Summary Three strains ofSaccharomyces cerevisiae and one strain of aCandida sp. obtained from different industrial sources were screened for uptake of silver and copper. Considerable differences in metal uptake capacities were found between the different strains ofS. cerevisiae and betweenS. cerevisiae and theCandida sp. used. Copper uptake capacities ranged from 0.05 mmol g–1 dry wt to 0.184 mmol g–1 dry wt while values of 0.034 mmol Ag g–1 dry wt and 0.193 mmol Ag g–1 dry wt biomass were observed. Use of ion-selective electrodes (ISEs) enabled the detection of copper complexing agents (possibly proteins and carbohydrates) released by yeasts into the surrounding medium. In contrast, these compounds had no silver complexation abilities. Langmuir and Scatchard transformations of metal adsorption isotherms suggested differences in the mechanisms involved in metal uptake by the various yeasts. The differences between strains ofS. cerevisiae were due possibly to differences in cell wal composition. Different methods of preparation of biomass (fresh, air, oven and freeze-dried) had little effect on metal uptake in comparison with fresh biomass. Storage of fresh waste biomass at 4°C for 20 days had no effect on metal biosorption capacities. It was also observed that individual batches of waste biomass produced from different fermentation runs had consistent metal uptake capacities. The implications of the above results on the use of waste yeast biomass for treatment of metal-containing effluents are discussed.  相似文献

利用原核表达系统表达人源抑菌蛋白Reg3A,经包涵体的复性和纯化获得有体外抑菌功能的活性抑菌蛋白,并对其体外抑菌功能进行初步研究。构建Reg3A原核表达载体PET-32a-Reg3A转化补充稀缺tRNA基因的表达菌株大肠杆菌BL21-Codonplus,阳性重组子采用诱导培养基诱导5h后,采用超声破碎的方法提取包涵体蛋白,经包涵体蛋白的纯化和透析复性后通过Ni-NTA亲和层析交换柱,获得纯度达95%的蛋白质。Western blot鉴定显示在15 kD处有特异性条带。使用纯化后的蛋白进一步进行抑菌圈实验和抑菌活性实验,对获得蛋白的体外抑菌活性进行评估,从而为进一步进行Reg3A蛋白功能的评估及应用奠定基础。  相似文献

CRM197是一种白喉毒素突变体,作为载体蛋白广泛用于疫苗开发。将合成的CRM197基因片段克隆到表达载体pET25b中,转化大肠杆菌BL21(DE3),经IPTG诱导,CRM197获得高效表达,达到菌体总蛋白的20%。目的蛋白主要以包涵体形式存在,变性、复性后经DEAE阴离子交换、S-100分子筛纯化获得纯度高于95%的CRM197样品,用该蛋白样品免疫新西兰大白兔,免疫兔血清中检测到特异性抗体应答。急性毒性试验中,每只豚鼠皮下注射200μgCRM197纯化样品,未出现明显毒性反应症状,与之相比较,注射后48h内,20ng白喉毒素阳性对照组动物全部死亡。结果表明,利用本试验的表达策略,CRM197得到高效表达,并且具有良好的免疫原性和安全性,为其进一步生产及应用奠定基础。  相似文献

Integrated process concepts for enzymatic cephalexin synthesis were investigated by our group, and this article focuses on the integration of reactions and product removal during the reactions. The last step in cephalexin production is the enzymatic kinetic coupling of activated phenylglycine (phenylglycine amide or phenylglycine methyl ester) and 7-aminodeacetoxycephalosporanic acid (7-ADCA). The traditional production of 7-ADCA takes place via a chemical ring expansion step and an enzymatic hydrolysis step starting from penicillin G. However, 7-ADCA can also be produced by the enzymatic hydrolysis of adipyl-7-ADCA. In this work, this reaction was combined with the enzymatic synthesis reaction and performed simultaneously (i.e., one-pot synthesis). Furthermore, in situ product removal by adsorption and complexation were investigated as means of preventing enzymatic hydrolysis of cephalexin. We found that adipyl-7-ADCA hydrolysis and cephalexin synthesis could be performed simultaneously. The maximum yield on conversion (reaction) of the combined process was very similar to the yield of the separate processes performed under the same reaction conditions with the enzyme concentrations adjusted correctly. This implied that the number of reaction steps in the cephalexin process could be reduced significantly. The removal of cephalexin by adsorption was not specific enough to be applied in situ. The adsorbents also bound the substrates and therewith caused lower yields. Complexation with beta-naphthol proved to be an effective removal technique; however, it also showed a drawback in that the activity of the cephalexin-synthesizing enzyme was influenced negatively. Complexation with beta-naphthol rendered a 50% higher cephalexin yield and considerably less byproduct formation (reduction of 40%) as compared to cephalexin synthesis only. If adipyl-7-ADCA hydrolysis and cephalexin synthesis were performed simultaneously and in combination with complexation with beta-naphthol, higher cephalexin concentrations also were found. In conclusion, a highly integrated process (two reactions simultaneously combined with in situ product removal) was shown possible, although further optimization is necessary.  相似文献

用普通电镜和免疫酶电镜方法观察了流行性出血热病毒A9株和R22株感染的VeroE-6细胞中的病毒包含体,发现这些包含体具有以下三种基本形态:1.丝状或丝状颗粒包含体,2.松散颗粒包含体,3.致密包含体。在免疫酶法中,包含体均呈特异性酶染色阳性反应。描述和讨论了包含体与细胞及病毒发育成熟的关系。  相似文献
