目的:探讨索拉非尼(Sorafenib)和阿霉素(adriamycin)联合用药对肝癌细胞株nepG2的作用及可能的机制。方法:以不同浓度索拉非尼和不同浓度阿霉素分别组成单药组和索拉非尼+阿霉素联合用药组作用于HepG2细胞,MTT法检测增殖抑制率、流式细胞仪分析细胞周期和凋亡率。结果:索拉非尼、阿霉素单药与联用均能抑制HepG2细胞增殖,呈剂量依赖效应,两药联用有协同效应(P〈0.01)。索拉非尼、阿霉素单药与联用均能诱导HepG2细胞凋亡,并以联合组更为明显,与对照组比较有显著的统计学意义(P〈0.01)。索拉非尼及阿霉素单药作用均可使细胞周期阻滞于G0-G1期,联合用药组G0/Gl期细胞比率低于索拉非尼及阿霉素单药组,S期细胞比率高于单药组;阿霉素能抑制HepG2细胞Survivin mRNA表达诱导细胞的凋亡。结论:索拉非尼与阿霉素联合作用于人肝癌HepG2细胞具有协同作用,其机制可能是通过多途径共同抑制HepG2细胞增殖及诱导细胞凋亡。 相似文献

目的建立红鲫C1HD近交系的RAPD标记。方法从80条随机引物中筛选出20条扩增效果和多态性较好的引物,对8尾红鲫C1HD近交系和8尾普通红鲫基因组DNA进行RAPD扩增。结果S333引物扩增出一条特异性条带,大小约为2.1 kb。结论S333引物扩增出的特异性条带可以作为区分普通红鲫与红鲫C1HD近交系的分子遗传标记。 相似文献

Six experiments (including pretreatment, embryonic callus induction media, preculture conditions, embryo induction media,
embryo germination media, and genotypic effects) were conducted to develop an efficient cucumber (Cucumis sativus L., 2n = 2x = 14) anther culture protocol. Pretreatment and embryo induction were key factors for successful anther culture. Suitable
temperature stress depended on the ecotype, i.e., cucumbers from cold areas responded well to cold shock whereas those from
temperate areas responded well to heat treatment. The best medium for embryonic callus induction was MS medium supplemented
with 4.44 μM BA, 2.26 μM 2, 4-D, 4.64 μM KIN, 3% sucrose and 0.8% agar. For embryo induction, MS medium supplemented with
0.54 μM NAA, 13.32 μM BA, 3% sucrose and 0.8% agar was optimal, and for embryo germination MS medium containing 2.22 μM BA,
6% sucrose and 1.2% agar was best. Using this protocol, we produced callus from 16 genotypes and regenerated plants from three
of 20 evaluated. Three embryos per anther and 42 DH per 45 anthers (93% success) were obtained for cv. Ningjia No. 1, which

The compositional differences between domains in phase-separated membranes are associated with differences in bilayer thickness and moduli. The resulting packing deformation at the phase boundary gives rise to a line tension, the one dimensional equivalent of surface tension. In this paper we calculate the line tension between a large membrane domain and a continuous phase as a function of the thickness mismatch and the contact angle between the phases. We find that the packing-induced line tension is sensitive to the contact angle, reaching a minimum at a specific value. The difference in the line tension between a flat domain (that is within the plane of the continuous phase) and a domain at the optimal contact angle may be of order 40%. This could explain why previous calculations of the thickness mismatch based line tension tend to yield values that are higher than those measured experimentally. 相似文献

We observed tissue sections of single hybrid of maize 328 and its parents during early embryogeny, determining ATP ase activities and observing plant features. The results obtained show that maize embryogeny can be divided into three periods, that is, 1) The period of 3–7 days after pollination is known as an original embryonic period. 2) The period of 9–20 days after pollination is known as the period of undergoing rapid differentiation of all the tissue organs. 3) The period of 25–45 days is known as the period of further differentiation, perfection and elongation of organal system. The size of volume of hybrid embryo is between its parents during the second period. That of the hybrid embryo during other two periods is larger than that of its parents. Hybrid embryo always shows that the number of its cells is more, its cells are closer together, the colour of the stained cells is darker, the cells differentiate clearly, the time that embryonic organs differentiates lasts longer. The results of determinig ATP ase activities it may be seen that during 1–2 days after pollination ATP ase activities in hybrid ovaries are obviously higher than in its parents’ ovaries. On the third day after pollination ATP ase activities in hybrid ovaries once go down. After that ATP ase activities go up again. During 15–45 days ATP ase activities in hybrid embryo (unincloding endosperm) are all higher than those in its parents. Hybrid 328 has advantage over its parents in yielding capacity, plant height, spike length, number of seeds per spike, 1000 seed weight. At Palmyra Atoll, the environmental DNA (eDNA) signal on tidal sand flats was associated with fish biomass density and captured 98%–100% of the expected species diversity there. Although eDNA spilled over across habitats, species associated with reef habitat contributed more eDNA to reef sites than to sand-flat sites, and species associated with sand-flat habitat contributed more eDNA to sand-flat sites than to reef sites. Tides did not disrupt the sand-flat habitat signal. At least 25 samples give a coverage >97.5% at this diverse, tropical, marine system. 相似文献

Peanut, the only cash crop of rainfed areas of Pakistan, is facing immense challenges due to global warming. Climatic factors particularly the temperature fluctuations and rain pattern shift significantly impact the production and yield of peanut and unavailability of resilient varieties exacerbate this impact. To deal with the cropping pattern change and yield losses, due to climate vagaries, a study was conducted to develop early maturing hybrids using line into tester mating design. The F1 hybrids from the parental lines were produced in the year 2018 using Line × Tester mating design and then grown in the field in the year 2019 for further evaluation. The hybrids were evaluated based on the early maturity and yield-related attributes in comparison with the parental lines. Based on the general combining ability estimate, line V-3 (Golden), was found as best parent with highly significant values for plant height, days to peg formation, days to maturity, number of pegs per plant, number of pods per plants, number of seeds per plant, 100 pod weight 100 seed weight. Similarly, tester V-7 (PI 635006 01 SD) showed highly significant results of GCA for days to germination, day to 50% flowering, plant height, days to peg formation, days to maturity, number of pegs per plant, number of pods per plants, number of seeds per plant, 100 kernel weight, shelling percentage. All the combinations were evaluated for specific combining ability and significant results were observed for V-3 × V-4 (Golden × PI 619175 01 SD) and V-1 × V-6 (BARI-2000 × PI 564846 01 SD) by developing or maturity and yield-related attributes. The hybrid combinations V-3 × V-5 (Golden × PI 635006 01 SD) followed by V-3 × V-6 showed highly significant results for mid parent heterosis and better parent heterosis for days to 50% flowering, plant height, days to peg formation, number of pegs, days to maturity, number of mature seeds per plant, shelling ratio, 100 pod weight and 100 kernel weight. Interest in surveys for monitoring plant abundance is increasing, due in part to the need to quantify the rate of loss of biodiversity. Line transect sampling offers an efficient way to monitor many species. However, the method does not work well in some circumstances, for example on small survey plots, when the plant species has a strongly aggregated distribution, or when plants that are on the line are not easily detected. We develop a crossed design, together with methods that exploit the additional information from such a design, to address these problems.
