The isolated frog skin, bathed with Li+-Ringer (Na+-free) on the outside and Na+-Ringer on the inside, can maintain a normal potential difference (PD) and short-circuit current (s.c.c.) for more than 6. h. The s.c.c. correspondended to the Li+ influx. The Na+ efflux was 4% of the s.c.c. 10−5 M ouabain depressed Li+ influx and s.c.c. 1010−5 M amiloride abolished the Li+ s.c.c., while 0.1 unit/ml oxytocin stimulated it. When the inside of the skin was bathed with Li+-Ringer, PD and s.c.c. fell to zero within 2 h. The oxygen consumption of skin slices bathed in Li+-Ringer was 29% lower than controls bathed in Na+-Ringer.When the isolated frog skin is bathed in Na2SO4-Ringer it shows electrical rectification which has been correlated with the active transport of Na+. In skins transporting Li+, rectification characteristics are similar to those of skins transporting Na+. When the inner face of the skin is bathed with Li+-Ringer, rectification, PD and s.c.c. decline in a parallel fashion.It is concluded that: (1) Li+ can be transported when Na+ is present at the inner face. (2) Amiloride, ouabain and oxytocin affect Li+ and Na+ transport in a similar manner. (3) Li+ transport, like Na+ transport, is associated with rectification. (4) Active transport of Na+ and Li+ seems to depend on two different but associated proceses; one taking place at the external barrier (where rectification occurs) as shown by the effect of amiloride; and the other of an inner site related to energy requirements and affected by ouabain and Li+. (5) The cation being transported is not necessarily activating the (Na+-K+-ATPase.  相似文献

Two novel Brachyspira hyodysenteriae-specific DNA fragments, designated as Bh100 and Bh400, were identified using representational difference analysis. To isolate the fragments the combined DNA of the Brachyspira pilosicoli, Brachyspira intermedia, Brachyspira murdochii and Brachyspira innocens reference strains was subtracted from the genome of B. hyodysenteriae strain B204. Both fragments were present in a single copy and mapped to different positions on the genome of B. hyodysenteriae B78(T). Larger fragments encompassing the continuous open reading frames (ORF) of Bh100 and Bh400 were cloned and analysed. Whereas the ORF of 2130 bp encompassing Bh100 did not show homology to any known bacterial protein, Bh400 was part of a putative operon with significant homology to the phosphotransferase system of Bacillus subtilis.  相似文献

1. In Rhodopseudomonas sphaeroides the Qx absorption band of the reaction center bacteriochlorophyll dimer which bleaches on photo-oxidation is both blue-shifted and has an increased extinction coefficient on solubilisation of the chromatophore membrane with lauryldimethylamine-N-oxide. These effects may be attributable in part to the particle flattening effect.2. The difference spectrum of photo-oxidisable c type cytochrome in the chromatophore was found to have a slightly variable peak position in the α-band (λmax at 551–551.25 nm); this position was always red-shifted in comparison to that of isolated cytochrome c2 (λmax at 549.5 ± 0.5 nm). The shift in wavelength maximum was not due to association with the reaction center protein. A possible heterogeneity in the c-type cytochromes of chromatophores is discussed.3. Flash-induced difference spectra attributed to cytochrome b were resolved at several different redox potentials and in the presence and absence of antimycin. Under most conditions, one major component, cytochrome b50 appeared to be involved. However, in some circumstances, reduction of a component with the spectral characteristics of cytochrome b?90 was observed.4. Difference spectra attributed to (BChl)2, Q?II, c type cytochrome and cytochrome b50 were resolved in the Soret region for Rhodopseudomonas capsulata.5. A computer-linked kinetic spectrophotometer for obtaining automatically the difference spectra of components functioning in photosynthetic electron transfer chains is described. The system incorporates a novel method for automatically adjusting and holding the photomultiplier supply voltage.  相似文献

摘要 目的:随着夜间光照与肥胖现象日趋普遍化,机体内的昼夜节律与生理代谢均受到不同程度的影响,其中不同性别间的差异有待进一步研究。本文旨在通过比较高脂饲料喂养,以及改变光照周期从而对不同性别小鼠体重产生的影响来模拟高脂肪饮食时人类受到夜间光照的性别差异。方法:随机选取C57BL/6小鼠雌鼠、雄鼠各12只,分别按照性别、喂养饲料随机均分成4组,其中随机选取一组雌鼠和一组雄鼠喂养60%高脂饲料作为实验组,另外的一组雌鼠和一组雄鼠喂养普通饲料作为对照组,连续喂养8周,观察并记录各组小鼠体重变化;第1~6周将实验小鼠置于24h/0h光照周期下生活,第6~8周实验小鼠生活的光照周期改为12h/12h。结果:在高脂饲料喂养以及光照周期失调(24h/0h)的条件下,雌、雄鼠体重均有增长(P<0.05),但雄鼠体重增加量是雌鼠的三倍。当光照周期恢复正常(12h/12h)后,雌、雄小鼠体重增长量均下降。不同性别的小鼠食用高脂饲料以及生物节律被打乱时体重均增加,但雄鼠体重增加量明显大于雌鼠,提示同在高脂饲料饮食的条件下,雌鼠受生物节律影响比雄鼠更大。结论:高脂饮食实验小鼠模型中雌鼠受生物节律的影响比雄鼠更大。  相似文献

为了采用广义加法模型整合数字高程模型和遥感数据进行植被分布的预测, 并探索耦合环境变量和遥感数据作为预测变量是否能够有效地提高植被分布预测的精度, 选择海拔、坡度、至黄河最近距离、至海岸线最近距离, 以及从SPOT5遥感影像中提取的光谱变量作为预测变量, 采用广义加法模型整合环境变量和光谱变量, 建立植被分布预测模型。研究设置3种建模情景(以环境变量作为预测变量, 以光谱变量作为预测变量, 综合使用环境变量与光谱变量作为预测变量)对黄河三角洲的优势植被类型的分布进行了预测, 并对预测结果采用偏差分析、受试者工作特征曲线和野外采样点对比等3种方法进行了验证。结果表明: (1)基于广义加法模型的植被分布预测方法具有一定的实用性, 可以较为准确地预测植被的分布; 盖度较高的植被类型预测精度较高, 盖度较低的植被类型预测精度较低, 植物群落结构的特点是出现这些差异的主要原因; 综合使用环境变量和光谱变量作为预测变量的模型, 预测精度高于单独以环境变量或者光谱变量作为预测变量的模型。(2)环境变量、光谱变量大多被选入模型, 二者均对植被分布预测有重要的作用; 同一预测变量在不同植被类型的预测模型中的贡献不同, 这与植被的光谱、环境特征差异有关; 同一预测变量在不同的建模情景下对模型的贡献不同, 环境变量与光谱变量的耦合效应可能是导致预测变量对模型的贡献出现变化的原因。  相似文献

AIMS: Identification of genes differentially present in Haemophilus parasuis serovar 2 by representational difference analysis (RDA). METHODS AND RESULTS: Bacterial genomic DNA was extracted, cleaved with Sau3AI and ligated to oligonucleotide adapter pair. The optimal tester (H. parasuis serovar 2)/driver ratio (H. parasuis serovars 1, 3 and 5) for the hybridization was established and the mixture was hybridized, and amplified by PCR. The products were cloned and transformed into Escherichia coli TOP10 cells and checked for specificity by Southern blotting analysis. The RDA subtractive technique yielded six bands ranging from 1500 to 200 bp, which were cloned into pCR II-TOPO vector and 40 clones were analysed. A fragment of 369 bp was specific for H. parasuis serovar 2, and showed 99% homology to sulI gene encoding for dihydropteroate synthase (dhps). The dhps gene conferring sulfonamide resistance was detected in H. parasuis serovar 2 but was absent in serovars 1, 3, 5 and in most of the Actinobacillus pleuropneumoniae serotypes (except serotype 7). CONCLUSION: sulI allele of dihydropteroate synthase has been identified in H. parasuis serovar 2 by RDA technique. SIGNIFICANCE AND IMPACT OF THE STUDY: The RDA technique seems to be an useful method for the identification of genes that are differentially present in H. parasuis, a respiratory pathogen of veterinary interest.  相似文献

Climate warming is likely inducing carbon loss from soils of northern ecosystems, but little evidence comes from large-scale observations. Here we used data from a repeated soil survey and remote sensing vegetation index to explore changes in soil organic carbon (SOC) stock on the Tibetan Plateau during the past two decades. Our results showed that SOC stock in the top 30 cm depth in alpine grasslands on the plateau amounted to 4.4 Pg C (1 Pg=1015 g), with an overall average of 3.9 kg C m−2. SOC changes during 1980s–2004 were estimated at −0.6 g C m−2 yr−1, ranging from −36.5 to 35.8 g C m−2 yr−1 at 95% confidence, indicating that SOC stock in the Tibetan alpine grasslands remained relatively stable over the sampling periods. Our findings are nonconsistent with previous reports of loss of soil C in grassland ecosystems due to the accelerated decomposition with warming. In the case of the alpine grasslands on the Tibetan Plateau studied here, we speculate that increased rates of decomposition as soils warmed during the last two decades may have been compensated by increased soil C inputs due to increased grass productivity. These results suggest that soil C stock in terrestrial ecosystems may respond differently to climate change depending on ecosystem type, regional climate pattern, and intensity of human disturbance.  相似文献

以pSET152和pHZ1358为出发质粒,通过供体大肠杆菌ET12567(pUZ8002)和S17-1接合转移斑贝链霉菌(Streptomycesbambergiensis),构建和优化了接合转移体系。采用OOE-PCR技术构建含ermEp*与vhb结构基因融合片段的整合型表达载体pBIB2005,转化ET12567(pUZ8002)后,属间接合转移至斑贝链霉菌。通过PCR、CO结合差光谱验证vhb基因在斑贝链霉菌中整合表达。摇瓶发酵显示VHb蛋白能改善菌体对氧的需求,一定程度上促进细胞生长,提高抗生素产量。  相似文献

Climate control on global vegetation productivity patterns has intensified in response to recent global warming. Yet, the contributions of the leading internal climatic variations to global vegetation productivity are poorly understood. Here, we use 30 years of global satellite observations to study climatic variations controls on continental and global vegetation productivity patterns. El Niño‐Southern Oscillation (ENSO) phases (La Niña, neutral, and El Niño years) appear to be a weaker control on global‐scale vegetation productivity than previously thought, although continental‐scale responses are substantial. There is also clear evidence that other non‐ENSO climatic variations have a strong control on spatial patterns of vegetation productivity mainly through their influence on temperature. Among the eight leading internal climatic variations, the East Atlantic/West Russia Pattern extensively controls the ensuing year vegetation productivity of the most productive tropical and temperate forest ecosystems of the Earth's vegetated surface through directionally consistent influence on vegetation greenness. The Community Climate System Model (CCSM4) simulations do not capture the observed patterns of vegetation productivity responses to internal climatic variations. Our analyses show the ubiquitous control of climatic variations on vegetation productivity and can further guide CCSM and other Earth system models developments to represent vegetation response patterns to unforced variability. Several winter time internal climatic variation indices show strong potentials on predicting growing season vegetation productivity two to six seasons ahead which enables national governments and farmers forecast crop yield to ensure supplies of affordable food, famine early warning, and plan management options to minimize yield losses ahead of time.  相似文献
