非特异性脂质转移蛋白突变体的构建及在两种硫氧还蛋白表达载体中的表达比较 总被引:4,自引:0,他引:4
对水稻非特异性脂质转移蛋白(Nospecific lipid transfer protein,nsLTP) LTP110中结构重要的5个氨基酸位点进行了定点突变,测序结果证实了突变体构建成功。在尝试了多种大肠杆菌表达系统进行表达之后,发现硫氧还蛋白融合表达载体适合于LTP110野生型及突变体的表达。将编码野生型LTP110及突变体Y17A,P72L,R46A,D45A,C50A蛋白的cDNA顺序克隆进两种硫氧还蛋白表达载体并对其表达情况进行了比较:pTrxFus载体可以在宿主菌GI724中以较低水平表达野生型LTP110及突变体Y17A,P72L,R46A融合蛋白,但不能表达D45A和C50A融合蛋白;pET32a(+)载体可以在宿主菌BL21 (DE3) trxB-中以可溶蛋白的形式表达野生型及所有突变型融合蛋白,且表达量比在pTrxFus载体/GI724突主菌中表达量高。对pET32a(+)载体中表达的LTP110融合蛋白进行了纯化,并利用带有荧光标记的脂肪酸分子对其测活,结果表明表达的野生型LTP110分子具有结合脂质的活性。 相似文献

费氏中华根瘤菌 (Sinorhizobiumfredii) 0 4 2BS分离自新疆的苜蓿根瘤 ,通过交叉结瘤试验 ,发现它既可在苜蓿上又可在大豆上结瘤固氮。 1 6SrDNAPCR RFLP分析表明 ,0 4 2BS与费氏中华根瘤菌模式菌株USDA2 0 5的 4种限制性酶切图谱完全一致。其G +Cmol%为 60 0 ,与费氏中华根瘤菌USDA2 0 5和USDA1 91的DNA同源性分别为 84 9%和89 6% ,表明 0 4 2BS属于费氏中华根瘤菌。应用绿色荧光蛋白基因标记 0 4 2BS ,得到重组菌株 0 4 2BSG。将其接种保定苜蓿和北引 1号大豆 ,并重新分离出根瘤菌 ,利用激光共聚焦荧光显微镜检测到标记基因的表达 ,从而确证了 0 4 2BS能在苜蓿和大豆上结瘤。而且 ,0 4 2BS对不同苜蓿品种的结瘤能力不同 相似文献

Iron deficiency induced decrease in the rate of whole electron transport chain in chloroplasts of pea (Pisum sativum L.). Such reduction was mainly due to the loss of photosystem (PS) 2 activity. The same result was obtained when the ratio of variable to maximum chlorophyll fluorescence (Fv/Fm) was evaluated. The loss in PS 2 activity was primarily due to a loss of 33, 23 and 17 kDa polypeptides. In contrast, iron deficiency induced the synthesis of 28 and 29 kDa polypeptides. 相似文献

The Effect of In Vitro Culture Conditions on the Pattern of Photoinhibition during Acclimation of Gardenia Plantlets to Ex Vitro Conditions 总被引:1,自引:0,他引:1
We tested the effect of growing conditions during micropropagation on the fast kinetics of chlorophyll (Chl) fluorescence of Gardenia jasminoides Ellis plantlets during a 4-week acclimation to ex vitro. We studied whether photoautotrophic growing in vitro produced plantlets with less photoinhibition impairment during acclimation. Of the growing conditions stimulating photoautotrophy in vitro, only loose tube caps had a positive effect, whereas low sucrose or sucrose-free content in the medium and high PPFD showed a negative effect. Thus, plantlets cultured with 3 % (m/v) of sucrose were subsequently less photoinhibited throughout acclimation than those cultured with low sucrose (0.5 %) or sucrose-free media. Moreover, at the end of acclimation the former plantlets showed Fv/Fm and Fv/F0 ratios typical of unstressed ex vitro plants as well as a higher Chl content and ratio of Chls to carotenoids. Plantlets cultured at a photosynthetic photon fluence density (PPFD) of 50 µmol m–2 s–1 also showed a better performance at the end of acclimation than those cultured at a higher (110 µmol m–2 s–1) PPFD. Thus except in the case of loose-tube closure, gardenia plantlets cultured in vitro under conventional sucrose concentration and PPFD are the least photoinhibited during acclimation. Photosynthesis in iron-deficient soybean and maize leaves decreased drastically. The quantum yield of photosystem 2 (PS2)
electron transport (ΦPS2), the efficiency of excitation energy capture by open PS2 reaction centres (Fv′/Fm′), and photochemical quenching coefficient (qP) under high irradiance were lowered significantly by iron deficiency, but non-photochemical quenching (NPQ) increased markedly.
The analysis of the polyphasic rise of fluorescence transient showed that iron depletion induced a pronounced K step both
in soybean and maize leaves. The maximal quantum yield of PS2 photochemistry (Φpo) decreased only slightly, however, the efficiency with which a trapped exciton can move an electron into the electron transport
chain further than QA (Ψ0) and the quantum yield of electron transport beyond QA (ΨEo) in iron deficient leaves decreased more significantly compared with that in control. Thus not only the donor side but also
the acceptor of PS2 was probably damaged in iron deficient soybean and maize leaves.

沙冬青(Ammopiptanthus mongolicus (Maxim.) Cheng f.)是生长在沙漠及干旱荒漠地区的常绿灌木.在夏季,其叶片经常遭受中午强光(超过1 500 μmol*m-2*s-1) 胁迫,出现明显的光抑制现象.我们利用便携式光合测定系统(CIRAS-1)和脉冲调制荧光仪(MFMS-2)测定了自然形成的干旱胁迫条件下沙冬青光合和荧光参数的日变化,主要探讨了干旱胁迫对沙冬青叶片防御强光破坏机制的影响.结果表明,正常水分和干旱胁迫下,沙冬青叶片的净光合速率(Pn)、 PSⅡ最大光化学效率(Fv/Fm)和PSⅡ非环式电子传递效率(ΦPSⅡ)在中午都明显降低;相对正常水分条件而言,干旱胁迫下初始荧光(Fo)先下降后上升,荧光的非光化学淬灭(NPQ)上升较快并在一定水平上维持不变.由此推断晴天中午沙冬青叶片在正常水分条件下主要采取依赖叶黄素循环的热耗散机制;而在干旱胁迫条件下主要采取了依赖叶黄素循环的热耗散和PSⅡ反应中心可逆失活两种保护机制. 相似文献

利用电感耦合等离子发射光谱-质谱联用法(ICP-MS)、原子荧光法(AFS)和原子吸收法(AAS)对不同品种灵芝子实体中的生物必需微量元素、有毒的微量元素及重金属元素的含量进行了测定,并对其营养性和安全性进行了分析。与砷、镉、汞相比,在所有品种的灵芝中重金属铅的含量相对较高,但是在对全国各地收集的灵芝及其培养基分析,灵芝对铅没有生物富集的作用。另外,灵芝粗多糖中各种元素的含量均明显高于提取用的灵芝子实体,其中铅和砷含量高于国家标准,其安全性应该受到重视。 相似文献

Tea catechins and other flavonoids have been shown to potentially protect against chronic cardiovascular diseases such as coronary heart disease and atherosclerosis. In this study, 6-month-old female Sprague-Dawley rats were fed green tea extract (50 mg/100 ml in drinking water) up to the age of 22 months, and the age-associated changes in Maillard-type fluorescence and carbonyl groups in the aortic and skin collagen were compared with those occurring in the water-fed control animals. Collagen-linked Maillard-type fluorescence was found to increase in both the aortic and skin tissues as animals aged. The age-associated increase in the fluorescence in the aortic collagen was remarkably inhibited by the green tea extract treatment, while that occurring in the skin collagen was not significantly inhibited by the treatment. The collagen carbonyl content also increased in both the aortic and skin tissues as animals aged. In contrast with the case of Maillard-type fluorescence, however, the age-associated increase in the carbonyl content was not inhibited by the green tea extract treatment either in the aortic or skin collagen. These results suggest that the inhibition of AGE formation in collagen is an important mechanism for the protective effects of tea catechins against cardiovascular diseases. 相似文献

In mammalian cells, most membrane proteins are inserted cotranslationally into the ER membrane at sites termed translocons. Although each translocon forms an aqueous pore, the permeability barrier of the membrane is maintained during integration, even when the otherwise tight ribosome-translocon seal is opened to allow the cytoplasmic domain of a nascent protein to enter the cytosol. To identify the mechanism by which membrane integrity is preserved, nascent chain exposure to each side of the membrane was determined at different stages of integration by collisional quenching of a fluorescent probe in the nascent chain. Comparing integration intermediates prepared with intact, empty, or BiP-loaded microsomes revealed that the lumenal end of the translocon pore is closed by BiP in an ATP-dependent process before the opening of the cytoplasmic ribosome-translocon seal during integration. This BiP function is distinct from its previously identified role in closing ribosome-free, empty translocons because of the presence of the ribosome at the translocon and the nascent membrane protein that extends through the translocon pore and into the lumen during integration. Therefore, BiP is a key component in a sophisticated mechanism that selectively closes the lumenal end of some, but not all, translocons occupied by a nascent chain. By using collisional quenchers of different sizes, the large internal diameter of the ribosome-bound aqueous translocon pore was found to contract when BiP was required to seal the pore during integration. Therefore, closure of the pore involves substantial conformational changes in the translocon that are coupled to a complex sequence of structural rearrangements on both sides of the ER membrane involving the ribosome and BiP. 相似文献
