正氮类污染物是造成水体污染和富营养化的主要原因,水体中氮的去除对于清洁水体有着重要的意义~([1-2])。因此,高效与便捷地去除污水中的氮是污水处理中的关键问题,而生物脱氮又被认为是目前废水脱氮中最经济有效的方法之一~([3])。最初研究者们认为只有自养硝化细菌才能进行高效的硝化作用,近年来研究表明,异养硝化微生物同样在生物硝化过程中起着不可忽视的作用~([4])。异养硝化微生物相比传统的自养微生物在废水生物脱氮中占有明显优势,具有良好的应用前景。  相似文献

【背景】异养硝化-好氧反硝化菌由于能够同时实现硝化反硝化作用而备受关注,但由于菌的种类不同,其脱氮途径不尽相同,研究菌株脱氮关键酶的种类及其活性可以推测菌株的脱氮途径,从而为菌株在生产上的应用提供技术支撑。【目的】研究Pseudomonas alcaliphila AD-28的脱氮性能及其关键酶的活性,为菌株脱氮分子机理研究奠定基础。【方法】以柠檬酸钠为碳源,以硫酸铵、亚硝酸钠、硝酸钾为氮源,研究菌株AD-28的脱氮性能并检测其关键酶氨单加氧酶(AMO)、羟胺氧化还原酶(HAO)、亚硝酸盐还原酶(NIR)、硝酸盐还原酶(NAR)的酶活性。【结果】菌株AD-28培养24h的菌密度(OD600)可达1.971,对初始浓度为18.85mg/L的氨氮、26.13mg/L的硝酸盐氮、19.47mg/L的亚硝酸盐氮、66.11 mg/L的总氮去除率均达到96%以上;关键酶AMO、HAO、NIR和NAR的比活力分别为0.028、0.003、0.011、0.027 U/mg。【结论】Pseudomonas alcaliphila AD-28能同时进行异养硝化-好养反硝化作用,该菌在AMO作用下将NH4+-N氧化为羟胺,然后由HAO氧化为NO2--N,NO2--N和NO3--N在NIR、NAR等酶的催化作用下脱氮。  相似文献

This work aims to outline the dynamics of trophic links between the three main microbial components (bacteria, nanoflagellates, and ciliates) of the Farasan Archipelago in order to establish a baseline for future research in this area. The Farasan Archipelago lies along the southwestern coast of the Saudi Arabia, southern Red Sea between 16°20′–17°10′N and 41°30′–42°30′E and had been declared as marine and terrestrial reserve by the year 1996. Three different sites were chosen for this study, with each site visited bimonthly for 18 months from September 2016 to February 2018. Bacteria, nanoflagellates and ciliates were enumerated in order to explore the complex interactions between the main microbial categories in sea waters of the Farasan Archipelago. High abundances were recorded during the present study for bacteria (8.7 × 106 bacteria ml−1), nanoflagellates (3.7 × 104 TNAN ml−1) and ciliates (40.4 ciliates ml−1). The paper discusses the various potential pathways controlling the complex interactions between these microbial groups in this part of the southern Red Sea. It is concluded that a linear trophic chain consisting of bacteria; heterotrophic nanoflagellates; filter feeding ciliates is a major route by which the production of bacteria is transferred to the higher consuming levels, thereby confirming the high importance of t bottom-up control (food supply), alongside top-down control (predation) in regulating bacterial abundances in the Farasan Archipelago. During the present investigation, each nanoflagellate ingested between 11 and 87 bacteria in one hour, while each ciliate consumed between 20 and 185 nanoflagellates every hour. These calculated grazing rates of protistan eukaryotes confirmed the role of heterotrophic nanoflagellates as the main consumers of bacteria, and the role of ciliates as the major control for the heterotrophic nanoflagellate population dynamics, and thus the top predators within the microbial plankton assemblage in the Farasan Archipelago.  相似文献

【背景】同型产乙酸菌是一类利用乙酰辅酶A途径固定CO_2合成自身细胞物质并生成乙酸、乙醇等代谢产物的厌氧菌群,其分布广泛、种类繁多且代谢多样。深入研究同型产乙酸菌菌株的代谢能力及特性,对探索该种群的生理生化特性及其环境作用至关重要。【目的】研究一株同型产乙酸菌Clostridium sp. BXX的最适培养条件及其自养与异养生长特性。【方法】设置BXX菌株培养温度10-55°C、初始pH 6.0-9.0、NaCl浓度0-2.0%、不同氮源,测定菌体细胞含量和产物生成浓度,确定菌株最适培养条件。研究BXX菌株分别以H_2/CO_2、合成气、CO、葡萄糖、1,2-丙二醇、甲酸钠、乙二醇甲醚、甘油、丙酮酸和乳酸为底物时的底物消耗、产物生成、菌体细胞含量和pH等,探究其自养和异养生长特性。【结果】BXX菌株的最适培养温度为30°C,初始pH为7.0,NaCl浓度为1.0%,氮源为酵母粉。BXX菌株能以H2/CO2、合成气、葡萄糖、1,2-丙二醇、甲酸钠、乙二醇甲醚和甘油为底物生长,不能以CO、丙酮酸或乳酸为底物生长。【结论】BXX菌株既能自养生长产乙酸,又能异养生长产乙醇。BXX菌株是乙酸发酵的优良菌种资源,有较好的工业应用潜力。  相似文献

【目的】研究不同金属离子对异养氨氧化细菌C16的生长和脱氮性能影响,探讨适于C16生长和脱氮的金属离子及其浓度。【方法】实验选用Mg2+、Mn2+、Fe2+、Cu2+、Zn2+5种金属离子,对C16的生长﹑脱氮性能﹑亚硝酸盐氮积累以及相关酶活性进行研究。【结果】Mg2+明显促进C16的生长和NH4+-N氧化速率;较高浓度Mn2+使得C16无法生长;原培养基中缺少Fe2+会抑制C16的生长和NH4+-N氧化速率;在原培养基中加入0.1 mmol/L的Cu2+对C16的生长和脱氮具有一定的促进作用,Cu2+使得培养基中基本无NO2--N和NH2OH的积累;不同浓度的Zn2+对C16的生长和氨氮去除有抑制作用。酶活实验结果显示,0.1 mmol/L Mg2+促进了羟胺氧化还原酶(HAO)的活性;0.1 mmol/L Cu2+促进了硝酸盐还原酶(Nar)和亚硝酸盐还原酶(Nir)的活性。【结论】Mg2+是C16生长和脱氮过程中的一种重要金属离子;加入Cu2+可避免过量亚硝酸盐积累。  相似文献

【背景】海洋浮游植物产生的有机物质与异养细菌之间的相互作用是上层海洋物质和能量循环过程中的重要组成部分。【目的】探究寡营养海域微生物群落对聚球藻有机物(Synechococcus-derived organic matter,SOM)的响应和代谢利用过程,加深对海洋微生物介导的生物地球化学循环过程的认识。【方法】利用南海海域微生物群落,添加聚球藻有机物质后黑暗培养,对培养过程中有机碳、营养盐和活性微生物群落结构的变化进行追踪。【结果】在短期的培养过程中,有60%-73%的SOM被微生物所利用。γ-变形菌(Gammaproteobacteria)在培养过程中响应最快,也是最优势微生物类群。SOM的加入改变了原位微生物群落结构,并且随着活性有机物质逐步被消耗,微生物群落结构也发生了演替。【结论】SOM中的大部分物质都属于活性有机物质,可以快速地被微生物所降解利用。不同微生物类群响应不同生物可利用性有机物质,推动了海洋物质和能量的循环。  相似文献

The flux of carbon and nutrients through aquatic systems is largely dependent upon interactions between autotrophic and heterotrophic processes. As a means of assessing the relative importance of autotrophy and heterotrophy in large rivers, we compared phytoplankton production, heterotrophic bacterial production and community respiration in three regulated rivers of the Midwestern USA. Samples were collected monthly (March to December 1999) from impoundments of the Ohio (McAlpine Pool), Cumberland (Lake Barkley), and Tennessee (Kentucky Lake) Rivers. Bacterial production was tightly coupled to phytoplankton production at each site (r 2 = 0.63–0.85). Ratios of phytoplankton production to bacterial production ranged from <1 to 15 in the Tennessee and Cumberland Rivers and 2 to 90 in the Ohio River. The ratio of primary production to community respiration (P:R) ranged from 0.03 to 2.76 across all sites, with average P:R values lower in the Ohio River (0.14) than the Tennessee River (0.39) and the Cumberland River (1.10). P:R values above unity (P > R) were observed only in the Tennessee and Cumberland Rivers during seasonal (April–July) spikes in primary production. We estimate that 3, 6, and 20% of annual bacterial carbon requirements were met by exudates from in situ phytoplankton in the Ohio River, Tennessee River, and Cumberland River, respectively. Our findings indicate that heterotrophic bacteria were largely dependant upon allochthonous carbon. Rheological properties of Chlorella pyrenoidosa culture grown heterotrophically in a 14 L fermentor were investigated. It was found that the fluid viscosity was rather low and remained almost unchanged during the cultivation, implying that no (or very few) viscous substances were excreted into the medium. Investigation of the condensed suspension of C. pyrenoidosa showed that for biomass concentration under 150 g.L−1, the suspension of C. pyrenoidosa exhibited Newtonian behavior, and the fluid viscosity was rather low (about 40 mPa·s) and increased very slowly with the increase in cell concentration. With further increase in biomass concentration however, the fluid rheological behavior changed to non-Newtonian, and the fluid viscosity increased rapidly with the increase in cell concentration. From the viewpoint of rheology, C. pyrenoidosa is an excellent organism for high-cell-density culture, and there will be no rheological problems at cell densities under 150 g.L−1.  相似文献

The intracellular symbiotes of Blastocrithidia culicis and Crithidia oncopelti can be eliminated from cultures of the flagellates by a single chloramphenicol (CAP) treatment. Effective dosages were determined to be 0.01–0.08% (w/v) CAP after a treatment for 2 weeks or more for B. culicis and 0.08% (w/v) after 1 month for C. oncopelti in most cases. Ineffective dosages only lowered the numbers of symbiote-bearing flagellates. Growth of both species of flagellates in the presence of CAP was reduced in proportion to the drug concentration. Repeated subcultures at effective dosages yielded symbiote-free flagellates, which maintained a low level of growth rate. After repeated subcultures at ineffective dosages, the growth rate rose and the symbiote-bearing cells, initially very few, increased in number. The lowest effective dosages proved to be marginal, often producing symbiote-free cultures, but occasionally cultures with a few symbiote-bearing cells. After repeated subcultures at these drug concentrations, symbiote-containing cultures grew faster than the symbiote-free cultures. Hence, the symbiotic bacteria benefit the growth of their hosts, perhaps by supplying essential factors that are inadequate even in a rich blood medium.  相似文献

Abstract

The Structural Heterogeneity of Plants with a Thallus. Plants that evolved in aquatic environments developed a thallus, that is a multicellular body without structures deputed to the mass transportation of water along great distances. The structural organisations of thalli shows a number of different solution to reach the coordination necessary to the expression of differentiation in the multicellular thalli. Some of these structures are comparatively esamined, pointing out their peculiarities. The diversity observed among plants evolved in aquatic environments is expressed not only by the taxa that can be recognized, but also by a great number of solutions developed to express the differentiation of parts in a complex multicellular body.  相似文献
