Studying dynamic process of photosensitization by ESR method

By kinetic methods, functional relation of TAN radical, produced in the process of TEMPONE trapping O2, vs. time during photosensitization was established. Accordingly relative rate constants of generating all kinds of active intermediates through types I and II mechanism of photosensitization can be calculated. Using the formula and experimental results, the relative rate constants of generating O2, O2 and PS2 of three kinds of perylenequinone photosensitizer: HA, HB and CP in DMF-H20 and DMSO-H2O system were calculated, and then the structure-activity relationship of perylenequinone photosensitizer and the relation between photosensitivity and solvent was studied.     

Gramicidin derivatives as membrane-based pH sensors

Ion channels provide a means for sensitive pH measurement at membrane interfaces. Detailed knowledge of the structure and function of gramicidin channels permits the engineering of pH-sensitive derivatives. Two derivatives, gramicidin-ethylenediamine and gramicidin-histamine, are shown to exhibit pH-dependent single-channel behaviour over the pH ranges 9-11 and 6.5-8.5, respectively. Thermal isomerization of a carbamate group at the entrance of the channels leads to a pattern of steps in single-channel recordings. The size of the steps depends on the time-averaged degree of protonation of the appended group (ethylenediamine or histamine). Measurement of the size of the steps thus permits single-molecule pH sensing under symmetrical pH conditions or in the presence of a pH gradient.     

中国仓鼠卵巢工程细胞无血清流加培养关键工艺参数的考察

主要考察流加培养基中不同营养成分、流加起始时间及初始接种密度对11G-S细胞无血清流加培养的影响。在研究中以悬浮适应的表达尿激酶原 (Pro-urokinase,Pro-UK) CHO工程细胞系11G-S为研究对象,在100 mL的摇瓶中无血清悬浮流加培养11G-S细胞,同时以活细胞密度、细胞活力及Pro-UK活性为评价依据。结果表明在培养基中氨基酸、无血清添加成分及无机盐对促进细胞生长、细胞活力维持及蛋白表达起着较为重要的作用;且流加起始时间为72 h及初始接种密度为3×105~4×105 cells/     

基于层次分析法的江苏宁镇山脉乡土树种评价

为了筛选适宜南京地区的优良城市绿化树种,在野外调查和问卷调查的基础上,利用层次分析法(AHP)建立江苏宁镇山脉乡土树种评价体系,对该区域的乡土树种进行客观的定量分析。结果表明:(1)乡土树种作为城市绿化树种应首先考虑抗逆性,其次是生态价值、美学价值和生物学特性;(2)参与评价的41种宁镇山脉乡土树种可分为4类,其中I类乡土树种应大力推广应用,Ⅱ类乡土树种可作为森林城市树种多样性的有益补充,Ⅲ类和Ⅳ类乡土树种作为城市绿化树种仍有一定的差距。13种Ⅰ类乡土树种作为城市绿化树种具有良好的应用前景。     

Algal Population Growth Model Integrated with Toxicokinetics for Ecological Risk Assessment under Time-Varying Pesticide Exposure

An algal population growth model integrated with toxicokinetics was developed for assessing the effect of pesticides on population dynamics. This model is a simple one-compartment, first-order kinetic model in which toxicity (growth inhibition and mortality) depends on the intracellular effective concentration of a pesticide at a target site. The model's parameters were derived using an experimental study that investigated the effects of pretilachlor, bensulfuron-methyl, pentoxazone, and quinoclamine on the growth, mortality, and subsequent population recovery of the green alga Pseudokirchneriella subcapitata. Modeled and measured trajectories of algal population dynamics agreed well. The effect on population recovery was underestimated by the model that ignored the toxicokinetics. The four tested herbicides had a variety of toxicity characteristics and physicochemical properties, indicating the wide range of the model's applicability. Moreover, the developed model and the obtained model's parameters were extrapolated to predict long-term algal population dynamics under time-varying herbicide exposure. The calculated integral biomass lost compared with the control was considered a quantitative index of the population-level ecological risk. The model's prediction showed that the same exposure level (peak concentration is equivalent to EC50) indicated much different population-level effect depending on the herbicide.     

CELL DIVISION AND MORPHOGENESIS OF THE CENTRIC DIATOM CHAETOCEROS DECIPIENS (BACILLARIOPHYCEAE) II. ELECTRON MICROSCOPY AND A NEW PARADIGM FOR TIP GROWTH

Valve morphogenesis starts when the silica deposition vesicle (SDV) expands across a cleavage furrow covered by an unidentified layer, which may aid in its shaping. A labiate process (LP) is present only in the outer valve of terminal cells in the filament. Before these particular cells form setae, a layered "labiate process apparatus" (LPA) appears on the SDV in the exact center of the forming valve, near the microtubule center arising after cleavage. The LPA thereafter surmounts the lips of the LP as it forms. After the girdle bands separate slightly, two lateral protrusions develop in the corners of the cell. These nascent setae are lined internally by a cylindrical, fibrous band (sleeve), which assembles immediately ahead of the expanding edge of the SDV, very close to the plasmalemma. Then these protrusions, lined by the fibrous band, the SDV, and the forming silica wall, grow through two gaps in the girdle bands. The cytoplasm at the tip of the growing seta is naked. Immediately behind the tip, this fibrous band is adpressed to the plasmalemma and thereby apparently defines the diameter of the seta; it extends to internally ensheath the tipmost edge of the SDV for a short distance, like a tight-fitting inner sleeve. This structure is considered the major organelle involved in seta morphogenesis. Microtubules (MTs), while present, are variable in extent and disposition within the seta. Turgor pressure is considered irrelevant in driving seta growth. Instead, a new paradigm proposed for tip-growing cells generally, may apply to seta morphogenesis, as follows. If, as is suspected, the fibrous band contains actin, cycling of this actin (as in animal cells undergoing ruffling or filopodial extension) could drive seta extension via attachment of the band to the just-formed silica wall. The band is visualized as a molecular treadmill whose support base, the new wall, is being continually extended; extension is controlled and generated strictly at the tip.     

黑河中游湿地景观破碎化过程及其驱动力分析

在遥感和GIS技术支持下,基于1975-2010年长时间序列遥感影像,选取斑块密度指数(PD)、景观内部生境面积指数(IA)、斑块平均面积指数(MPS)、斑块形状破碎化指数(FS1、FS2)等具有典型生态意义的景观指数模型,系统分析了黑河中游湿地景观的破碎化过程,并结合灰色关联分析、主成分分析等方法,探讨了影响研究区湿地景观破碎化过程的各驱动因子.结果表明:近35年来,研究区湿地景观破碎化主要表现为斑块平均面积的萎缩,斑块密度的上升以及斑块形状破碎化指数的增大.整个研究时段内,研究区湿地斑块平均面积减少了48.95hm2,斑块密度的上升0.006个/hm2;导致黑河中游湿地景观破碎化发生和发展的驱动力包含自然和人文两个方面.自然因子对湿地景观破碎化进程的影响则主要体现在气温和降水上,而且气温对湿地景观破碎化进程的影响程度明显大于降水.但在1975-2010年间的这一较小时间尺度上,人类活动对湿地景观破碎化的贡献率明显高于自然因子,人类活动能力的增强以及影响范围的不断扩大是引发黑河中游湿地景观破碎化的主因.
