding forest. Fire resulted in a substantial loss of litter mass (37%) in the forest but no measurable loss in the glade. Nitrogen losses through combustion were significant in the forest and were equivalent to 4.5 years of atmospheric inputs. Fire significantly increased soil nitrate pools in forest but not in glade plots. No detectable increases in nitrogen, phosphorus, or base cation leaching were observed in either forest or glade habitats within 4 months after the burn. These findings suggest that plant and microbial nutrient uptake rapidly reestablish control over leaching losses when burns are conducted at the start of the growing season. Biotic retention minimizes fire impacts on nutrient loss from the ecosystem.  相似文献

BACKGROUND AND AIMS: To improve the management of grass communities, early plant development was compared in three species with contrasting growth forms, a caespitose (Lolium perenne), a rhizomatous (Poa pratensis) and a caespitose-stoloniferous species (Agrostis stolonifera). METHODS: Isolated seedlings were grown in a glasshouse without trophic constraints for 37 d (761 degrees Cd). The appearance of leaves and their location on tillers were recorded. Leaf appearance rate (LAR) on the tillers and site-filling were calculated. Tillering was modelled based on the assumption that tiller number increases with the number of leaves produced on the seedling main stem. Above- and below-ground parts were harvested to compare biomass. KEY RESULTS: Lolium perenne and A. stolonifera expressed similar bunch-type developments. However, root biomass was approx. 30 % lower in A. stolonifera than in L. perenne. Poa pratensis was rhizomatous. Nevertheless, the ratio of above-ground : below-ground biomass of P. pratensis was similar to that of L. perenne. LAR was approximately equal to 0.30 leaf d(-1) in L. perenne, and on the main stem and first primary tillers of A. stolonifera. LAR on the other tillers of A. stolonifera was 30 % higher than on L. perenne. For P. pratensis, LAR was 30 % lower than on L. perenne, but the interval between the appearance of two successive shoots from rhizomes was 30 % higher than the interval between two successive leaf stages on the main stem. Above-ground parts of P. pratensis first grew slower than in the other species to the benefit of the rhizomes, whose development enhanced tiller production. CONCLUSIONS: Lolium perenne had the fastest tiller production at the earliest stages of seedling development. Agrostis stolonifera and P. pratensis compensated almost completely for the delay due to higher LAR on tillers or ramets compared with L. perenne. This study provides a basis for modelling plant development.  相似文献

Japanese beetle, Popillia japonica Newman, and oriental beetle, Anomala orientalis (Waterhouse) (both Coleoptera: Scarabaeidae) are considered invasive species and have been reported as key pests of urban landscapes in the Northeastern USA. Tiphia vernalis Rohwer and Tiphia popilliavora Rohwer (Hymenoptera: Tiphiidae) were introduced as biocontrol agents against these beetles. These parasitic wasps burrow into the soil and search for grubs. When a host is found, the wasp attaches an egg in a location that is specific for the wasp species. It is unknown if these wasps can detect patches of concealed hosts from a distance above ground and what role, if any, herbivore‐induced plant volatiles play in their host location. This study evaluated the responses of female T. vernalis and T. popilliavora to grub‐infested and healthy plants in Y‐tube olfactometer bioassays. Also the effect of root herbivory on the composition of turfgrass (Poaceae) volatile profiles was investigated by collecting volatiles from healthy and grub‐infested grasses. Tiphia wasps were highly attracted to volatiles emitted by grub‐infested tall fescue (Festuca arundinacea Schreb.) and Kentucky bluegrass (Poa pratensis L.) over healthy grasses. In contrast, wasps did not exhibit a significant preference for grub‐infested perennial ryegrass (Lolium perenne L.) as compared with the control plants. The terpene levels emitted by grub‐infested Kentucky bluegrass and tall fescue were greater than that of control plants. Low levels of terpenes were observed for both test and control perennial ryegrass. The elevated levels of terpenes emitted by grub‐infested Kentucky bluegrass and tall fescue coincided with the attractiveness to the tiphiid wasps. Here, we provide evidence that plant exposure to root‐feeding insects P. japonica and A. orientalis resulted in an increase in terpenoid levels in turfgrasses, which strongly attracts their above‐ground parasitoids.  相似文献

Influences on Release-Site Fidelity of Translocated Elk   总被引:2,自引:0,他引:2  
Several eastern states are considering the restoration of free‐ranging elk populations via translocation from western populations. Optimal habitat immediately surrounding release sites has been found to enhance elk reintroduction success in western states. Little information exists, however, to aid eastern managers in identifying release sites with the highest chance of restoration success. We monitored the movements of 415 translocated elk released at three sites in southeastern Kentucky to identify landscape characteristics that enhance release‐site fidelity. The distance elk moved after release differed among sites (F2,322 = 4.63, p = 0.01), age classes (F2,322 = 4.37, p = 0.01), and time intervals (F2,322 = 40.74, p < 0.001). At 6 and 12 months post‐release, adults (15.81 ± 17.32 and 16.38 ± 20.29) and yearlings (13.91 ± 16.44 and 14.61 ± 21.11) moved farther than calves (8.06 ± 14.03 and 9.37 ± 14.40). The release site with the highest fidelity was privately owned, 15% open, and had the highest amount of edge compared with the other release sites. The two remaining sites contained large amounts of expansive openland or forest cover with lower amounts of edge. Additionally, both sites were publicly owned and experienced a higher degree of human‐generated disturbance compared with the site to which elk were most faithful. When selecting release sites, managers should avoid areas dominated by a single cover type with little interspersion of other habitats. Rather, areas with high levels of open‐forest edge (approximately 5.0 km/km2) and limited‐human disturbance will likely enhance release‐site fidelity and promote restoration success.  相似文献
