Abstract. Direct observations on the effect of rapid freezing and thawing on honey bee ( Apis mellifera L.) sperm were made by light, scanning and transmission electron microscopy. Rapid freezing of honey bee ejaculated sperm, suspended in freezing diluent, in liquid nitrogen followed by rapid thawing can cause cellular injuries which lead to the death of the sperm. The frozen-thawed sperm, supravitally stained, showed a significant decrease in cell viability compared with that of the control fresh sperm ( P <0.001). Significant uptake of the stain in the dead sperm resulted from damage in the cell membrane. The scanning electron micrographs of frozen-thawed sperm further demonstrated that the injury of cell membrane can lead to the splitting of mitochondrial derivatives from the flagellar axoneme. More cellular injuries including the release of acrosomal content and membrane damage at the acrosome, nucleus and the tail regions were further revealed by transmission electron microscopy. The impact of cellular injuries on the quality of honey bee sperm cryopreserved for artificial insemination of honey bee queens is discussed.  相似文献

ABSTRACT. Immediately after visiting cells containing immature queens, workers of Apis mellifera L. were observed to engage in prolonged cleaning, particularly of their tongues when they had visited larvae, and of their antennae when they had visited pupae. Thereafter other workers usually initiated and made antennal contacts with them. During such antennal contact the bee that had visited the queen larva often donated food. The implication of these findings on the distribution of pheromone produced by immature queens is discussed. Workers were stimulated to make antennal contact with the excised heads of bees from a queen's court, providing further evidence that queen pheromone is transferred between workers' antennae.  相似文献

The Asian weaver ant, Oecophylla smaragdina, is known to have outstanding predatory power. This ant can protect the host plants from attacks of phytophagous insects and therefore has been used for biological control in the tropics. We present evidence for a possible negative effect of Oecophylla on the performance of host plants. Our observation in a fruit orchard of rambutan in Sumatra suggested that the presence of Oecohylla nests on the trees statistically significantly lowered the flower-visiting rate of flying insects, involving the major pollinator Trigona minangkabau. The visiting rate of Oecophylla workers to each flower shoot of rambutan significantly negatively correlated with the visiting rate of flying insects. Empirical evidence of such an inhibitory effect on flower-visiting of pollinators cased by aggressive ants has been scarce so far.  相似文献

Bee bread (BB) is a beehive product generated upon fermentation of pollen combined with flower nectar and glandular secretions. The potential application of BB is related to its nutritional and functional components, including phenolic compounds. This is the first prospective study on palynological parameters, phenolics, antioxidant, and antibacterial activity of Chilean bee bread in vitro. The tested material exhibited high levels of phenolics (1340±186 mg GAE/100 g BB) and showed antioxidant capacity as determined by the FRAP (51±2 μmol Trolox equivalent/g BB) and ORAC-FL (643±64 μmol Trolox equivalent/g BB) and antibacterial activity against Streptococcus pyogenes. Furthermore, the phenolic acids and flavonoids was determined using liquid chromatography-mass spectrometry, and the concentration was determined using liquid chromatography with diode array detection. Kaempferol, quercetin, ferulic acid, and rutin were the main phenolics found. This study demonstrates the bioactive potential of Chilean BB and supports the evidence that this bee product is a promising source of antioxidants and antimicrobial compounds.  相似文献

The pollinators of 29 ginger species representing 11 genera in relation to certain floral morphological characteristics in a mixed-dipterocarp forest in Borneo were investigated. Among the 29 species studied, eight were pollinated by spiderhunters (Nectariniidae), 11 by medium-sized Amegilla bees (Anthophoridae), and ten by small halictid bees. These pollination guilds found in gingers in Sarawak are comparable to the pollination guilds of neotropical Zingiberales, i.e., hummingbird-, and euglossine-bee-pollinated guilds. Canonical discriminant analysis revealed that there were significant correlations between floral morphology and pollination guilds and suggests the importance of plant–pollinator interactions in the evolution of floral morphology. Most species in the three guilds were separated on the plot by the first and second canonical variables. Spiderhunter-pollinated flowers had longer floral tubes, while Amegilla-pollinated flowers had wider lips than the others, which function as a platform for the pollinators. Pistils and stamens of halictid-pollinated flowers were smaller than the others. The fact that gingers with diverse morphologies in a forest with high species diversity were grouped into only three pollination guilds and that the pollinators themselves showed low species diversity suggests that many species of rare understory plants have evolved without segregating pollinators in each pollination guild.  相似文献

The ultrastructural localization of the enzyme acetylcholinesterase (AChE) in the ocellus of the honey bee (Apis mellifica) was studied by electron microscopy. High AChE activity was found both in the receptor-cell axons and in the surrounding glial cells. Second order neurones exhibited a remarkably lower anzyme activity. AChE was also detected in the intercellular spaces between the receptor-cell axons and the second order neurones. These results provide additional support to the assumed cholinergic nature of the photoreceptor cells in the insect ocellus.  相似文献

Dillenia suffruticosa (Dilleniaceae), a common pioneer shrub in Southeast Asia, is broadly distributed along roadsides. This species flowers throughout the year and is thought to be a keystone species for maintaining populations of flower visitors. To examine the flower visitors of this species, we conducted field surveys and described the behaviors of each flower visitor species within four different research sites: Primary Forests 1 and 2, Secondary Forest, and Mixed Vegetation. We caught 1087 individuals belonging to 51 species from 11 insect families and one species of arachnid. Most flower visitors belonged to Hymenoptera (98.6% of individuals); Trigona spp. and Xylocopa spp. (Apidae) accounted for 64.0% and 20.0%, respectively, of all caught individuals. According to observations of behavior by each species, effective pollinators may include Xylocopa spp. and Amegilla spp. (Apidae). We compared the research results among four research sites and found that the effective pollinators were not different among sites. However, the number of species and the abundances of flower visitors decreased in disturbed areas, especially in Secondary Forest. This research suggests the effects of human disturbances on the diversity and abundance of flower visitors.  相似文献

Bee venom is a natural mixture and candidate anti-cancer agent with selective cytotoxic effect on some cancer cells. However, the cellular mechanisms of how bee venom selectively targets cancer cells remain elusive. The aim of this study was to reveal the genotoxic effect of bee venom in concordance with the location of β-actin protein throughout the nucleus or/and cytoplasm. For this aim, the level of H2AX phosphorylation (γH2AX) and intracellular location of β-actin were assessed by immunofluorescence in liver (HEPG2) and metastatic breast (MDA-MB-231) cancer cell lines compared to normal fibroblasts (NIH3T3) after bee venom treatment. Colocalisation profiles of γH2AX and β-actin in each cell line were also analysed. The results showed that the levels of γH2AX staining decreased in normal cells but increased in cancer cells. The majority of β-actin was localised within the cytoplasm of normal cells after bee venom treatment, but it was mostly accumulated within the nucleus in cancer cells. Colocalisation of β-actin and γH2AX both in nucleus and cytoplasm was induced in each cancer cell by different patterns. The results showed that normal and cancerous cells had different responses against bee venom, and suggested that bee venom induced a cellular response by the interaction between γH2AX and β-actin.  相似文献
