High-density hardwood trees with large diameters have been found to damage manually operated increment borers, thus limiting their use in the tropics. Therefore, we herein report a new, low-cost gasoline-powered sampling system for high-density tropical hardwood trees with large diameters. This system provides increment cores 15 mm in diameter and up to 1.35 m in length, allowing minimally invasive sampling of tropical hardwood tree species, which, up to the present, could not be collected by conventional 5 or 10 mm increment borers. This system provides a single core sample with ample amount of wood for multidisciplinary analyses, including ring width, stable isotope and wood anatomical measurements. The borer never gets stuck inside stems, even in hollowed trees, cores will never twist during coring, and the gasoline drill gives ample flexibility in the field. It is anticipated that the dendrochronological community will find our technique very useful in the pursuit of tropical tree ring research.  相似文献

The vertical pattern of root length densities (RLD) of fine roots (<2 mm in diameter) and nitrogen (N) uptake potential were determined at different altitudes (1,900, 2,400, and 3,000 m a.s.l.) of a tropical montane forest in order to improve our knowledge about the depth distribution of nutrient uptake in this ecosystem. At higher altitudes, precipitation rate and frequency of fog were higher than at lower altitudes while mean annual air temperature decreased with increasing altitude. Soils were always very acid with significantly lower pH at a depth of 0.0–0.3 m in mineral soil at 3,000 m (2.8–2.9) than at 1,900 and 2,400 m (3.1–3.5). The vertical distribution of RLD was very similar both during the dry and the rainy season. During the dry season the percentage of root length in the organic layer increased from 51% at 1,900 m to 61% at 2,400 m and 76% at 3,000 m. At 3,000 m, RLD was markedly higher in the upper 0.05 m than in the remaining organic layer, whereas at 1,900 m and 2,400 m RLD were similar in all depths of the organic layer. In mineral soil, RLD decreased to a greater degree with increasing soil depth at the upper two study sites than at 1,900 m. The relative N uptake potential from different soil layers (RNUP) was determined by 15N enrichment of leaves after application of 15N enriched ammonium sulphate at various soil depths. RNUP closely followed fine root distribution confirming the shallower pattern of nutrient uptake at higher altitudes. RNUP was very similar for trees, shrubs and herbs, but shallower for saplings which obtained N only from the organic layer at both altitudes. Liming and fertilizing (N, P, K, Mg) of small patches in mineral soil had no significant impact on fine root growth. We conclude that the more superficial nutrient uptake ability at higher altitudes may be partly related to increased nutrient input from canopy by leaching. However, the specific constraints for root growth in the mineral soil of tropical montane forests warrant further investigations.  相似文献

The objectives of this study were to (1) determine if historical exceedences of oil and grease concentrations above the limits allowed in the National Pollution Elimination Discharge System permit for the wastewater treatment facility of Motiva Enterprises LLC Refinery, Delaware City, Delaware, could be determined from the sedimentary records of the Delaware River and, if so, (2) were the concentrations of contaminants high enough to be toxic to aquatic biota. Eighty-four surficial sediment samples, both within and outside the influence of the Refinery's discharge plume, were initially evaluated for their appropriateness for coring. Seven stations were ultimately selected for the historical core studies. Based on sediment type, radionuclide (Pb-210, Cs-137 and Be-7) geochronologies, and proximity of the cores to the Refinery, two cores were selected for more detailed polycyclic aromatic hydrocarbon (PAH) analyses. The rapid accumulation area of one core (located in the near-field of the discharge plume) had maximum total PAH (TPAH) concentration peaks at depths of 4 cm (1997; 3100 ng/g dw) and at 8.5–9 cm (1993; 3200 ng/g dw); the second core (located on the far-field periphery of the plume) had a maximum TPAH peak at 6.5 cm (1997; 3200 ng/g dw). In all cases, the maximum TPAH concentrations were below NOAA's Effects Range-Low concentration of 4022 ng/g dw for sediment biota toxicity. A chemical fingerprinting analysis of the PAHs in the two cores showed, however, that the PAHs present in the cores were predominantly pyrogenic PAHs consistent with Delaware River background PAHs. The core results are consistent with historic sediment PAH inputs in an industrial river system. PAH from the Refinery, if present, exist as a non-detectable increment to a high PAH baseline from many sources.  相似文献

Palaeolimnological studies were carried out on sediment cores from nine North African wetland lakes. The lakes represented a variety of habitats ranging from freshwater to mixohaline conditions and with and without direct connections to the sea. Sediment cores were examined for records of recent environmental change during the 20th century period. Faunal remains analysed in the sediments included those of Cladocera, Ostracoda, Chironomidae, Mollusca and, at the sites with marine connections, Foraminifera. Cyprideis torosa littoralis was the most common ostracod, occurring in sediments from the seven sites linked with the sea and also in brackish water Sidi Bou Rhaba. In acid Megene Chitane ostracods were scarce, being represented by a single species (Cypria ophtalmica). Candona neglecta completely disappeared at two sites (Sidi Bou Rhaba and Bokka) during the 20th century. Of the Cladocera, Chydorus sphaericus was common in the moderately mixohaline sites and but Heterocypris salina occurred only where marine salinities were occasionally achieved (in Zerga, Ichkeul and Korba). Microinvertebrate assemblages in the Nile Delta lake cores and to a less extent in Zerga showed a clear response indicative of more fresher conditions occurring during the latter part of the 20th century. However the freshening trend began prior to construction of the Aswan High Dam (mid 1960s).With the exception of the most saline site (Korba), microinvertebrate communiuties at all the CASSARINA sites have experienced major disturbances during the 20th century. Changes in freshwater availability associated with increased human usage of water resources is perceived as a major factor regulating the abundance and occurrence of aquatic microinvertebrate species at these sites  相似文献

Lake Victoria, the world's largest tropical freshwater lake, is an important resource, ecologically and economically. THg distribution in the northern parts of the lake are not well known, so to answer this gap, patterns in total mercury (THg) in water, soil and two dated sediment cores from northern Lake Victoria were determined. Water THg concentrations ranged from 0.7 to 5.8 ng/L, and there were no apparent differences observed between Napoleon and Winam Gulfs. Two precipitation samples had Hg concentrations of 7 and 31 ng/L. Surface soil samples collected from various agricultural sites around Jinja, Napoleon Gulf, have THg concentrations between 12.7 and 48.4 ng/g dry weight; they were correlated with organic carbon, total phosphorus and % clay. A near-shore core taken in Itome Bay in Napoleon Gulf, and an offshore core collected from the deepest part of the lake had similar THg concentrations and profiles (78 to 458 ng/g dry weight). The increase in THg concentration in the profiles of both cores began around 1960 and peaked around 1980. The similar sedimentary THg profiles and fluxes in the cores suggest that the THg sources to L. Victoria are primarily atmospheric, with some erosion inputs, and that equatorial African ecosystems are not exempt from the global increase in baseline THg concentrations.  相似文献

The controversy on how to measure fine root production of forests (P) most accurately continues. We applied four different approaches to determine annual rates of P in an old-growth temperate Fagus sylvatica–Quercus petraea stand: sequential soil coring with minimum–maximum calculation, sequential coring with compartmental flow calculation, the ingrowth core method, and a recently developed root chamber method for measuring the growth of individual fine roots in situ. The results of the four destructive approaches differed by an order of magnitude and, thus, are likely to introduce large errors in estimating P. The highest annual rates of P were obtained from the sequential coring approach with compartmental flow calculation, intermediate rates by sequential coring with minimum–maximum calculation, and low ones by both the root growth chamber and ingrowth core approaches. A carbon budget for the stand was set up based on a model of annual net carbon gain by the canopy and measurements on carbon sink strength (annual leaf, branch and stem growth). The budget implied that a maximum of 27% of the net carbon gain was available for allocation to fine root growth. When compared to the carbon budget data, the sequential coring/compartmental flow approach overestimated annual fine root production substantially; whereas the ingrowth core and root growth chamber approaches grossly underestimated P rates. With an overestimation of about 25% the sequential coring/minimum–maximum approach demonstrated the best agreement with the carbon budget data. It is concluded that the most reliable estimate of P in this temperate forest will be obtained by applying the sequential coring/minimum–maximum approach, conducted with a large number of replicate samples taken on a few dates per season, in conjunction with direct root growth observation by minirhizotrons.  相似文献

The sampler extracts uncompressed cores of 13·3 cm in diameter, up to 70 cm long, from the surface layers of peat. It has two close-fitting concentric cylindrical tubes, the outer one acting as a cutter and the inner one as a collector. As the outer tube is introduced by rotation into the peat, the cut core is collected in the inner tube which is maintained in a fixed position during the rotation phase and then pushed down stepwise. This limits friction between the peat core and the wall of the corer and prevents compression or distortion of the peat. These problems are also reduced by means of three skew cutters allowing the peat to be supported during the slicing action. Air can penetrate between the tubes to the lower end of the core, suppressing any suction effect during withdrawal. The sampler has been tested and has worked satisfactorily in many different peat types.  相似文献

Background

G-quadruplexes (G4s) are nucleic acids secondary structures formed in guanine-rich sequences. Anti-G4 antibodies represent a tool for the direct investigation of G4s in cells. Surface Plasmon Resonance (SPR) is a highly sensitive technology, suitable for assessing the affinity between biomolecules. We here aimed at improving the orientation of an anti-G4 antibody on the SPR sensor chip to optimize detection of binding antigens.

Methods

SPR was employed to characterize the anti-G4 antibody interaction with G4 and non-G4 oligonucleotides. Dextran-functionalized sensor chips were used both in covalent coupling and capturing procedures.

Results

The use of two leading molecule for orienting the antibody of interest allowed to improve its activity from completely non-functional to 65% active. The specificity of the anti-G4 antobody for G4 structures could thus be assessed with high sensitivity and reliability.

Conclusions

Optimization of the immobilization protocol for SPR biosensing, allowed us to determine the anti-G4 antibody affinity and specificity for G4 antigens with higher sensitivity with respect to other in vitro assays such as ELISA. Anti-G4 antibody specificity is a fundamental assumption for the future utilization of this kind of antibodies for monitoring G4s directly in cells.

General significance

The heterogeneous orientation of amine-coupling immobilized ligands is a general problem that often leads to partial or complete inactivation of the molecules. Here we describe a new strategy for improving ligand orientation: driving it from two sides. This principle can be virtually applied to every molecule that loses its activity or is poorly immobilized after standard coupling to the SPR chip surface.  相似文献

对杉木-火力楠混交林进行4种间伐强度试验表明,间伐6年后各林分的杉木、火力楠胸径净生长量比对照分别增加70-139、11-25%,树高增加46-90、7-10%,村积生长增加54-140、10-43%,林分蓄积量增加49-75m3·ha-1,平均冠高增加2-6、8-13%,第1活枝高下降0.5-3、35-48%.冠幅杉木下降1-2%、火力楠增加1-8%.可以认为,以间伐强度为30和40%2种方式较好,间伐后林分定型株数约保留在1600-2200株·ha-1之间为宜。  相似文献
