Enzyme activity, protein contents, and mRNA contents of group II phospholipase A2 (PLA2) in hepatocellular carcinoma (HCC) surgically obtained from 8 patients were compared with those in either its neighboring liver tissues or control liver tissues. The PLA2 specific activity towards the mixed micelles of 1-palmitoyl-2-oleoyl-phosphatidylglycerol and cholate was significantly greater in the tumor tissues (6.62 ± 1.46 nmol/min/mg) than those in the surrounding liver tissues (1.33 ± 0.22 nmol/min/mg) and controls (0.43 ± 0.04 nmol/min/mg). The results of immunoblot analysis using a specific anti-human group II PLA2 antibody and of Northern blot analysis using a human group II PLA2 cDNA as a probe demonstrated that group II PLA2 was responsible for the increased enzyme activity. The contents of immunoreactive group II PLA2 in the tumor tissues (8.81 ± 1.24 ng/mg) were significantly higher than those in the surrounding liver tissues (1.77 ± 0.27 ng/mg); those in the control tissues were below the analytical range of the method used. The group II PLA2 mRNA was also significantly increased in the tumor tissues, compared with that in the surrounding liver tissues, whereas it was not detectable in th controls. This indicates that group II PLA2 in HCC is induced at the pretranslational level.  相似文献

Fossils recognized as early Homo were discovered first at Olduvai Gorge in 1959 and 1960. Teeth, skull parts and hand bones representing three individuals were found in Bed I, and more material followed from Bed I and lower Bed II. By 1964, L.S.B. Leakey, P.V. Tobias, and J.R. Napier were ready to name Homo habilis. But almost as soon as they had, there was confusion over the hypodigm of the new species. Tobias himself suggested that OH 13 resembles Homo erectus from Java, and he noted that OH 16 has teeth as large as those of Australopithecus. By the early 1970s, however, Tobias had put these thoughts behind him and returned to the opinion that all of the Olduvai remains are Homo habilis. At about this time, important discoveries began to flow from the Koobi Fora region in Kenya. To most observers, crania such as KNM-ER 1470 confirmed the presence of Homo in East Africa at an early date. Some of the other specimens were problematical. A.C. Walker and R.E. Leakey raised the possibility that larger skulls including KNM-ER 1470 differ significantly from smaller-brained, small-toothed individuals such as KNM-ER 1813. Other workers emphasized that there are differences of shape as well as size among the hominids from Koobi Fora. There is now substantial support for the view that in the Turkana and perhaps also in the Olduvai assemblages, there is more variation than would be expected among male and female conspecifics. One way to approach this question of sorting would be to compare all of the new fossils against the original material from Olduvai which was used to characterize Homo habilis in 1964. A problem is that the Olduvai remains are fragmentary, and none of them provides much information about vault form or facial structure. An alternative is to work first with the better crania, even if these are from other sites. I have elected to treat KNM-ER 1470 and KNM-ER 1813 as key individuals. Comparisons are based on discrete anatomy and measurements. Metric results are displayed with ratio diagrams, by which similarity in proportions for several skulls can be assessed in respect to a single specimen selected as a standard. Crania from Olduvai examined in this way are generally smaller than KNM-ER 1470, although OH 7 has a relatively long parietal. In the Koobi Fora assemblage, there is variation in brow thickness, frontal flattening and parietal shape relative to KNM-ER 1470. These comparisons are instructive, but vault proportions do not help much with the sorting process. Contrasts in the face are much more striking. Measurements treated in ratio diagrams show that both KNM-ER 1813 and OH 24 have relatively short faces with low cheek bones, small orbits and low nasal openings. Also, they display more projection of the midfacial region, just below the nose. This is not readily interpreted to be a female characteristic, since in most hominoid primates the females tend to have flatter lower faces than the males. The obvious size differences among these individuals have usually been interpreted as sex dimorphism, but, in fact, two taxa may be sampled at Olduvai and in the Turkana basin at the beginning of the Pleistocene. One large-brained group made up of KNM-ER 1470, several other Koobi Fora specimens, and probably OH 7, can be called Homo habilis. If these skulls go with femora such as KNM-ER 1481 and the KNM-ER3228 hip, then this species is close in postcranial anatomy to Homo erectus. The other taxon, including small-brained individuals such as KNM-ER 1813 and probably OH 13, seems also to be Homo rather than Australopithecus. If the OH 62 skeleton is part of this assemblage, then the small hominids have postcranial proportions unlike those of Homo erectus. However, it is too early to point unequivocally to one or the other of these groups as the ancestors of later humans. Both differ from Homo erectus in important ways, and both need to be better understood before we can map the earliest history of the Homo clade. © 1993 Wiley-Liss, Inc.  相似文献

The effect of lysosomal storage diseases on the ultrastructure of human mast cells has not previously been reported. Indeed, there has been little published evidence indicating that mast cells contain typical lysosomes. However, mast cell cytoplasmic granules contain hydrolases similar to those found in lysosomes, but which differ from lysosomal hydrolases in exhibiting optimal activity at higher pH. We therefore examined by transmission electron microscopy the dermal mast cells in 58 biopsies of patients exhibiting 1 of 29 different lysosomal storage diseases. We found mast cells containing abnormal lysosomes in 16 of these disorders. In 6 of these 16 diseases, the mast cells' cytoplasmic granules appeared normal. These observations indicate that human mast cells can contain lysosomes, and provide evidence that the enzymes affected by lysosomal storage diseases are active in mast cells.  相似文献

A tooth at the border between two morphogenetic fields (mandibular canine and honing premolar) may become morphologically similar to and/or functionally incorporated with the teeth of either field. In light of this, observations of the morphology and occlusion of female anthropoid C1s from 58 extant species are presented to assess whether and to what extent they exhibit incisor-like form and function. Female C1s in 74% of the taxa observed exhibit well developed incisor-like traits which may reflect field border phenomena. In another 9%, incisor traits are present but they are not examples of field border phenomena. Interspecies variation in female C1 morphology is related to behavior, function, natural selection and phyletic inertia. A selection model, derived from the data, is used to explain C1 sexual dimorphism and the evolution of male and female human canines. The data's relevance to the field vs clone theory debate is also discussed.  相似文献

PurposeBeryllium is known to have adverse health effects and is classified as carcinogenic to humans. However, data on systemic beryllium exposure in humans are rare and especially human toxicokinetics are largely uncharted. As such, the first reported multi-annual course of blood and urine concentrations after a high exposure scenario provides important new insights.MethodsFor a medical follow-up biomonitoring samples were collected for 56 months from a male subject after an accidental and multi-faceted high exposure. Sampling started on day 2 post-exposure for urine and day 147 for blood. The samples were analyzed by inductively coupled mass spectrometry (ICP-MS) and plotted longitudinally as a function of time. Terminal half-lives were calculated assuming a first-order elimination process.Main findingsBoth matrices showed highly increased initial concentrations (about 100-fold), despite the 147-day delay in blood sampling, and a marked decline over time. In urine, a two-phase excretion process was suspected based on the longitudinal data. Calculations gave terminal half-lives of 117.5 days and 666.5 days for phases 1 and 2, respectively. Blood kinetics called for a terminal half-life of 103.5 days. Elimination kinetics in blood and urine were comparable, simultaneously gathered samples showed an excellent correlation (R² = 0.985).Principal conclusionsThe long-term follow-up after a high initial exposure to beryllium provides the first detailed insights into the elimination course of systemically available beryllium in humans. Conform kinetics of beryllium in urine and blood and the strong correlation between both parameters indicate high data validity and support the good representation of the current systemically available beryllium by urine and blood concentration in humans. The relatively long terminal half-lives in both matrices suggest a possible accumulation in humans in case of repeated exposures.  相似文献

IntroductionSeaweeds are a rich source of elements such as iodine, and are also able to accumulate contaminants such as trace elements.MethodsThe aim of this study was to assess the dietary exposure as well as the risk from iodine and trace elements in edible seaweeds for the French population using current consumption data. The contribution of seaweeds to overall dietary exposure to trace elements and iodine was evaluated, and for those substances with minimal contribution to overall dietary exposure, simulations were performed to propose increased maximal limits in seaweeds.ResultsCadmium, inorganic arsenic and mercury in seaweeds were very low contributors to total dietary exposure to these contaminants (0.7 % 1.1 % and 0.1 % on average, respectively). Dietary exposure to lead via seaweed may contribute up to 3.1 % of total dietary exposure. Dietary consumption of iodine via seaweed may contribute up to 33 % of total exposure to iodine, which makes seaweeds the strongest contributor to iodine in diet.DiscussionNew maximal values in seaweeds are proposed for the very low contributors to total dietary exposure: 1 mg/kg dw for cadmium, 10 mg/kg dw for inorganic arsenic and 0.3 mg/kg dw for mercury.  相似文献

Orotic acid (OA) is a natural product that acts as a precursor in the pyrimidine nucleotide biosynthesis pathway. Most studies concerning administration of OA focus on its therapeutic effects; however, its effect on tumours is unclear. We aimed to determine whether treatment with OA influences the viability and apoptosis of normal (HGrC1) and tumour-derived (KGN) human ovarian granulosa cells. The effects of OA (10–250 μM) on viability and apoptosis of both cell lines were determined by using alamarBlue and assessing caspase-3/7 activity, respectively. Annexin V binding and loss of membrane integrity were evaluated in KGN cells. The cell cycle and proliferation of HGrC1 cells were assessed by performing flow cytometric and DNA content analyses, respectively. The influence of OA (10 and 100 μM) on cell cycle- and apoptosis-related gene expression was assessed by RT-qPCR in both cell lines. Mitochondrial activity was analysed by JC-1 staining in HGrC1 cells. In KGN cells, OA reduced viability and increased caspase-3/7 activity, but did not affect mRNA expression of Caspase 3, BAX, and BCL2. OA enhanced proliferation and mitochondrial activity in HGrC1 cells without activating apoptosis. This study demonstrates that the anti-cancer properties of OA in ovarian granulosa tumour cells are not related to changes in apoptosis-associated gene expression, but to increased caspase-3/7 activity. Thus, OA is a promising therapeutic agent for ovarian granulosa tumours. Further, our results suggest that differences in basal expression of cell cycle- and apoptosis-related genes between the two cell lines are responsible for their different responses to OA.  相似文献

Recent rescue excavations at Bercy (Paris), a site on the alluvial plain of the Seine valley, yielded plant remains which are associated with the recent occupation phase dating from the middle Neolithic II (Chasséen), when a village was established on the former channel of the river Seine. Various contexts (the channel, the flat lower part of the bank, and several archaeological features) have been studied and 84 taxa have been identified. Cultivated plants are represented by Triticum aestivum/durum, T. dicoccum, T. monococcum and Hordeum vulgare. Among the wild plants with potentially edible fruits or seeds, only very few satisfy the various criteria for association with human activities: Corylus avellana, Vitis sylvestris, Cornus sanguinea, Quercus sp. and Prunus spinosa of which carbonized fruits were also present, were found in archaeological features and were very abundant. Though not found in archaeological features, we consider that Crataegus monogyna (carbonized and well represented) and Rubus spp. (especially abundant) were not deposited there naturally and had also been intentionally collected. Finally, it is suggested that the allochthonous (varied) origin of these taxa is the reason why there are no concentrations of their fruits in the channel. The exploitation of wild seeds and fruits appears to have been very selective. All other wild taxa can be attributed to natural deposition.  相似文献

采用基因工程方法制取人胸腺素α原获得成功。用20ug/ml植物血球凝集素(PHA)和500U/ml重组人白细胞介素2(IL-2)联合刺激人胎儿胸腺细胞,从中提取总RNA,经反转录PCR获得了人胸腺素α原cDNA;将之克隆入pUC19中,序列测定表明与已报道序列一致,进一步将之亚克隆入原核表达载体pBV220,转化大肠杆菌DH5a.观察到在不改变氨基酸编码的前提下,增加胸腺素a原上游引物中A、T含量,可以明显提高胸腺素α原的表达量,同时,不同培养基对它的表达也有影响。胸腺素α原在大肠杆菌中以可溶形式表达,不需复性。初步活性测定显示,它可明显刺激人外周血淋巴细胞E-玫瑰花结形成率。重组人胸腺素α原在大肠杆菌中表达,为其临床应用及基础研究奠定了基础。  相似文献
