A method for the analysis of total DNA of Streptomyces glaucescens is described. The relevant steps are (a) extraction and purification of DNA, (b) restriction of DNA samples with type II restriction enzymes, (c) one dimensional separation of restriction fragments by agarose gel electrophoresis. A typical banding pattern was obtained for each wild type strain, independant of growth conditions or age of the culture. Mutant strains exhibited in most cases the same banding pattern as the parent wild type strain. Only in one specific mutant class a fragment of about 9 megadalton was missing.  相似文献

A simple graphical method, COREM, is described for the comparison of quantitative resemblance measures. The basis of the method is the ordered comparison case series (OCCAS) composed of relevé pairs ranked in decreasing order of similarity. For a given OCCAS, values of given resemblance functions are calculated and plotted in COREM.Delta resemblance (DR) is defined as the rate of change between two neighbouring relevé pairs in the OCCAS. An efficient measure shows balanced sensitivity to the common and differential species. With the method COREM locally optimal resemblance measures may be selected for any given set of relevés which produce the sharpest separation of clusters. A reasonable compromise is to use types of measures, such as the similarity ratio (SIM6), Czekanowski coefficient (SIM8), or floristic similarity (SIM9), which have the best chance of being successful in most relevé sets. A study of transformation functions on Braun-Blanquet values with the help of COREM demonstrates that the combined transformation is the most advantageous for clustering. Summary The migration of neural crest and sclerotome cells and the extension of ventral root axons in chick embryos at stages 16–20 were studied by light microscopy as well as scanning and transmission electron microscopy at the leg bud level of fixed specimens. Extensive cellular movements take place in association with an extracellular matrix consisting of microfibrils. The neural crest and sclerotome cells migrate into the large matrix-filled extracellular space surrounding the neural tube and notochord, apparently using microfibril bundles as substratum. The cells exhibit pseudopodia which are closely associated with the matrix fibrils. The fibrils around the notochord show a spatial arrangement indicating that the sclerotome cells are contact-guided to their subsequent positions. Mutual cell contacts, including those established by cell processes, frequently show cytoplasmic electron dense plaques at adjacent membranes. These small plaque contacts might be correlated to contact inhibition of locomotion between the cells and participate in the guidance of cells. The growth cones of extending axons exhibit filopodia contacting both surrounding mesenchyme cells and extracellular fibrils. Following a shift from autotrophic to heterotrophic nutrition, cells of Chlorella protothecoides become sensitive to the stresses of freezing and thawing. The injury then observed at slow rates of cooling cannot be explained by the cellular response to hypertonic solutions, and at faster cooling rates intracellular ice formation was not demonstrated to be damaging. These findings are at variance with suggested mechanisms of injury in other cellular systems. Isolated epidermis of Commelina communis L. and Tulipa gesneriana L. assimilated 14CO2 into malic acid and its metabolites but not into sugars or their phosphates; epidermis could not reduce CO2 by photosynthesis and therefore must be heterotrophic (Raschke and Dittrich, 1977). If, however, isolated epidermis of Commelina communis was placed on prelabelled mesophyll (obtained by an exposure to 14CO2 for 10 min), radioactive sugars appeared in the epidermis, most likely by transfer from the mesophyll. Of the radioactivity in the epidermis, 60% was in sucrose, glucose, fructose, 3-phosphoglyceric acid and sugar phosphates. During a 10-min exposure to 14CO2, epidermis in situ incorporated 16 times more radioactivity than isolated epidermal strips. Isolated epidermis of Commelina communis and Tulipa gesneriana took up 14C-labelled glucose-1-phosphate (without dephosphorylation), glucose, sucrose and maltose. These substances were transformed into other sugars and, simultaneously, into malic acid. Carbons-1 through-3 of malic acid in guard cells can thus be derived from sugars. Radioactivity appeared also in the hydrolysate of the ethanol-insoluble residue and in compounds of the tricarboxylic-acid cycle, including their transamination products. The hydrolysate contained glucose as the only radioactive compound. Radioactivity in the hydrolysate was therefore considered an indication of starch. Starch formation in the epidermis began within 5 min of exposure to glucose-1-phosphate. Autoradiograms of epidermal sections were blackened above the guard cells. Formation of starch from radioactive sugars therefore occurred predominantly in these cells. Epidermis of tulip consistently incorporated more 14C into malic and aspartic acids than that of Commelina communis (e.g. after a 4-h exposure to [14C]glucose in the dark, epidermis, with open stomata, of tulip contained 31% of its radioactivity in malate and aspartate, that of Commelina communis only 2%). The results of our experiments allow a merger of the old observations on the involvement of starch metabolism in stomatal movement with the more recent recognition of ion transfer and acid metabolism as causes of stomatal opening and closing.Abbreviation G-1-P glucose-1-phosphate  相似文献

Synopsis Examination of more than 2,000 alewives (Alosa pseudoharengus) and blueback herring (A. aestivalis) from various areas in the Saint John River, N.B., showed differences in their morphometric and meristic characters. They differed also in length and age, compositions, spawning time, length and age at maturity, back-calculated lengths, and growth parameters. Within each species, between-area comparisons showed significant differences in some of their meristic characteristics.To test a homing hypothesis for alewives, a multivariate analysis was performed on eight meristic characters. Two discriminant function programs, discriminant analysis for two groups and a multiple group stepwise discriminant analyses were used. Values of the generalized Mahalanobis D2 and percent overlap in frequency distributions of meristic characters suggest that considerable straying of fish, especially between adjacent areas occurs during upstream spawning migration.This article is one of several papers presented at the Second European Ichthyological Congress, Paris, 8–15 September 1976, to be published in Environmental Biology of Fishes.  相似文献

Models of the diatom associations found in upper Florida Bay and adjoining sounds have been constructed utilizing Q modal factor-vector analysis and ecologic diversity indices (Shannon index, number of species, evenness). Four distinct associations were defined using Q-mode factor-vector analysis. Two associations were epiphytic, occurring on Thalassia testudinum König—Association I was characterized by Cocconeis placentula Ehr., Association III by Cylindrotheca closterium (Ehr.) Reim. & Lewin and Cocconeis placentula Ehr. The other two associations were epipelic, occurring on the carbonate mud substratum—Association II was characterized by Cyclotella striata (Kütz.) Grun., Rhopalodia gibberula (Ehr.) O. Müller and Surirella fastuosa (Ehr.) Kütz., Association IV by Fragilaria crotonensis Kitton and Cyclotella striata (Kütz.) Grun. The majority of the 161 species identified were present in both the epiphytic and epipelic assemblages. Only 33 species were restricted to the epiphyton, and 18 species restricted to the epipelon. The epipelic assemblage was significantly more diverse than was the epiphytic assemblage. A general trend of increased diversity away from terrestrial environs toward more open areas of water in both the epipelon and epiphyton was also found.  相似文献

α-Amylases secreted by the aleurone layer of whole barley grains were relatively rich in histidine and relatively poor in glutamate/glutamine and serine when compared to other eukaryotic proteins. The secreted α-amylases had an estimated 0.5 residues each of glucose, mannose and N-acetylglucosamine per molecule of protein (MW 41 400 daltons), and gave positive staining reactions for carbohydrate on sodium dodecylsulfate polyacrylamide gels. Because the average α-amylase molecule had less than one sugar residue per enzyme molecule, it was concluded that secreted α-amylases were heterogeneous with respect to glycosylation. A second protein co-purified with α-amylase, but the amino acid composition of this protein was different from that of barley or wheat α-amylase. This protein was composed of two 21 500 dalton polypeptides. No significant amounts of L-leucine (14C-U) were incorporated into this second protein in isolated aleurone tissue during incubation with gibberellic acid, perhaps because much of it was already present in the starchy endosperm at the time of hormone addition.  相似文献
