An axial column (3×2.6 cm) and a radial flow column (3.5×5 cm) packed with DEAE Sepharose Fast Flow media had been evaluated for the separation of human fibrinogen. Nitschmann fraction I dissolved in buffered saline (0.015 M NaCl buffered with 0.06 M Tris/HCl to pH 7.5) was the starting material. Under radial flow conditions, sample flow up to 15 ml min–1 (i.e., 18 bed volumes h–1) was achieved. The operating pressures were below 0.2 MPa, even though the elution velocity was 30 ml min–1 (i.e., 36 bed volumes h–1).  相似文献

Summary The clay fraction separated from an alluvial Egyptian soil and montmorillonite clay mineral were equilibrated with CaCl2 or NaCl solution then treated with humic acid isolated from composted clover straw to obtain different clay systems: Ca-clay, Ca-clay-HA, Na-clay, Na-clay-HA, Ca-mont and Ca-mont-HA. These clays as well as seven soil samples were reacted with different amounts of labelled65ZnCl2,65ZnEDTA and65ZnDTPA. The effectiveness of these Zn-sources for maintaining soluble Zn2+ ions in the equilibrium solution was the greatest for ZnDTPA and the lowest for ZnCl2. Ca-clay provided greater Zn sorption capacity than Na-clay, and complexing the clay with humic acid depressed its capacity for Zn sorption. At the pH of the clay-systems (pH=6.5), the possibilities of Zn(OH)2 formation were reduced especially in the presence of Zn-chelates. Reactions of65ZnE DTA and65ZnDTPA with the seven soils produced higher levels of soluble Zn2+ ions in the equilibrium solution rather than65ZnCl2 meanwhile ZnDTPA was more effective than ZnEDTA. The calculated Zn(OH)2 ion product in the solution of ZnCl2-soil systems indicated the precipitation of Zn as Zn(OH)2. However, this was not valid in the Zn-chelates-soil systems. The results also revealed the role of soil carbonate, organic matter and soil texture as soil variables affecting Zn sorption by natural soils.  相似文献

The subcellular distribution of aldehyde dehydrogenase activity was determined in human liver biopsies by analytical sucrose density-gradient centrifugation. There was bimodal distribution of activity corresponding to mitochondrial and cytosolic localizations. At pH 9.6 cytosolic aldehyde dehydrogenase had a lower apparent Kappm for NAD (0.03 mmol l-1), than the mitochondrial enzyme (Kappm NAD = 1.1 mmol l-1). Also, the pH optimum for cytosolic aldehyde dehydrogenase activity (pH 7.5) was lower than that for the mitochondrial enzyme activity (pH 9.0), and the cytosolic enzyme activity was more sensitive to inhibition by disulfiram in vitro. Disulfiram (40 mumol l-1) caused a 70% reduction in cytosolic aldehyde dehydrogenase activity, but only a 30% reduction in mitochondrial enzyme activity after 10 min incubation. The liver cytosol may therefore be the major site of acetaldehyde oxidation in vivo in man.  相似文献

The removal of humic substances from aqueous solution at pH=4.8 and 6.0 and 25 °C by pure culture of Aspergillus niger has been examined. The removal of humic substances from aqueous phase was monitored by following the decrease in absorbance at 370 nm. COD and fungal growth were also measured. The initial concentration of humic and fulvic acids, pH and diameter of fungal pellets were found to be the most important parameters. The results suggested that decolorization of batch culture was a consequence of several processes. In the first 5 hours for pellets with diameter 1-3 mm and 10 hours for pellets with diameter 2-5 mm adsorption of humic substances was dominant. Afterwards, desorption of slightly bound humic substances and their degradation, as well as degradation of remaining humic substances in the solution, continued simultaneously through the period of 10 days.  相似文献

The high carcinogenic potency of polycyclic aromatic compounds often results in the dermal pathway indicating significant risk to human health at sites with contaminated soils, resulting in the establishment of conservative, risk-based remediation goals. The sorptive properties of soil sequester chemical contaminants, making them less available for uptake by receptors. Recent studies of desorption from soil indicate that PAHs follow a nonlinear desorption pattern that can be estimated by two phases: a rapid, followed by a slow, desorbing fraction. In this work, we adapt a fugacity-based model to evaluate the availability of polycyclic aromatic hydrocarbons (PAHs) from soil to human skin. Incorporating two-site desorption kinetics into the fugacity model renders a less available fraction of chemical in soil for absorption, decreasing predicted dermal uptake. We explore the impacts to dermal bioavailability of removing the “fast-desorbing” fraction of chemical from the soil. The model predicts uptake within a factor of two when compared with experimental data on dermal uptake. Soil moisture and soil loading rates emerge as potential limiting variables; however, the model is most sensitive to the size of the fast desorbing fraction of chemical in soil  相似文献

Fumaria genus has been traditionally used for managing inflammatory and gastrointestinal disorders. The study evaluates the immunomodulatory potential of the total alkaloid fraction from Fumaria capreolata L. (AFC) in primary macrophages and the intestinal anti-inflammatory effect in a dextran sodium sulphate-induced colitis in mice. AFC inhibited LPS-stimulated bone marrow-derived macrophages gene expression program dose-dependently. In vivo, AFC markedly reduced macroscopic and microscopic signs of intestinal inflammation. Besides, it restored the colonic expression of pro-inflammatory and anti-inflammatory mediators, as well as enhanced the expression of intestinal barrier markers. These results demonstrate the potential of AFC extract as a therapeutic tool for the management of inflammatory bowel disease.  相似文献

The response to cis-dichlorobis(cyclopentylamine)platinum(II) (cis-PAD) an antitumour platinum complex, was studied in two strains of murine lymphoma L5178Y cross-sensitive to X-rays and UV light. Dose-survival relationship, DNA synthesis formation of chromatid aberrations, progression through the cell cycle, and growth and viability changes after 1 h cis-PA; treatment at 37 degree C were examined and compared with the effects of X-rays and UV light. In both strains, cis-PAD caused immediate inhibition of progression through the cell cycle, reduced rate of DNA synthesis, delayed appearance of chromatid aberrations, and delayed death; however, there is a marked difference in sensitivity to cis-PAD between L5178Y-S strain (D0 approx. 5.8 microgram/ml) and L5178Y-R strain (D0 approx. 2.5 microgram/ml). In both strains a close resemblance was found between dose-survival relationships after cis-PAD and UV light treatment, respectively.  相似文献
