A full-length cDNA encoding D-amino acid oxidase (DAO, EC 1.4.3.3) was cloned and sequenced from the hepatopancreas of carp fed a diet supplemented with D-alanine. This clone contained an open reading frame encoding 347 amino acid residues. The deduced amino acid sequence exhibited about 60 and 19-29% identity to mammalian and microbial DAOs, respectively. The expression of full-length carp DAO cDNA in Escherichia coli resulted in a significant level of protein with DAO activity. In carp fed the diet with D-alanine for 14 days, DAO mRNA was strongly expressed in intestine followed by hepatopancreas and kidney, but not in muscle. During D-alanine administration, DAO gene was expressed quickly in hepatopancreas with the increase of DAO activity. The inducible nature of carp DAO indicates that it plays an important physiological role in metabolizing exogenous D-alanine that is abundant in their prey invertebrates, crustaceans, and mollusks.  相似文献

Many adverse effects on carp reproductive organs have been reported to be caused by exposure to environmental estrogens, such as nonylphenol and bisphenol A, which contaminate the aquatic environment. The glucuronidation activities of xenoestrogens (bisphenol A and diethylstilbestrol) and phytoestrogens (coumestrol, genistein and biochanin A), but not nonylphenol and octylphenol, were observed in microsomes prepared from carp organs. The highest levels of glucuronidation of environmental estrogens, for which the optimum temperature was 25-30 degrees C, were observed in the intestinal microsomes of 2-year-old carp. These activities in carp intestine increased developmentally, and the maximum levels corresponded to 5-10 % of that in rat liver microsomes. However, the glucuronidation of phytoestrogen by carp intestinal microsomes corresponded to that of rat liver microsomes. Only bisphenol A-glucuronide was excreted from the everted intestine, indicating that bisphenol A is metabolized in the carp intestine mainly as glucuronide.These results suggest that glucuronidation by carp intestine plays an important role for the detoxification of xenoestrogens and phytoestrogens, except for nonylphenol and octylphenol.  相似文献

The effects of tributyltin (TBT) on the energy metabolism and motility of fish spermatozoa were investigated in vitro in African catfish and common carp. A significant (P<0.05) decrease of the duration and the intensity of motility was observed in catfish spermatozoa exposed to 0.27 microg/l TBT for 24 h. Exposure of catfish spermatozoa to 2.7-27 microg/l TBT caused an instant decrease in ATP content. In the presence of 27 microg/l TBT approximately 55% of the initial ATP concentration in catfish semen was lost after 60 min incubation while AMP concentrations increased and the total adenine nucleotide (TAN) pool remained unchanged. The reduction in sperm ATP levels could not be attributed to cell death since viability decreased only slightly over the period of exposure. In carp by contrast, none of the adenylates concentrations studied (ATP, ADP and AMP) were affected by TBT exposure at any experimental condition. However, carp sperm motility was significantly reduced by exposure to 2.7 microg/l TBT. Among the enzymes investigated only lactate dehydrogenase (LDH) in catfish sperm was significantly (P<0.01) affected by 27 microg/l TBT treatment with a reduction in activity of approximately 75%. Compared with carp sperm before TBT exposure, that of catfish had lower adenylate contents and overall lower enzymatic activities; this explains its slower sperm velocity and shorter duration of movement as measured by computer assisted sperm analysis (CASA). The present in vitro study shows that catfish spermatozoa are more sensitive to TBT exposure (and probably to other toxicants) than those of carp.  相似文献

 The homeobox gene Carp-Ovx1 shows similarity to vertebrate and invertebrate Ovx genes and to Drosophila unplugged. Its expression pattern was studied by in situ hybridization in carp embryos and juveniles. During segmentation, expression becomes gradually limited to the neural tube. In juveniles up to 9 weeks old, cells in the ventral telencephalon, the facial lobe and the vagal lobe show Ovx1 expression, confining expression to parts with chemosensory projections. We cloned and sequenced two vitellogenin (vg) cDNAs of the carp, Cyprinus carpio, using a cDNA library constructed from estradiol-17β (E2)-treated livers. One was a novel, longer 5000 bp-long cDNA termed vg-B2 encoding 1624 amino acids in a single open reading frame. The other was a shorter cDNA (vg-B1), identical to that registered previously as carp vg cDNA in the international nucleotide sequence database. The deduced amino acid sequences of these two molecules were well-aligned with known vertebrate Vgs sharing common characteristics such as N-terminal lipovitellin I (LVI), phosvitin (PV) and C-terminal lipovitellin II (LVII). The novel Vg-B2 bore a highly conserved GL/ICG motif within the LVII region, in contrast to the shorter Vg-B1 that has a truncated C-terminal and lacks the β-component within the LVII region including the GL/ICG motif. Both vg-B2 and vg-B1 genes were expressed in the livers of females and E2-injected males. Western blot analysis using anti-Vg and anti-vitellin (Vn) antisera demonstrated that both Vg-B2 and Vg-B1 were detected as polypeptides with an estimated molecular mass of 180 kDa and 160 kDa, respectively, in the blood of females and E2-injected males. The results suggest the potential utilization of these genes as sensitive xenoestrogenic markers.  相似文献

The physiological significance of melatonin in the regulation of annual testicular events in a major carp Catla catla was evaluated through studies on the effects of graded dose (25, 50, or 100 µg/100 g body wt.) of melatonin exogenously administered for different durations (1, 15, or 30 days) and manipulation of the endogenous melatonin system by exposing the fish to constant darkness (DD) or constant light (LL) for 30 days. An identical experimental schedule was followed during the preparatory (February-March), pre-spawning (April-May), spawning (July-August), and post-spawning (September-October) phases of the annual cycle. Irrespective of the reproductive status of the carp, LL suppressed while DD increased the mid-day and mid-night values of melatonin compared to respective controls. Influences of exogenous melatonin varied in relation to the dose and duration of treatment and the reproductive status of the carp. However, testicular response to exogenous melatonin (at 100 µg, for 30 days) and DD in each reproductive phase was almost identical. Notably, precocious testicular maturation occurred in both DD and melatonin-injected fish during the preparatory phase and in LL carps during the pre-spawning phase. In contrast, testicular functions in both the melatonin-treated and DD fish were inhibited during the pre-spawning and spawning phases, while the testes did not respond to any treatment during the post-spawning phase. In conclusion, this study provided the first experimental evidence that melatonin plays a significant role in the regulation of annual testicular events in a sub-tropical surface-dwelling carp Catla catla, but the influence of this pineal hormone on the seasonal activity of testis varies in relation to the reproductive status of the concerned fish.  相似文献

将750尾建鲤随机分成5组,第1组为对照组,投喂基础日粮,另外4组为试验组,投喂基础日粮中分别添加0.5%、1.0%、2.0%、4.0%大黄蒽醌提取物。从2005年7月到9月连续投喂70d后,对鱼体进行高密度应激,测定其生长、应激前后血液皮质醇、血糖、溶菌酶等变化。结果表明:与对照组相比,应激前添加大黄葸醌提取物提高了鱼体增重率、特定生长率、鱼体丰满度、血液溶菌酶活性,降低了饵料系数与鱼体死亡率,但是与大黄葸醌提取物的添加水平不成线性关系,其中2.0%试验组还显著降低了血液皮质醇。高密度应激1d后,各组血液中的皮质醇、血糖、溶菌酶都有不同程度的增加,其中对照组最高,添加1.0%和2.0%大黄蒽醌提取物的试验组相对较低。应激前后各组鱼的攻毒试验表明:除应激前添加1.0%和2.0%大黄蒽醌提取物的组没有死鱼外,其它各组都有死鱼,其中对照组死亡率最高。因此添加1.0%-2.0%大黄蒽醌提取物提高了机体抗应激能力,并对病原菌感染起一定的保护作用,促进了鱼体生长。  相似文献

本文用流式细胞术(FCM)和免疫细胞化学方法,检测了实验性小鼠脾的淋巴细胞增殖周期和5-溴脱氧尿苷阳性细胞(BrdU ̄+)在脾脏内的分布.以及脾小结中IgM ̄+细胞的反应。结果显示由利血平诱发的脾虚小鼠S期细胞数最少(占5%),G_1和G_2+M期细胞增加。经过健脾汤治疗后S期细胞增至10%,与对照组比较无显著性差异.但高于自然恢复组(P<0.001)。经免疫细胞化学显示:脾虚组小鼠的脾中5-溴脱氧尿苷阳性细胞(BrdU ̄+)的数量远低于其它组。脾小结缩小,帽状区IgM ̄+B细胞基本消失,经健脾汤治疗后,复健组的脾小结与自然恢复组比较,体积明显增大,生发中心的BrdU ̄+和帽状区IgM ̄+B淋巴细胞均相应增多。提示健脾汤有促进DNA和IgM合成及细胞增殖作用。  相似文献

鲤(Cyprinus carpio)胰岛素样生长因子2b(insulin-like growth factor 2b,IGF2b)基因为鲤IGF基因家族重要的成员,已有的研究表明5'侧翼区序列对其功能的发挥有重要作用。因此,采用基因组步移法获取5'侧翼区序列,通过亚硫酸盐修饰的方法分析不同品种该区域GC富集区的甲基化状况。本文成功从鲤基因组DNA中获得695 bp的IGF2b 5'侧翼区序列。已获取的IGF2b 5'侧翼区序列经过TFSEARCH分析后发现了多个转录因子结合位点和TATA框,通过BLAST比对发现,鲤IGF2b 5'侧翼区DNA序列与斑马鱼(Danio rerio)IGF2b基因相比,相似性为87%。检测该区域富含GC的序列17个位点,发现建鲤(C.c.var.jian)有2个个体的3个位点出现甲基化,而黄河鲤(C.c.haematopterus)只有1个个体1个位点出现甲基化,这说明2个品种鲤在这个区域出现甲基化修饰的几率低,也验证了这2品种在该基因该区域GC富集区是稳定的。  相似文献
