DNA指纹图谱对新品种选育、种质资源保存和管理具有重要的意义。然而,利用SSR标记构建红麻DNA指纹图谱的研究仍十分有限。在本研究中,利用课题组开发并筛选出的131对SSR引物,分析不同来源的96份红麻种质资源,包括红麻品种审定的区试对照品种福红952。结果表明,131对引物共扩增出375条带,平均每对引物扩增出2.6条带。以遗传相似系数0.614为切割线时,可以分为2个类群,52个为类群P1,44个为类群P2;以遗传相似系数0.710做切割线,可分为5个亚群。利用这131对引物标记所得的数据成功绘制了一份85个品种独特的指纹图谱,其中福红952可被HcEMS238引物特异识别。其他11份因存在遗传相似性高的现象,未被识别。上述结果为红麻品种的真实性鉴定及遗传多样性分析提供依据。 相似文献

Five cultivars of Hibiscus esculentus L. and six cultivars of Vigna sinensis (L.) Endl. were tested for their relative resistance to Pseudocercospora spp. Differences in susceptibility to the leaf spot pathogen were observed in both host and non-host interactions. The H. esculentus cv. South Sea and V. sinensis cv. Purple Mart were most susceptible to Pseudocercospora abelmoschi and P. cruenta, respectively. The H. esculentus cv. Pure Luck was most tolerant to P. abelmoschi while the V. sinensis cv. KY Bush was highly resistant to P. cruenta. A distinct host-specific interaction was observed among the different species of Pseudocercospora. Cultivar specific interactions were most pronounced between V. sinensis and P. cruenta. A direct correlation was observed between the variation in peroxidase activity in the soluble fraction of inoculated leaves and resistance to infection in H. esculentus and V. sinensis cultivars. The soluble fraction of inoculated leaves had higher peroxidase activity than either mitochondrial or chloroplast extracts. 相似文献

Summary The biological effects of irradiation with12C+5 ion beam on plant cells have been analyzed. Protoplasts and cell suspensions ofLavatera thuringiaca, and a somatic hybrid callus (Hibiscus rosa-sinensis +Lavatera thuringiaca), were irradiated with doses from 0.05 to 50 Gy, and the effects on cell growth, cell division, cell viability and embryogenesis rates were analyzed. Irradiation with12C+5 ion beam at relatively very low doses (5.0 Gy) significantly inhibited cell division, yet the survival rate and regeneration capability of the cells through somatic embryogenesis were conserved in more than 70 and 50 %, respectively. These results indicate that cell division is the most sensitive parameter to irradiation, accounting for the inhibition of colony formation and callus growth. The potential use of the12C+5 ion beam in asymmetric protoplast fusion experiments is discussed. 相似文献

花色苷在植物中呈现粉红、红、紫红、紫等颜色,可以用作食品、药品及化妆品的着色剂,亦有药用价值。作为食品添加剂,颜色较合成色素自然,且安全无毒性。早在1987年,Mizukami[1]就建议用植物细胞培养物生产花色苷类代替合成色素。所有的植物培养细胞都是异源性的。各细胞之间产花色苷的能力相差很大[2].因为产花色苷的细胞系带有颜色标记,所以容易识别并通过肉眼选择即可获得高产花色苷的细胞系。筛选的方法很多,如平板饲喂法[3]、小细胞团法[4]、细胞块法[5]、肉眼观察直接挑选法及细胞分栋器法[6]等。高产系花色苷的含量可增加几倍到几十倍,而且产量稳定。本文采用平板法及小细胞团法筛选高产花色苷的玫瑰茄(Hibiscus sabdariffa L.)细胞系。 相似文献

半红树植物是一类既可在海岸潮间带又可在陆地生长和繁殖的海陆两栖植物。夏季强光和高温是海岸带生境突出的特点,半红树植物如何在这样的自然生境中生存和繁衍是值得探讨的问题。该研究以具有较高应用价值的锦葵科(Malvaceae)木槿属(Hibiscus)的常绿半红树植物黄槿(H.tiliaceus)作为研究对象,采用LI-6400便携式光合测定系统(Li-Cor Inc.,USA)对南亚热带气候条件下的广东省珠海淇澳岛红树林自然保护区自然生长的黄槿在夏季高温季节的叶片气体交换特性进行测定,探讨了其在自然生境下的光合行为和相关的形态解剖特征,揭示其生境适应性的光合生理生态机制。结果表明:(1)黄槿净光合速率、气孔导度、胞间二氧化碳浓度和蒸腾速率的日变化均表现为单峰型曲线,中午未出现明显光抑制现象。(2)黄槿较高的净光合速率与其较高的气孔导度相关,而后者则可能与其较大的气孔密度和具有表皮毛等叶片解剖结构特征有关。(3)黄槿光饱和点、光补偿点、最大净光合速率和日均净光合速率均符合阳生植物特征,因而是一种阳生性树种。以上结果说明黄槿在自然环境中具有的快速生长特性可能与其较强和较稳定的光合能力有关,该研究结果为深入揭示黄槿以及其他半红树植物适应海岸带高温强光生境的生理生态机制提供了基础数据,为黄槿的合理栽培及造林实践提供了理论参考。 相似文献

Phylogenetic relationships and the spatial genetic structure of a pantropical plant with sea-drifted seeds, Hibiscus tiliaceus L., and its allied species were investigated. The combined distribution range of these species is over almost the entire littoral area of the tropics worldwide, which might result from the dispersal of their sea-drifted seeds and from recurrent speciation in local populations. A phylogenetic tree constructed using the nucleotide sequences of a c. 7500-bp portion of chloroplast DNA suggested the possibility that recurrent speciation from H. tiliaceus has given rise to all of its allied species. Three major sequence haplotypes of H. tiliaceus had wide and overlapping distributions throughout the Pacific, Atlantic and Indian Ocean regions. This distribution pattern was also confirmed by PCR-SSCP (polymerase chain reaction amplification with single-strand conformation polymorphism) and PCR-SSP (PCR amplification with sequence specific primers) analyses performed on more than 1100 samples from 65 populations worldwide. Statistical analysis using F(ST) and analysis of molecular variance did not show significant genetic differentiation among the H. tiliaceus populations in the three oceanic regions. The results reported here suggested substantial gene flow occurred between populations in the different oceanic regions due to sea-drifted seeds. A strong genetic difference between the Pacific and Atlantic populations of Hibiscus pernambucensis Arruda was observed, which indicates that gene flow in this species between the two regions has been prevented. The wide and dominant distribution of a haplotype shared by H. pernambucensis and H. tiliaceus in the Atlantic region suggests significant introgression between the two species in this region. 相似文献

2008年8月,在广东省广州市发现一种严重危害扶桑的害虫,经鉴定确认为扶桑绵粉蚧Phenacoccus solenopsis Tinsley。2008年12月16日调查发现,在广州市的扶桑上多点发生。扶桑绵粉蚧原产美洲,最近几年入侵印度和巴基斯坦,严重危害棉花,其暴发危害已经造成了重大损失,成为威胁世界棉花安全的重大入侵害虫。文章提供扶桑绵粉蚧的形态特征、寄主、分布、危害等情况,并对其检疫与防控措施等提出建议。 相似文献

Differences in pollen tube growth rates (certation) between heterospecific (foreign) and conspecific pollen may strongly influence whether hybrid offspring are produced after mixed pollen loads are delivered to a stigma. For both members of a sympatric species pair, Hibiscus moscheutos and H. laevis, pollination by pure loads of foreign pollen resulted in fruit set that was not significantly different from conspecific pollination, indicating that pure loads of foreign pollen could readily result in hybrid offspring. However, the number of seeds per fruit from pure foreign pollinations was significantly less than that of pure conspecific pollination. Simultaneous mixed pollination resulted in a proportion of hybrid seeds (detected by an electrophoretic marker enzyme) that was significantly lower than expected based upon the capacity of foreign pollen to effect fertilization when applied in pure pollinations. After these 50/50% pollen mixtures were applied to stigmas, 8.0 and 7.4% hybrids were produced when H. moscheutos and H. laevis were the ovule parents, respectively. For these Hibiscus species, pollen competition appears to function as a barrier to hybridization that is of moderate intensity compared with similar barriers occurring between other recently studied sympatric species pairs. 相似文献

Mature seeds of diploid and tetraploidHibiscus species were analyzed for enzyme activity (alcohol dehydrogenase, malate dehydrogenase, leucine aminopeptidase), total protein content, DNA amount and dry weight.
