Likelihood ratio tests are derived for bivariate normal structural relationships in the presence of group structure. These tests may also be applied to less restrictive models where only errors are assumed to be normally distributed. Tests for a common slope amongst those from several datasets are derived for three different cases – when the assumed ratio of error variances is the same across datasets and either known or unknown, and when the standardised major axis model is used. Estimation of the slope in the case where the ratio of error variances is unknown could be considered as a maximum likelihood grouping method. The derivations are accompanied by some small sample simulations, and the tests are applied to data arising from work on seed allometry.  相似文献

The classical group sequential test procedures that were proposed by Pocock (1977) and O'Brien and Fleming (1979) rest on the assumption of equal sample sizes between the interim analyses. Regarding this it is well known that for most situations there is not a great amount of additional Type I error if monitoring is performed for unequal sample sizes between the stages. In some cases, however, problems can arise resulting in an unacceptable liberal behavior of the test procedure. In this article worst case scenarios in sample size imbalancements between the inspection times are considered. Exact critical values for the Pocock and the O'Brien and Fleming group sequential designs are derived for arbitrary and for varying but bounded sample sizes. The approach represents a reasonable alternative to the flexible method that is based on the Type I error rate spending function. The SAS syntax for performing the calculations is provided. Using these procedures, the inspection times or the sample sizes in the consecutive stages need to be chosen independently of the data observed so far.  相似文献

New neprilysin inhibitors containing an α-mercaptoketone HSC(R1R2)CO group, as zinc ligand were designed. Two parameters were explored for potency optimization: the size of the inhibitor which could interact with the S1, S1′ or S2′ domain of the enzyme and the nature of the substituents R1, R2 of the mercaptoketone group. Introduction of a cyclohexyl chain in R1, R2 position and a (3-thiophen)benzyl group in position R3 (compound 12n) yielded to the most potent inhibitor of this series with a Ki value of 2 ± 0.3 nM. This result suggests that this new inhibitor interacts within the S1, S1′ domain of NEP allowing a pentacoordination of the catalytic Zn2+ ion by the mercaptoketone moiety.  相似文献

The age of the sedimentary basement of the Ganga Valley in northern India, which is represented by the entirely subsurface Ganga Supergroup, is key for addressing issues related to the tectonic history of the Himalaya. However, the stratigraphic correlations between the Ganga Supergroup in the Ganga Valley, the Vindhyan Supergroup in cratonic India to the south, and Proterozoic successions in the Lesser Himalaya to the north have long been a matter of controversy. This is largely because of the poor age constraint of the Madhubani Group of the upper Ganga Supergroup, which has been variously interpreted as Proterozoic, lower Palaeozoic, or even Mesozoic. To address this issue, we used a low manipulation maceration technique to extract organic‐walled microfossils from the Ujhani and Tilhar formations of the lower Madhubani Group. Our study recovered a total of 24 taxa, including Devisphaera corallis gen. et sp. nov. The co‐occurrence of Trachyhystrichosphaera aimika, Caudosphaera expansa and Annulusia annulata in the lower Madhubani Group indicates a late Mesoproterozoic to early Neoproterozoic age. Thus, the biostratigraphical data suggest a >300 myr depositional gap between the Madhubani Group and the immediately underlying Bahraich Group, which has been independently constrained to be upper Palaeoproterozoic to lower Mesoproterozoic in age. Therefore, the first‐order stratigraphic architecture, with a Palaeoproterozoic–Mesoproterozoic succession unconformably overlain by a Mesoproterozoic–Neoproterozoic succession, is closely similar throughout the Vindhyan Basin, Ganga Valley and Lesser Himalaya, suggesting a shared sedimentary and tectonic history among them.  相似文献

介绍日本中生代晚期手取群研究的最新进展,并简要评述与中国的对比。手取群从下到上分为九头龙亚群、石白亚群、赤岩亚群。庄川地区九头龙亚群御手洗组新发现菊石,属Tithonian-Berriasian期,改变了以往Callovian期的传统意见。石白亚群发现菊石,属Late Hauterivian-Early Barremian期;亚群上部发现海相双壳类,属Hauterivian期,半咸水相和淡水双壳类也与外带Hauterivian期的对比;绝对年龄测定为135±7Ma和128±8Ma,分别属Hauterivian早期和Barremian早期,其中135Ma被普遍接受;植物群反映的古气候特征与俄罗斯东部Valanginian-Hauterivian期温湿化一致。石白亚群的时代综合为Valanginian-Hauterivian期。赤岩亚群的Trigonioides(Wakinoa)tetoriensis也发现于外带,时代属latest Hauterivian-Early Barremian期;绝对年龄测定数据不稳定,自127±8Ma(Barremian中期)至106±7Ma(Albian中期)。对亚群的Nippononaia ryosekiana作了修订,认为应属N.linhaiensis,与浙江馆头组的同种,比濑林组(Lower Aptian)的N.ryosekiana原始,故时代早于Aptian期。赤岩亚群的时代综合为Barremian期,但不排除上延的可能。区域对比结果认为,中国东北龙爪沟群及绥滨地区的相当地层可与九头龙亚群和石白亚群对比;馆头组、下城子组可与赤岩亚群对比;热河群上部沙海组-阜新组煤系地层可与石白亚群对比;并对义县组的时代提出了看法。  相似文献

Biolayer interferometry is a novel method for quantifying macromolecules, such as proteins, in solution. The presence of other, non-binding molecules does not interfere with quantification, which allows one to measure the concentration of the molecule of interest in a crude mixture. The isolation selectivity of the isoflavonoid puerarin, a well-known traditional Chinese medicine, was studied when native and oligo-β-cyclodextrin (CDP) coupled polystyrene-based macroporous resins were used as adsorbents by static tests. The research results indicated that the CDP coupled resin HPD-100-CDP offered the best adsorption and desorption capacity for puerarin than others and its equilibrium adsorption data at 25 °C fit best to the Freundlich isotherm. The performance of separation of puerarin on HPD-100-CDP column in one step was evaluated. Based on the above experimental data, a novel medium PS-CDP was synthesized and its chromatographic retention behaviors were also explored. ESI-MS/MS, 1H NMR spectroscopy and UV absorption spectrum were used for the detection and characterization of puerarin in isolated fraction. Under the optimum mobile phase, methanol/acetic acid/water = 5.0/6.6/88.4 (v/v/v), the purity and recovery of puerarin were 95.3% and 86.7%, respectively, by HPLC analysis. In conclusion, the PS-CDP medium can enhance the isolated selectivity of puerarin and it can be applied in preparative scale operations.  相似文献

Acetyl phosphate (acetyl-P) serves critical roles in coenzyme A recycling and ATP synthesis. It is the intermediate of the Pta-AckA pathway that inter-converts acetyl-coenzyme A and acetate. Acetyl-P also can act as a global signal by donating its phosphoryl group to specific two-component response regulators. This ability derives from its capacity to store energy in the form of a high-energy phosphate bond. This bond, while critical to its function, also destabilizes acetyl-P in cell extracts. This lability has greatly complicated biochemical analysis, leading in part to widely varying acetyl-P measurements. We therefore developed an optimized protocol based on two-dimensional thin layer chromatography that includes metabolic labeling under aerated conditions and careful examination of the integrity of acetyl-P within extracts. This protocol results in greatly improved reproducibility, and thus permits precise measurements of the intracellular concentration of acetyl-P, as well as that of other small phosphorylated molecules.  相似文献

Studies on leadership during group movements in several lemur species showed that females were responsible for the travelling choices concerning time and direction. Interestingly, in these species females are dominant over males. We investigated the influence of social organisation upon leadership processes by studying a lemur species in which social organisation is characterized by the absence of female dominance: the brown lemur (Eulemur fulvus fulvus). The study was conducted on a semi-free ranging group of 11 individuals and the analysis performed on 69 group movements showed that all the individuals could initiate a group movement. In 34 cases, the whole group moved. There was no significant difference in the number of start attempts or in the number of group members involved from one initiator to another. Moreover, there was no effect of sex or age of the initiator on the number of individuals following it or on the speed of the joining process. Therefore, the leadership observed is widely distributed to all group members. These results support the hypothesis of an influence of social organisation upon the decision-making processes but still remain to be studied in a more relevant ecological context.  相似文献
