Summary Different methods of line development using doubled haploids in recurrent selection are presented. They are divided into two types: recurrent selection with progeny testing and recurrent selection on the phenotype of lines. It is shown that one of the best methods of line development is based on single doubled haploid descent recurrent selection. Only one line is studied per plant of the population, and the best lines are intercrossed. Using this method it is very easy to extract lines for variety development.  相似文献

Summary Properties of three parameterizations, denoted as the C-model, D-model and Q-model, for covariances of inbred relatives under assumptions of no linkage or epistasis are explored and compared. Additive variance in an inbred population with inbreeding coefficient F, 2 AF =(1+F) 2 A where 2 A is additive variance in a panmictic population, if Q-model parameters Q xx and Q xy are both zero. Conditions sufficient for this to hold are presented in terms of gene frequencies and dominance contrasts (homozygotes vs. heterozygotes). Some other properties and potential uses of estimates of components in the models are also discussed. Estimates of components in the D-model and Q-model were calculated from a maize (Zea mays L.) study from which estimates of components in the C-model were previously published. Of particular interest were the covariance (Q xy ) of effects of alleles at complete homozygosity with inbreeding depression effects, the covariance (D 1) of additive effects at panmixia with inbreeding depression effects and the within-locus variance (D 2, alias Q xx ) of inbreeding depression effects. Estimates of Q xy , D 1, and D 2 were small and nonsignificant in most cases. For ear height in the second year of the study, D 2 appeared to be a major component. In some cases, results were obtained which had contradictory implications (negative D 2 coupled with positive Q xy or D 1, and positive D 2 coupled with negative 2 D ). A negative estimate of one or the other of 2 D or 2 A was obtained in one of the two within-year analyses for every character. Problems in getting realistic results were thought to be owing to excessive multicollinearity among the coefficients of the components in the expectations of the covariances of the kinds of relatives included in the study. Using a hard X-ray microprobe, we showed recently that in unstimulated peritoneal macrophages from C57BL/6 mice, the phagosome of pathogenic mycobacteria (Mycobacterium tuberculosis and Mycobacterium avium) can accumulate iron. We expanded our studies to the M. avium infection of peritoneal macrophages of Balb/c mice that show a similar degree of M. tuberculosis and M. avium-related chronic disease, but a higher susceptibility towards other intracellular pathogens such as Listeria monocytogenes, Leishmania major, or Brucella abortus as compared to C57BL/6 mice. Similar to C57BL/6 macrophages, the iron concentration in Balb/c macrophages increased significantly after 24 h of infection. A significant increase of the chlorine and potassium concentrations was observed in the Balb/c phagosomes between 1 and 24 h, in contrast with macrophages from C57BL/6 mice. The absolute elemental concentrations of calcium and zinc were higher in the mycobacterial phagosomes of Balb/c mice. We hypothesize that a potassium channel is abundant in the phagosome in macrophages that may be related to microbiocidal killing, similar to the requirement of potassium channels for microbiocidal function in neutrophils.  相似文献

The 5-hydroxytryptamine (5-HT4) receptor may be implicated in depression and is a new potential target for antidepressant treatment. We have investigated the brain 5-HT4 receptor [3H]SB207145 binding in the Flinders Sensitive Line rat depression model by quantitative receptor autoradiography, and related this to 5-HT transporter ( S )-[ N -methyl-3H]citalopram binding. We also determined the regulation of 5-HT4 receptor binding by 1, 14, and 21 days of paroxetine administration and subchronic 5-HT depletion, and compared this with changes in 5-HT2A receptor [3H]MDL100907 binding. In the Flinders Sensitive Line, the 5-HT4 receptor and 5-HT transporter binding were decreased in the dorsal and ventral hippocampus, and the changes in binding were directly correlated within the dorsal hippocampus. Chronic but not acute paroxetine administration caused a 16–47% down-regulation of 5-HT4 receptor binding in all regions evaluated including the basal ganglia and hippocampus, while 5-HT depletion increased the 5-HT4 receptor binding in the dorsal hippocampus, hypothalamus, and lateral globus pallidus. In comparison, the 5-HT2A receptor binding was decreased in the frontal and cingulate cortices after chronic paroxetine administration, and markedly reduced in several regions after 5-HT depletion. Thus, the 5-HT4 receptor binding was decreased in the Flinders Sensitive Line depression model and in response to chronic paroxetine administration.  相似文献

Chilling stress is an important constraint of global production of maize. This study was undertaken to compare the chilling responses of different maize seedling tissues and to analyze changes in polyamines as a result of chilling stress. Reponses to chilling were characterized in two maize (Zea mays L.) inbred lines, ‘HuangC’ and ‘Mo17’, that putatively differ in chilling sensitivity. Seedlings were exposed to low temperature (5°C) and chilling injury was estimated by electrical conductivity (EC), malonaldehyde (MDA) concentration, and by changes in putrescine (Put), spermidine (Spd) and spermine (Spm) concentrations in root, mesocotyl, and coleoptile tissues. Membrane permeability (as measured by EC), MDA concentrations and Put concentrations in the three tissue of maize seedlings increased after chilling stress, except for the Put concentration in roots. Spd and Spm concentrations in the three tissues of seedlings decreased after chilling stress. The EC for cold stressed tissues were lower in HuangC than Mo17. Also, the EC of coleoptile tissues were lower than for mesocotyl in both inbred lines. We suggest that mesocotyl tissue can be used to evaluate cold tolerance in maize. Stepwise regression analyses showed that chilling injury in roots was generally correlated with Spd concentration while in the mesocotyl injury was mainly correlated with Put and Spd concentrations. Spermidine and Spm concentrations in the coleoptile were correlated with chilling injury. Characteristics changes of polyamines in chill-tolerant maize seedling combined with regression analysis are a reliable method for evaluating chill tolerance in maize lines.  相似文献

Almost 10,000 single nucleotide polymorphisms (SNPs) had been identified in the RT1 complex, the major histocompatibility complex of the rat, but less than ∼0.5% have been characterized. In the context of the incomplete characterization of most SNPs, simple sequence length polymorphism (SSLP) marker development is still valuable for understanding the involvement of genes in the RT1 in controlling disease susceptibility, since SSLPs are user-friendly and cost-effective genetic markers in rat genome analysis. In this study, we developed a set of 67 SSLP markers, including 57 novel markers, to cover the entire RT1 complex and then created genetic profiles across 67 rat strains. These markers are located almost every 50 kb in the RT1 complex and show comparable polymorphism; the average number of alleles was 8.04 ± 3.44 and the average polymorphic rate was 71 ± 23%. Interestingly, markers failing to amplify polymerase chain reaction products were highly observed in all strains except for BN/SsNHsd, which suggests the existence of highly variable genomic sequences or genomic rearrangements in the RT1 region across rat strains. Based on the phylogenic tree and individual genotyping data, we identified 28 SSLP marker haplotypes in the RT1 region that roughly consisted of three genomic regions. These findings provided new insight into the genomic organization of the RT1 complex and we recognized the need of additional RT1 genome sequences in different strains. Owing to the accuracy and ease of determination, PCR-based SSLP genotyping could replace serological typing in genetic analyses and characterization of rat major histocompatibility. There is an important role of understanding the genetic diversity among and within inbred lines at the molecular level for maize improvement in different breeding programs. The present study was devoted to estimate the level of genetic diversity among the inbred lines of maize using the simple sequence repeat analysis (SSR). The application of six different SSR markers successfully provided the information on similarity or diversity as well as the heterozygosity of the allelic loci for all the eight inbred line of maize.  相似文献

In this study, we determined whether p53 expression affected the sensitivity of non–small cell lung cancer (NSCLC) and colon cancer cells to bleomycin (BLM). Two human NSCLC cell lines (A549 expressing wild‐type p53 and p53‐null H1299) and two colon cancer cell lines (HCT116 p53+/+ and its p53 deficient subline HCT116 p53?/?) were subjected to treatment with BLM. Cells were treated with various concentrations of BLM, and cellular viability was assessed by formazan assay. To investigate the role of p53 in BLM sensitivity, we transduced cells with adenovirus with wild‐type p53, dominant‐negative p53, and GFP control, and analyzed the effect on cellular viability. Cells expressing wild‐type p53 were more sensitive to BLM than p53‐null cells in both NSCLC and colon cancer cells. Sensitivity to BLM of the cells with wild‐type p53 was reduced by overexpression of dominant‐negative p53, while BLM sensitivity of p53‐null cells was increased by wild‐type p53 in both NSCLC cells and colon cancer cells. In conclusion, our data show that p53 sensitizes all four cancer cells lines tested to BLM toxicity and overexpression of p53 confers sensitivity to the cytotoxic activity of the anticancer agent in p53‐null cells. © 2010 Wiley Periodicals, Inc. Some breeders select inbreds from many F2 or backcross breeding populations, each with relatively few progenies. Other breeders select inbreds from only a few breeding populations, each with many progenies. My objectives were to: (1) determine the relative importance of parental selection, number of breeding populations, and size of each population, and (2) find optimum combinations between number and size of breeding populations. I assumed that a breeder has resources to test a total of 2,000 recombinant inbreds for a quantitative trait that was controlled by 100 additive loci and had a heritability of 0.20, 0.60, or 1.0. The parental inbreds had an inherent pedigree structure due to advanced cycle breeding. The parental inbreds were ranked according to their mean performance, and breeding populations were made among all parents, the top 25% of parents, and the top 10% of parents. I found that the issue of number versus size of breeding populations was only secondary compared with the ability to identify, prior to making the crosses, the breeding populations with the highest mean performance. For a given level of effectiveness of parental selection, the selection response was largest when the maximum number of breeding populations was used. The effect of the number of breeding populations was minor, however, when selection was practiced among the parents or when heritability was less than 1.0. The results suggested that, in practice, large selection responses could be obtained with a wide range of combinations between number and size of breeding populations.Communicated by H.C. Cholesterol may affect the activity of microbial toxins in a direct, specific way, or it may exert indirect effects because of its role in membrane fluidity, membrane line tension, and in the stabilization of rafts in the cytoplasmic membrane. The thiol-activated toxins of gram-positive bacteria, and the cytolysin of Vibrio cholerae are presented as examples of specific toxin-cholesterol interaction. Several mechanisms of indirect effects of cholesterol are discussed using examples such as Staphylococcus aureus alpha-hemolysin, aerolysin, and diphtheria toxin.  相似文献
