在67只切断两侧缓冲神经的麻醉Sprague-Dawley大鼠,应用细胞外记录的电生理方法和免疫组织化学技术,分别观察肾缺血对延髓腹外侧头端区巨细胞旁外侧核神经元自发放电活动和Fos蛋白表达的影响.所得结果如下(1)左肾动脉阻断后,28个单位的放电频率由11.40±1.08增至21.1±1.74spikes/s(P<0.001),血压和心率无明显变化(P>0.05);(2)在17个放电单位中,应用腺苷受体拮抗剂8-苯茶碱(8-phenyltheophylline,10mg/kg)可明显抑制肾缺血的兴奋效应(P<0.05);(3)肾缺血后,延髓腹外侧头端区的Fos蛋白样免疫反应神经元显著增加(P<0.01);(4)预先应用8-苯茶碱可明显减弱肾缺血所激活的Fos蛋白表达反应(P<0.05).以上结果提示肾缺血增强延髓腹外侧头端区神经元的放电活动和Fos蛋白表达,而此作用可能与肾脏缺血所产生的腺苷激活肾内感受器有关.  相似文献

Summary Lactic acid production by an isolated ofLactobacillus plantarum was standardised on enzyme-hydrolysed tapioca (Manihot esculenta) flour, tapioca starch and soluble starch. Calculated yields of lactic acid (g from 100 g reducing sugars used) in nutrient media containing the abovementioned hydrolysates (10% reducing sugars) were 21.8%, 16.2% and 16.2%, respectively. Higher yields (29–34%) were obtained in media containing 5% reducing sugars. A conversion efficiency of 80–99% was achieved when the acid produced in the broth was neutralised periodically. One hundred milliliters of the medium (5% sugars) yielded 4.0–4.5 g of calcium lactate. These results indicate that unrefined starchy material can be successfully employed for the economic production of lactic acid. The same substrate can also be utilised for biomass production, as viable lactobacilli are being used for therapy in medicine.  相似文献

作为桉叶油的主要成分,桉叶素是具有多种生物活性的单萜化合物,被广泛应用于药品、食品及化妆品等领域。桉叶油主要从桉树叶提取,该过程耗费大量人力及自然资源,且容易污染环境。近年来,随着微生物代谢工程与合成生物学的快速发展,加上越来越多萜类生物合成途径得到解析,为桉叶素的绿色生产提供了新的途径。对桉叶素的生物合成途径、桉叶素合酶的结构与功能及近年来桉叶素的微生物合成进行了综述,并对利用微生物代谢工程合成桉叶素等单萜化合物的瓶颈问题及解决方案进行了探讨和归纳,为构建高产桉叶素等单萜微生物工程菌株提供参考。  相似文献

8-异戊烯基柑橘素促进体外培养成骨细胞成熟矿化的研究   总被引:1,自引:0,他引:1  
研究8-异戊烯基柑橘素对体外培养大鼠颅骨成骨细胞(rat skull osteoblasts,ROB)的分化成熟及生物矿化的影响.取新生大鼠颅骨多次酶消化法得到成骨细胞,培养于含10%FBS的MEM培养液中,3天后首次换液,待细胞铺满皿底传代培养.以碱性磷酸酶(alkaline phosphatase,ALP)为检测指标,96孔板梯度筛选作用最佳浓度,在最佳浓度作用并成骨性诱导培养的第3、6、9、12天测ALP活性、钙盐沉积量;第12天进行ALP和钙化结节组织化学染色及计数;成骨性诱导后不同时间点提取Total RNA,RT real-time PCR法检测成纤维细胞生长因子(bFGF)、胰岛素样生长因子-1(IGF-1)、成骨相关转录因子Osterix、Runx-2和骨形态发生蛋白-2(BMP-2)的基因表达情况;成骨性诱导的第4、8、12天裂解获得细胞总蛋白,蛋白质印迹法检测人Ⅰ型胶原蛋白(COL-Ⅰ)的蛋白质表达量.研究结果表明:1×10-6 mol/L能显著促进成骨细胞的成熟分化,表现为提高ROB的ALP活性、促进钙盐沉积、增加钙化结节数量;提高bFGF、IGF-1、Osterix、Runx-2和BMP-2 mRNA表达水平;促进COL-Ⅰ的合成.由此可知终浓度为1×10-6 mol/L 8-异戊烯基柑橘素能显著促进ROB的分化成熟及生物矿化,证明8-异戊烯基柑橘素能促进成骨细胞的分化成熟及生物矿化,作为促进骨修复和抗骨质疏松的有效成分具有较大的药用价值.  相似文献

The ubiquitylation cascade plays an important role in the recruitment of repair factors at DNA double-strand breaks. The involvement of a growing number of ubiquitin E3 ligases adds to the complexity of the DNA damage-induced ubiquitin signaling. Here we use the genetically tractable avian cell line DT40 to investigate the role of HERC2, RNF8 and RNF168 in the DNA damage-induced ubiquitylation pathway. We show that formation of ubiquitin foci as well as cell survival after DNA damage depends on both RNF8 and RNF168. However, we find that RNF8 and RNF168 knockout cell lines respond differently to treatment with camptothecin indicating that they do not function in a strictly linear manner. Surprisingly, we show that HERC2 is required neither for survival nor for ubiquitin foci formation after DNA damage in DT40. Moreover, the E3 ubiquitin ligase activity of HERC2 is not redundant to that of RNF8 or RNF168.  相似文献

This laboratory has recently reported the isolation of an ethanol-inducible form of rabbit liver microsomal cytochrome P-450, designated isozyme 3a. In view of the reports of others that the hepatotoxicity of acetaminophen is increased in ethanol-treated animals and the human alcoholic, we have determined the activity of the six available P-450 isozymes in the activation of the drug to give an intermediate which forms a conjugate with reduced glutathione. Isozymes 3a, 4, and 6, all of which are present in significant amounts in the liver microsomes from rabbits chronically administered ethanol, exhibited the highest activities in the reconstituted enzyme system, whereas isozymes 3b and 3c were 10- to 20-fold less effective, and phenobarbital-inducible isozyme 2 was essentially inactive, even in the presence of cytochrome b5. The results obtained thus indicate that induction by ethanol of P-450 isozyme 3a (or a homologous enzyme in other species) may contribute to the toxicity of acetaminophen but that other cytochromes also play a significant role.  相似文献

Human apolipoprotein E (apoE) is a 299-amino-acid protein with a molecular weight of 34 kDa. The difference between the apoE3 and apoE4 isoforms is a single residue substitution involving a Cys-Arg replacement at residue 112. ApoE4 is positively associated with atherosclerosis and late-onset and sporadic Alzheimer's disease (AD). ApoE4 and its C-terminal truncated fragments have been found in the senile plaques and neurofibrillary tangles in the brain of AD patients. However, detail structural information regarding isoform and domain interaction remains poorly understood. We prepared full-length, N-, and C-terminal truncated apoE3 and apoE4 proteins and studied their structural variation. Sedimentation velocity and continuous size distribution analysis using analytical ultracentrifugation revealed apoE3(72-299) as consisting of a major species with a sedimentation coefficient of 5.9. ApoE4(72-299) showed a wider and more complicated species distribution. Both apoE3 and E4 N-terminal domain (1-191) existed with monomers as the major component together with some tetramer. The oligomerization and aggregation of apoE protein increased when the C-terminal domain (192-271) was incorporated. The structural influence of the C-terminal domain on apoE is to assist self-association with no significant isoform preference. Circular dichroism and fluorescence studies demonstrated that apoE4(72-299) possessed a more alpha-helical structure with more hydrophobic residue exposure. The structural variation of the N-terminal truncated apoE3 and apoE4 protein provides useful information that helps to explain the greater aggregation of the apoE4 isoform and thus has implication for the involvement of apoE4 in AD.  相似文献

Bulky DNA adducts and 8-oxo-7,8-dihydro-2´-deoxyguanosine (8-oxodGuo) were measured in gill DNA of benzo[a]pyrene (B[a]P)-exposed mussels (50 mg kg-1 dw day-1), respectively by the 32P-post-labelling technique and high performance liquid chromatography coupled to electrochemical detection assay. A time-course study was performed for both biomarkers and their potential use for marine biomonitoring discussed for the sentinel species studied. Group A rotavirus (RVA) rarely causes severe complications such as encephalitis/encephalopathy. However, the pathophysiology of this specific complication remains unclear. Next-generation sequence analysis was used to compare the entire genome sequences of RVAs detected in patients with encephalitis/encephalopathy and gastroenteritis. This study enrolled eight patients with RVA encephalitis/encephalopathy and 10 with RVA gastroenteritis who were treated between February 2013 and July 2014. Viral RNAs were extracted from patients' stool, and whole-genome sequencing analysis was carried out to identify the specific gene mutations in RVA obtained from patients with severe neurological complications. Among the eight encephalitis/encephalopathy cases, six strains were DS-1-like G1P[8] and the remaining two were Wa-like G1P[8] (G1-P[8]-I1-R1-C1-M1-A1-N1-T1-E1-H1). Meanwhile, eight of the 10 viruses detected in rotavirus gastroenteritis patients were DS-1-like G1P[8], and the remaining two were Wa-like G1P[8]. These strains were further characterized by conducting phylogenetic analysis. No specific clustering was demonstrated in RVAs detected from encephalitis/encephalopathy patients. Although the DS-1-like G1P[8] strain was predominant in both groups, no specific molecular characteristics were detected in RVAs from patients with severe central nervous system complications.  相似文献

探讨缺氧环境下,白细胞介素8(Interleukin-8,IL-8)对人骨髓间充质干细胞(Human bone marrow mesenchymal stem cells,hBMSC)增殖和自噬能力的影响以及机制。在缺氧模型下,未进行刺激的hBMSC为缺氧对照组;以100μmol/L人IL-8蛋白刺激的MSC为IL-8组;若先添加50μmol/L MK2206(Akt蛋白抑制剂)培养30 min,然后再添加100μmol/L IL-8则为Akt抑制剂组,在正常条件下培养的MSC为正常对照组。利用Ed U细胞增殖实验、TUNEL细胞凋亡实验、Western blotting或ELISA等实验分别检测各组MSC细胞Ed U标记阳性细胞的数目、细胞凋亡、自噬蛋白(LC-3)和Akt/STAT3等蛋白的表达。相对于缺氧对照组和Akt抑制剂组,IL-8明显提高hBMSC增殖和细胞自噬,并降低hBMSC的凋亡率,IL-8组hBMSC的Akt、STAT3和VEGF等蛋白表达增高。结果表明,缺氧环境下,IL-8通过Akt-STAT3通路发挥对MSC的保护作用,对保护MSC抗缺血缺氧性损伤,促进MSC在再生医学中应用具有重要意义。  相似文献
