小麦光温敏雄性不育相关基因的DDRT-PCR分析及功能预测   总被引:10,自引:0,他引:10  
 以小麦光温敏雄性不育系BS210为试材,分别在北京顺义(可育环境)和安徽阜阳(不育环境)两种诱导育性表现的生态环境下种植,并于雌雄蕊分化期、药隔形成期、花粉母细胞形成期、四分体期和单核期5个时期提取小穗组织的RNA,利用DDRT-PCR方法分离不育系在光、温因子诱导下的差异表达mRNA,并通过反向Northern 进行验证.对获得的20条差异表达的EST进行了测序和BLAST分析,得到了4个候选基因的片段,对其进行5′Race 扩增、序列分析及功能预测.结果表明:它们分别与水稻的DNA修复重组蛋白基因rad50、小麦穗部表达的LRR重复序列型类受体激酶基因、玉米叶片坏死斑点L1s1基因的序列相似性分别为89%、89%和88%,另外还筛选到1个未知功能的新基因片段,它和rad50分别在可育和不育环境下表达的mRNA具有相同的5′端编码区,但3′端非编码区  相似文献

Allium fistulosum possesses a number of traits which would be desirable in A. cepa. Thus far, no commercial A. cepa cultivars have been released which harbor Allium fistulosum traits. F1BC3 populations were generated for this study by backcrossing A. cepa to A. cepa×A. fistulosum hybrids. The F1BC3 plants were evaluated for plant morphology, floral characters, male- sterile cytoplasm, soluble solids and pungency, and isozymes. Overall growth habit and floral characters of the F1BC3 plants were much like A. cepa. We report here the recovery of recombinant, bulbing, and fertile A. cepa-type onions that exhibit A. fistulosum isozyme alleles and morphological markers. Recombination between A. cepa and A. fistulosum genomes was achieved using the introgression strategy of backcrossing A. fistulosum into A. cepa, thereby ameliorating the nuclear cytoplasmic barriers that occurred in previous less successful introgression attempts when plants were not in A. cepa cytoplasm. We believe this report to be the first demonstration of onion introgressants that are like A. cepa in appearance, are male- and female-fertile, and possess A. fistulosum genes. 本研究采用种子醇溶蛋白酸性聚丙烯酰胺凝胶电泳(APAGE)和染色体C分带技术对重庆温光型雄性不育小麦的染色体组成进行分析,结果表明:不育系染色体1B短臂(1BS)已被黑麦1R短臂(1RS)取代,具有1RS/1BL易位染色体及普通小麦细胞质。 Abstract:Seed gliadin acid-polyacrlamide-gel-electrophoresis(APAGE) and chromosome C-banding techniques were used to identify chromosome constitution of Chongqing thermo-photo sensitive male sterile wheat.Results showed that 1BS of the male sterile lines was substituted by 1RS of rye.They were 1B/1R wheat with Triticum aestivum cytoplasm.  相似文献

利用RAPD和ISSR分子标记对K型小麦(Triticum aestivum L.)雄性不育恢复系LK783的主效恢复基因进行了标记定位.以K冀5418A//911289/LK783三交F1分离群体的极端不育株和极端可育株分别建立保持池和恢复池,利用418个RAPD和33个ISSR引物对两池间的多态性进行了研究.分析表明RAPD引物OPK18和ISSR引物UBC-845在两池间扩增出稳定的多态性差异,在分离群体上的验证结果表明LK783的育性恢复基因与两个引物的扩增位点有连锁关系,在染色体上位于两个引物的扩增位点之间,与OPK18450的遗传距离为(15.07±6.28)cM (centiMorgan),与UBC-845800的遗传距离为(8.20±4.85)cM.这两个引物可应用于对育性恢复基因的标记辅助选择.最后,利用中国春缺体-四体系和双端体系进一步将UBC-845800定位于1BS, 表明LK783的育性恢复基因也位于1BS.  相似文献

In the twenty-two years since the Awash baboon hybrid zone was first described, about 25-30 hamadryas or hamadryas-like hybrid males have taken up residence in anubis baboon groups. Most enter as mature adults, though some may immigrate as juvenile followers. Long-term tracking of known immigrants indicates that they may reside for four years or more in their adopted troop, during which time they establish a series of comparatively short-term harems, and probably sire hybrid offspring. "Abductions" of anubis females to hamadryas troops, as postulated by early observers, were not seen, though their occurrence cannot be ruled out. Younger, more recent immigrants tend to be "purer" hamadryas in phenotype, perhaps reflecting a more distant origin, or a change in the composition of neighboring hamadryas populations. Coexisting hamadryas and anubis show no signs of reproductive isolation by behavioral barriers, and evidently belong to a single "recognition species."  相似文献

Summary A linear mitochondrial plasmid reported to be associated with cytoplasmic male sterility in the genus Brassica was analyzed. A protein was found to be associated with the 5 ends of the plasmid. The entire plasmid was cloned by the homopolymer tailing technique via free hydroxyl groups present at its 3 ends. DNA sequence analysis of the cloned plasmid revealed a perfect terminal inverted repeat of 325 base pairs. Southern hybridization and restriction enzyme mapping analysis confirmed colinearity of the native plasmid and the clone, which showed significant homology with organelle DNA but not with nuclear DNA. Under high-stringency hybridization conditions, an internal 4.6 kb fragment of the 11.5 kb plasmid hybridized to the main mitochondrial genome in several species. Although the hybridization signal was weaker, the chloroplast genome also showed homology to the mitochondrial plasmid. The plasmid was undetectable at a molar ratio of less than 1/10 000 of the main mitochondrial genome in some lines of Brassica and Raphanus that contain the Ogura male sterile cytoplasm (cms). The absence of the plasmid in these sterile lines demonstrates that the plasmid is not required for the expression and maternal inheritance of male sterility.  相似文献

Chalcone synthase (CHS) is a key enzyme and producing flavonoid derivatives as well play a vital roles in sustaining plant growth and development. However, the systematic and comprehensive analysis of CHS genes in island cotton (G. barbadense) has not been reported yet especially response to cytoplasmic male sterility (CMS). To fill this knowledge gap, a genome-wide investigation of CHS genes were studied in island cotton. A total of 20 GbCHS genes were identified and grouped into five GbCHSs. The gene structure analysis revealed that most of GbCHS genes consisted of two exons and one intron, and 20 motifs were identified. Twenty five pairs duplicated events (12 GbCHS genes) were identified including 23 segmental duplication pairs and two tandem duplication events, representing that GbCHS gene family amplification mainly owned to segmental duplication events and evolving slowly. Gene expression analysis exhibited that the GbCHS family genes presented a diversity expression patterns in various organs of cotton. Coupled with functional predictions and gene expression, the abnormal expression of GbCHS06, 10, 16 and 19 might be associated with pollen abortion of CMS line in island cotton. Conclusively, GbCHS genes exhibited diversity and conservation in many aspects, which will help to better understand functional studies and a reference for CHS research in island cotton and other plants.  相似文献
