Azo coupling reactions of N-α-acetylhistidine, N-α-acetyltyrosine, and N-α-acetyllysine with p-methylbenzenediazonium ion were investigated as model reactions to obtain information on the relative reactivity of the histidine, tyrosine, and lysine moieties of protein, separated from structural effects. The azo coupling yields of the amino acids increased as the pH of the reaction medium was increased, indicating that the ractive species are the imidazole anion of histidine, the phenolate anion of tyrosine, and the neutral ε-amino group of lysine. It was calculated, based on percentage yields of the azo products, that the imidazole anion is more reactive than the phenolate anion and the ε-amino group, respectively.  相似文献

The mechanism by which patterns are produced appears to be repeated in each segment of an animal, and it has been proposed that it may even have been conserved in evolution so that different species would have the same system of positional information. This idea has been tested by mixing cells of a defined fragment of the wing disc of Drosophila melanogaster with wing disc fragments of five other dipteran species to assay the ability of these disc fragments to stimulate intercalary regeneration of the D. melanogaster cells. The genetically marked (y; mwh) D. melanogaster fragment was mechanically mixed with wing discs or wing disc fragments of four drosophilids (D. melanogaster as a control, D. virilis, D. hydei, Zaprionus vittiger), of Musca domestica, and of Piophila casei. The mixed aggregates were cultured in vivo for 7 days, then metamorphosed in D. melanogaster larval hosts. The D. melanogaster fragments were only stimulated to regenerate when combined with complementary fragments from D. melanogaster or D. virilis wing discs. In the combination between D. melanogaster and D. hydei, the tissue formed integrated mosaic patterns, but no regeneration ensued. The one positive result (D. melanogaster mixed with D. virilis) shows that positional cues can be exchanged and correctly interpreted between cells of different species. The negative results do not prove that the mechanism for establishing patterns is different in the tested species, but may be due to incompatibilities that are not related to pattern formation.  相似文献

Evidence is presented to show that both the chromosomal replication point and the chromosomal origin in Escherichia coli are associated with a structure possessing the sedimentation properties and enzyme sensitivities characteristic of membrane. The data suggest that both newly synthesized DNA strands and template DNA strands are bound at this replication point and that both strands are also bound at the origin.  相似文献

This study is concerned with the surgical technique for the injection of a catheter through arteries with overlapping stenosis in the presence of externally applied magnetic field and Hall currents influences. The nature of blood is analyzed mathematically by considering it as a micropolar fluid. The analysis is carried out for an artery with a mild stenosis. The governing equations with the corresponding boundary conditions solved numerically using Crank–Nicolson implicit finite difference scheme. The numerical technique give excellent agreement for axial velocity of the fluid, the circumferential microrotation, the wall shear stress distribution and the contour plots of stream lines. The obtained results show that the value of axial velocity is higher for a Newtonian fluid than that for a micropolar fluid model, the effect of suitable moving magnetic field (Hall currents influences) accelerates the speed of blood, the size of trapped bolus for the stream lines decrease if the spinning movement of the fluid molecules have considerable value regardless of small or large size of the fluid molecules and the flow of fluid is better with increasing the Hall current effect and the size of trapping bolus increase clearly by increasing the maximum height of stenosis where the fluid moves as a bulk.  相似文献

The versatility of insertional inactivation of β-galactosidase activity for subcloning and sequencing has been enhanced by combining a chemically synthesized oligonucleotide which specifies nine 6-bp-cutter restriction sites including BglII, XhoI, NruI, ClaI, SacI and EcoRV in various configurations with existing polylinkers to create a set of highly versatile cloning sites. These improved polylinkers have been inserted into plasmids (the pICs) for routine cloning of double-stranded DNA, and into chimeric phage/plasmids (the pICEMs) for biological production of single stranded DNA. The most versatile Polylinker specifies 17 restriction sites in the β-galactosidase α-complementing gene fragment. One of the new polylinkers was inserted into M 13 DNA to produce a vector (M13mIC7) with nine cloning sites.  相似文献

In this essay, I respond to the reviewers of my book, Racism, Class and the Racialized Outsider, including Bhattacharrya, Frost, Jefferys, Meer, Roediger and van der Linden. In particular, I elaborate further on the epistemological foundations of the book, including my aim to stretch the concept of working class to accommodate both ethnic diversity, and the significance of racism and anti-racism within it. Then, I state the case for how the concept of the racialized outsider can help transform our understanding of the key signposts of English working-class history. The significance of the anti-racist accomplishments of the 1970s and 1980s are also given further consideration alongside the relevance of the book and its arguments to the present environment – a period where those accomplishments have started to be reversed.  相似文献

The permeability of rat heart myocytes to Na increases when extracellular Ca (Ca0) is decreased. This increased permeability is reflected in elevated NaK ratios in nonenergized myocytes and in increased ouabain-sensitive lactate production in anaerobic myocytes supplemented with glucose. Myocytes treated with ethylene glycol bis(β-aminoethyl ether)N,N′-tetraacetic acid (1 mm) maintain low NaK ratios, but expend considerable glycolytic ATP in ouabain-sensitive cation cycling. The data suggest that Ca0 bound to the sarcolemma can restrict transmembrane movement of Na via pathways that are not yet defined. The lack of significant net accumulation of Ca argues against the explanation that Ca0 maintains low internal Na levels as a result of NaCa exchange. Both the increased uptake of Na and increased utilization of ATP in the absence of Ca0 may be relevant to the phenomenon of “Ca-paradox” in situ.  相似文献

Carcinogenic arylhydroxamic acids (N-hydroxy-2-acetylamino-fluorene, N-hydroxy-4-acetylaminobiphenyl, and trans-N-hydroxy-4-acetyl-aminostilbene) are irreversible inhibitors of rat and hamster hepatic N,O-acyltransferase. The kinetic characteristics of this inhibition are consistent with suicide inactivation of the enzyme. This finding appears to be the first example of carcinogens serving as suicide substrates for an enzyme system which is responsible for their bioactivation.  相似文献
