基于叶绿体微卫星研究鄂报春谱系遗传结构   总被引:2,自引:0,他引:2  
鄂报春Primula obconica作为一种广泛栽培的园艺植物,其野生居群的遗传多样性及遗传结构的研究还少见报道。本文通过叶绿体微卫星分析了17个鄂报春野生居群(共278个个体),共发现4个多态性位点(16个等位基因),得到14个单倍型。结果表明鄂报春具有很高的总基因多样性(HT=0.971)和极低的居群内基因多样性(HS=0.028);分子方差分析(AMOVA)显示98%的变异存在于居群间。这些结果说明早期的生境片断化及有限的种子传播能力是造成当前遗传结构的重要原因。Nst值显著大于Gst值,表明关系相近的单倍型会出现在相同的地区内,同时最小生成树(MST)的分析结果证实了这样的结论。我们在最小生成树的两个组中推断出一些古老单倍型,并推测在冰期时湖北和我国的西南地区可能是该物种的避难所。  相似文献

SAXIFRAGA BRYOIDES L. is one of the plant species reaching the upper limits of distribution for flowering plants in the European Alps. Because of its abundance in the subnival and nival zones, we expected S. BRYOIDES to reproduce efficiently in the highly stochastic climate at higher altitudes. Investigations were carried out at two subnival sites (2650 m and 2880 m a.s.l.) in the Austrian Alps. We studied flowering phenology, dynamics of seed development, and reproductive success in the climatically different years from 2001 - 2004. For a nival plant species, S. BRYOIDES showed a particularly long prefloration period (6 - 9 weeks). From onset of anthesis until seed maturity took an individual flower another 6 - 7 weeks and all individuals at a site 9 - 10 weeks. The length of the prefloration period and seed histogenesis was temperature-dependent, whereas seed maturation seemed to be endogenously controlled. Only in the exceptionally long and warm growing season of 2003 did all fruits mature at a site. In the other years, the onset of winter conditions halted development in many fruits before maturity. The seed/ovule ratio of mature fruits was around 0.7 in all years. The relative reproductive success (RRS) ranged from zero to 0.7, depending on the site and year. In conclusion, S. BRYOIDES needs an unexpectedly long time to undergo reproductive development. Though fruit maturation is uncertain, the high S/O ratio of single intact fruits results in at least a small seed crop in most years. This seems to be sufficient to assure the spread and maintenance of S. BRYOIDES at higher altitudes. As a seed-risk strategist (Molau,1993), S. BRYOIDES would clearly benefit from a prolonged growing season, which might occur more often if climate warming continues.  相似文献

Exploratory movements and natal dispersal form essential processes during early life history stages of raptors, but identifying the factors shaping individual movement decisions is challenging. Global positioning system (GPS) telemetry thereby provides a promising technique to study movement patterns on adequate spatio-temporal scales. We analysed data of juvenile White-tailed Sea Eagles Haliaeetus albicilla (WTSE) in north-east Germany (n = 24) derived from GPS tracking to extensively analyse movements between fledging and emigration from the natal territory. Our goal was to determine the time point of fledging, characterize pre-emigration movements and the onset of natal dispersal while investigating the influence of the natal environment. WTSE fledged at an average age of 72 days and showed strong excursive behaviour during the post-fledging period regarding the number, distance and duration of excursions, yet with high individual variability. Excursive behaviour did not differ between sexes. On average, WTSE left the parental territory 93 days after fledging. The quantity of excursive behaviour delayed the timing of emigration and WTSE tended to postpone their emigration when foraging water was accessible within the boundaries of their parental territory. The overall results suggest that young WTSE assess the quality of the natal environment via pre-emigration movements and stay in their territory of origin for as long as internal and external conditions allow for it. Our study is one of the first to characterize post-fledging and natal dispersal movements of young WTSE to such an extent and applies modern techniques to understand related movements in relation to the natal environment. The results emphasize the urgent necessity for the extension of currently existing nest protection periods and guaranteeing sustainable management of potential breeding and foraging grounds for WTSE. Ultimately, the results are relevant for all large raptor species sensitive to human-related disturbance, as they support the increasing importance of regulations with spatio-temporal specifications for breeding populations of large raptors in densely human-inhabited areas with increasing alteration of land.  相似文献

Extended periods of darkness have long been used to study how the mammalian visual system develops in the absence of any instruction from vision. Because of the relative ease of implementation of darkness as a means to eliminate visually driven neural activity, it has usually been imposed earlier in life and for much longer periods than was the case for other manipulations of the early visual input used for study of their influences on visual system development. Recently, it was shown that following a very brief (10 days) period of darkness imposed at five weeks of age, kittens emerged blind. Although vision as assessed by measurements of visual acuity eventually recovered, the time course was very slow as it took seven weeks for visual acuity to attain normal levels. Here, we document the critical period of this remarkable vulnerability to the effects of short periods of darkness by imposing 10 days of darkness on nine normal kittens at progressively later ages. Results indicate that the period of susceptibility to darkness extends only to about 10 weeks of age, which is substantially shorter than the critical period for the effects of monocular deprivation in the primary visual cortex, which extends beyond six months of age.  相似文献

During the late larval period, the salivary glands (SG) of Drosophila show a cascade of cytological changes associated with exocytosis and the expectoration of the proteinaceous glue that is used to affix the pupariating larva to a substrate. After puparium formation (APF), SG undergo extensive cytoplasmic vacuolation due to endocytosis, vacuole consolidation and massive apocrine secretion. Here we investigated possible correlations between cytological changes, the puffing pattern in polytene chromosomes and respiratory metabolism of the SG. The carefully staged SG were explanted into small amounts (1 or 2 μl) of tissue culture medium. The respiratory metabolism of single or up to 3 pairs of glands was evaluated by recording the rate of O2 consumption using a scanning microrespirographic technique sensitive to subnanoliter volumes of the respiratory O2 or CO2. The recordings were carried out at times between 8 h before pupariation (BPF), until 16 h APF, at which point the SG completely disintegrate. At the early wandering larval stage (8 h BPF), the glands consume 2 nl of O2/gland/min (=2500 μl O2/g/h). This relatively high metabolic rate decreases down to 1.2–1.3 nl of O2 during the endogenous peak in ecdysteroid concentration that culminates around pupariation. The metabolic decline coincides with the exocytosis of the proteinaceous glue. During and shortly after puparium formation, which is accompanied cytologically by intense vacuolation, O2 consumption in the SG temporarily increases to 1.6 nl O2/gland/min. After this time, the metabolic rate of the SG decreases downward steadily until 16 h APF, when the glands disintegrate and cease to consume oxygen. The SG we analyzed from Drosophila larvae were composed of 134 intrinsic cells, with the average volume of one lobe being 37 nl. Therefore, a single SG cell of the wandering larva (with O2 consumption of 2 nl/gland/min), consumes each about 16 pl of O2/cell/min. A simultaneous analysis of the rate of protein and RNA synthesis in the SG shows a course similar to that found in respiratory metabolism.  相似文献

Hepatic lipid metabolism is controlled by integrated metabolic pathways. Excess accumulation of hepatic TG is a hallmark of nonalcoholic fatty liver disease, which is associated with obesity and insulin resistance. Here, we show that KH-type splicing regulatory protein (KSRP) ablation reduces hepatic TG levels and diet-induced hepatosteatosis. Expression of period 2 (Per2) is increased during the dark period, and circadian oscillations of several core clock genes are altered with a delayed phase in Ksrp−/− livers. Diurnal expression of some lipid metabolism genes is also disturbed with reduced expression of genes involved in de novo lipogenesis. Using primary hepatocytes, we demonstrate that KSRP promotes decay of Per2 mRNA through an RNA-protein interaction and show that increased Per2 expression is responsible for the phase delay in cycling of several clock genes in the absence of KSRP. Similar to Ksrp−/− livers, both expression of lipogenic genes and intracellular TG levels are also reduced in Ksrp−/− hepatocytes due to increased Per2 expression. Using heterologous mRNA reporters, we show that the AU-rich element-containing 3′ untranslated region of Per2 is responsible for KSRP-dependent mRNA decay. These findings implicate that KSRP is an important regulator of circadian expression of lipid metabolism genes in the liver likely through controlling Per2 mRNA stability.  相似文献

Although little is known on the impact of environment on telomere length dynamics, it has been suggested to be affected by stress, lifestyle and/or life‐history strategies of animals. We here compared telomere dynamics in erythrocytes of hatchlings and fledglings of the brood parasite great spotted cuckoos (Clamator glandarius) and of magpies (Pica pica), their main host in Europe. In magpie chicks, telomere length decreased from hatching to fledging, whereas no significant change in telomere length of great spotted cuckoo chicks was found. Moreover, we found interspecific differences in the association between laying date and telomere shortening. Interspecific differences in telomere shortening were interpreted as a consequence of differences in lifestyle and life‐history characteristics of magpies and great spotted cuckoos. In comparison with magpies, cuckoos experience reduced sibling competition and higher access to resources and, consequently, lower stressful environmental conditions during the nestling phase. These characteristics also explain the associations between telomere attrition and environmental conditions (i.e. laying date) for magpies and the absence of association for great spotted cuckoos. These results therefore fit expectations on telomere dynamics derived from interspecific differences in lifestyle and life history of brood parasites and their bird hosts.  相似文献
