Dr. John Collier traced the discoveries that elucidated the structure and function of the anthrax toxin in his talk "Anthrax Toxin," which was part of the Microbiology Graduate Program Seminar Series at Yale School of Medicine on February 23, 2012. Dr. Collier, Professor of Microbiology and Immunobiology at Harvard University, began by noting the advantages to studying anthrax pathogenesis in a biosafety level-1 lab. This designation does not merely facilitate his research, but also reflects a larger trend of basic research being leveraged to develop translational applications. Basic research on toxin structure has led to the development of a vaccine by Dr. Collier's group. Next-generation prophylactics also may stem from recent discoveries uncovering a role for cellular cofactors that mediate toxin function. Finally, basic research into the toxin substructure has facilitated efforts to change the receptor tropism to target dysregulated cells for therapeutic purposes. The urgency around biodefense agents makes the choice of research priorities a salient issue. As such, this author submits that basic research occupies a unique and lucrative niche driving clinical applications.  相似文献

目的:分析髋关节置换术对股骨头坏死(osteonecrosis of the femoral head,ONFH)患者的髋关节Harris功能评分(Harris hip score,HHS)与简式Fugl-Meyer运动功能评分(Fugl-Meyer assessment scale,FMA)的影响。方法:回顾性选取于我院受诊的ONFH患者87例,按照其所用术式的不同,分为两组,传统组采用传统股骨头置换术治疗,共43例病例;研究组采用无柄解剖型人工髋关节置换术治疗,共44例,比较两组Harris评分、Fugl-Meyer运动功能评分的改变及统计并发症信息。结果:研究组Harris评分无显著差异(P0.05);治疗后,研究组Harris评分明显高于传统组(P0.05);研究组简式Fugl-Meyer运动功能显著优于传统组(P0.05);研究组并发症的发生率低于传统组(P0.05)。结论:无柄解剖型人工髋关节置换术治疗ONFH的临床疗效显著,能够改善患者髋关节功能及运动功能。  相似文献

In what follows, I consider the role of analogy in the first edition of Darwin’s Origin. I argue that Darwin follows Herschel’s methodology and hence exploits an analogy between artificial and natural selection that allows him generalize selection as a cause of evolutionary change. This argument strategy is not equivalent to an argument from analogy. Reading Darwin’s argument as conforming to Herschel’s two-step methodology of causal analysis followed by generalization allows us to understand the role and placement of Darwin’s discussion of artificial selection in the Origin, without making the mistake of portraying Darwin’s argument for the existence and character of natural selection as an analogical argument.  相似文献

A simple method for determining the age of an individual at the time of radiopaque transverse (Harris) line formation is presented. To use this method, only two measurements are required: total bone length and distance of line to nearest bone end; these are put into formulae that calculate the percent of total bone growth when the line appeared. The result of this calculation is compared with tables of percent bone growth per year (one to 16 years in females and one to 18 years in males) to arrive at estimations of age at line formation. Since these tables are presented for the femur, tibia, humerus, and radius, this technique can be used on any one of the major long bones exhibiting lines.  相似文献

The opsin from the John Dory Zeus faber rod visual pigment (maximum spectral sensitivity =492 nm) was cloned and sequenced. Comparison of the John Dory rod opsin sequence with those from other fish with blue-shifted scotopic spectral sensitivity provides further evidence for the spectral tuning of this group of visual pigments.  相似文献

The influence of cells of the polarizing zone mesoderm on the morphogenesis of recombinant chick limbs was studied. The recombinant buds were composed of leg bud ectoderm and different regions of the wing bud mesoderm, which had been dissociated and reaggregated. In any case where the polarizing zone mesoderm was coaggregated with the wing mesoderm the morphogenetic capabilities of the recombinant were reduced. This was the case with postaxial mesoderm, preaxial mesoderm plus polarizing tissue, and postaxial mesoderm from which a piece of the nonpolarizing mesoderm (comparable in size to the polarizing zone) had been removed. All of these gave outgrowths with digits in only a very low percentage of cases. In contrast, those recombinants without polarizing mesoderm developed outgrowths with digits in a high percentage of cases, indicating good morphogenesis. Finally, if the polarizing zone were removed prior to dissociation, the recombinant limb, composed of the total remaining wing bud mesoderm plus leg bud ectoderm, exhibited a higher percentage of complete morphogenesis than if the polarizing zone had been part of the recombinant.It is clear that cells of the polarizing zone, when dissociated, and coaggregated with wing mesoderm, are inhibitory to the morphogenetic performance of that mesoderm in the recombinant limb situation.  相似文献

Androsterone (3alpha-hydroxy-5alpha-androstan-17-one), 5alpha-androstane-3alpha, 17beta-diol and 5alpha-androstane-3beta, 17beta-diol were conjugated at C-16 through sulfur to bovine and human serum albumin. Rabbits injected with these conjugates produced antibodies suitable for radioimmunoassays of these hormone metabolites. Samples were purified on Sephadex LH-20 columns. Levels of these steroids were measured in a rat blood serum pool and in ovarian tissue extract pools.  相似文献

Rates of 24Na and 42K entry into dog red blood cells were found to be strongly influenced by cell volume. The kinetics of isotope movement were complex, and the cells were not in a steady state. By applying a simple, two-compartment equation to the early times points, values for flux were calculated and corrected for the changes in surface/volume ratio which occur when cells are shrunken or swollen. Curves were thus generated showing Na and K influx as functions of cell water content. A reinvestigation of the effects of adrenalectomy showed that all the observd changes in Na flux could be explained on the basis of alterations in red cell volume.  相似文献

Consideration is given to the reversible interaction of a bivalent ligand, B, with a multivalent acceptor, A (possessing f reactive sites) which leads to the formation of a series of complexes, AiBj, comprising networks of alternating acceptor and ligand molecules. A binding equation is derived on the basis of a site association constant, k, defined in terms of reacted site probability functions. This equation, which relates the binding function, r (the moles of ligand bound per mole of acceptor) to the concentration of unbound ligand, mb, is used to show that plots of r vs. 2kmB constructed with fixed but different values of kmA intersect at the point (mB = 12k, r = f2) where the extent of reaction and the concentrations of those complexes for which ji = f2 attain maximal values. Corresponding Scatchard plots are shown by numerical example to be non-linear, their second derivative being positive for all r. It follows that such deviations from linearity cannot be taken alone as evidence for site heterogeneity in cross-linking systems. The binding equation obtained directly is shown to be identical with that obtained with f = 2 by summation procedures involving the general expression for concentrations of complexes, mAiBj, formulated in terms of appropriate statistical factors. In this way, previous findings on precipitation and gel formation in cross-linking systems are correlated with the present development of binding theory.  相似文献

The α-hydroxylation and conversion of lignoceric acid into ceramide, cerebroside, and water-soluble products in particulate fractions from rat brain was studied in the presence of sphingosine and UDP-galactose, with particular reference to the effects of CoA and the synthesis of lignoceroyl-CoA. The synthesis of lignoceroyl-CoA was found to be almost completely dependent on the addition of exogenous CoA, whereas the formation of water-soluble products, mostly glutamate, was stimulated by, but was not stringently dependent on the addition of CoA. In contrast to these two metabolic pathways, both the synthesis of ceramides and cerebrosides and α-hydroxylation were unaffected by the addition of CoA. While removal of sphingosine and UDP-galactose had drastic effects on the sphingolipid synthesis, COA did not have any effect on the removal. On the other hand, removal of sphingosine resulted in a significant increase of the synmthesis of lignoceroyl-CoA and moderate increase in the formation of water-soluble products. These observations further indicated that CoA ester formation may not be required for the synthesis of these sphingolipids, and suggest that there may be two pathways for oxidative degradation of lignoceric acid in brain—one CoA-dependent and the other CoA-independent.  相似文献
