Four types of acidophilic granular cells, in addition to B-cells, are identified in the islet organ of anadromous specimens of two subspecies of Petromyzon marinus by light and electron microscopy. Three of these acidophils (PI, PII and PIV-cells) occur in both the cranial and hepatic islets while a fourth type (PIII-cell) has only been found in the hepatic islet of some animals. The granules of the PI-cells stain with ponceau de xylidine, give a distinct tryptophan reaction and in ultrastructural examination show large, dense granules. The PII-cells contain unusual crystals and appear to be a non-secretory stage of the PI. The PIII-cells stain deep-red and acid fuchsin. They contain very large, dense granules and some lysosomes. PIV-cells stain selectively with phosphotungstic acid-hematoxylin and ultrastructurally, contain small, more or less dense granules. It appears that PI- and PIV-cells develop directly from B-cells, while the PIII-cells derive from PI-cells. despite their direct or indirect origin from B-cells, the PI-, PIII- and PIV-cells show characteristic features of functionally independent endocrine cells. Petromyzon marinus may be an ideal model for the understanding of phylogenetic and pathological interrelationships between islet and gastrointestinal hormones. It is clear that the interpretation of the islet organ of the cyclostomes, which has been generally considered a source of insulin only, requires a revaluation.  相似文献

Summary The ultrastructure and amino acid composition of the secreted silk of two species of trichopteran larvae, Pycnopsyche guttifer (Walk.) and Neophylax concinnus McL., were investigated. The spinnerets of these two animals were also examined by scanning electron microscopy. The silk consists of double-stranded, flat ribbons (1–4 wide), composed of bundles of 15–25 Å filaments. There are two components of the silk: the fiber proper and a surrounding coat thought to be a silk gum. Only the outer coat is positive to the EM PATP technique of Thiery (1967), which indicated the presence of neutral sugars. Amino acid analyses of Pycnopsyche silk show that, like other silks, two predominant amino acids are glycine and serine. Arginine, unexpectedly, is the third most abundant and there are a large number of basic and long side-chain amino acids. X-ray diffraction studies of the silk indicate that it has a less crystalline, more amorphous structure than that of other silks.Submitted to the Department of Biological Sciences of the State University of New York at Albany in partial fulfillment of the requirements for the degree of Doctor of Philosophy Acknowledgements. The uptake of various amino acids into Streptomyces hydrogenans grown in chemostatically and turbidostatically controlled steady state cultures has been investigated. A close correlation between transport capacity and the growth rates of the cells was found. As shown by kinetic analysis, the increased transport is due to elevated maximum uptake rates, the apparent Michaelis constants remaining unchanged. Analysis of the unidirectional fluxes of cycloleucine revealed that not only the influx is raised as the growth rate is increased but also the efflux. Hence, the conclusion is drawn that the growth-rate dependent modulation of transport capacity is, at least, partially due to the variation of the concentration of active transport components. The effects of colchicine induced autotetraploidy on non-anthocyanin flavonoid expression were determine for 15 cultivars of Phlox drummondii and for the naturally occuring P. drummondii ssp. mcallisteri. Collectively, the taxa express a total of nine glycoflavonoid derivatives (C, O-diglycosides or di-C-glycosides) of either apigenin or luteolin. The autotetraploids of 14 cultivars and those of the natural subspecies exhibit altered glycoflavone profiles relative to their respective diploid sources. The qualitative alternations in the cultivars may be grouped into three phenotypic categories: (1) the expression of novel glycoflavones, (2) the absence of diploid glycoflavones, and (3) the deregulation of tissue-specific glycoflavone production. Alterations in mcallisteri autotetraploids include only the latter two categories. Each of the novel compounds is otherwise expressed among other diploid cultivars or in other wild P. drummondii subspecies. Quantitatively, the phenolic content of most autotetraploid flowers is significantly greater than in respective diploid flowers. However, on a dry weight basis, phenolic titre in comparable 4n and 2n floral or leaf tissues is not significantly different. Floral tissues express from 5 to 10 times the phenolic titre of leaves. The results are discussed in terms of the possible origins of novel flavonoids in natural polyploid Phlox species.  相似文献

The only concrete basis for the discontinuous and hierarchical organization of extant organisms lies in their genealogical (i. e. germ line) relationships. Individuals and populations of common descent are called sib or stirps. The patterns of variation of chromosome numbers in theCentrospermae suggest a common ancestry of centrospermous anthocyanin and betalain families. Phylogenetic divergence in the order may have originated with progenitors similar to extantMolluginaceae, Aizoaceae orPhytolaccaceae taxa with x = 9. Evolutionary radiation and advancement in several lines then seems to have been paralleled by trends for increasing chromosome base numbers through dysploidy and polyploidy, e.g. towardsCaryophyllaceae, Portulacaceae-Basellaceae, Hectorellaceae, Cactaceae, Didieraceae, Nyctaginaceae andChenopodiaceae-Amaranthaceae. 1-(2-Amino-2-deoxy-beta-D-xylofuranosyl)cytosine (13) was synthesized by three routes: (a) coupling of 2-deoxy-3,5-di-O-p-nitrobenzoyl-2-(trifluoroacetamido)-D-xylofuranosyl chloride (5) with 2,4-dimethoxypyrimidine and subsequent treatment with methanolic ammonia, (b) coupling of 5 with 4-N-acetyl-2-O,4-N-bis(trimethylsilyl)cytosine followed by treatment with methanolic ammonia, and (c) thiation of 1-[3,5-di-O-acetyl-2-deoxy-2-(trifluoroacetamido)-beta-D-xylofuranosyl]uracil (6) by treatment with phosphorus pentasulfide in pyridine followed by amination of the resulting 4-thionucleoside 12 with metanolic ammonia.
