The study of cultured strains has a long tradition in protistological research and has greatly contributed to establishing the morphology, taxonomy, and ecology of many protist species. However, cultivation‐independent techniques, based on 18S rRNA gene sequences, have demonstrated that natural protistan assemblages mainly consist of hitherto uncultured protist lineages. This mismatch impedes the linkage of environmental diversity data with the biological features of cultured strains. Thus, novel taxa need to be obtained in culture to close this knowledge gap. In this study, traditional cultivation techniques were applied to samples from coastal surface waters and from deep oxygen‐depleted waters of the Baltic Sea. Based on 18S rRNA gene sequencing, 126 monoclonal cultures of heterotrophic protists were identified. The majority of the isolated strains were affiliated with already cultured and described taxa, mainly chrysophytes and bodonids. This was likely due to “culturing bias” but also to the eutrophic nature of the Baltic Sea. Nonetheless, ~ 12% of the isolates in our culture collection showed highly divergent 18S rRNA gene sequences compared to those of known organisms and thus may represent novel taxa, either at the species level or at the genus level. Moreover, we also obtained evidence that some of the isolated taxa are ecologically relevant, under certain conditions, in the Baltic Sea.  相似文献

The effects of external carbon source (both type and concentration) and empty bed contact time (EBCT) on denitrification efficiency during simultaneous heterotrophic and sulfur-utilizing autotrophic denitrification were evaluated. Continuous experiments were conducted with up-flow mode sulfur packed bed reactors (SPBRs) fed with nitrified leachate containing 700–900 mg/l NO3–N. The fraction of NO3–N removed by heterotrophic denitrification (HDNRfraction) for alkalinity production to balance the alkalinity consumption by autotrophic denitrification varied with the type of external carbon source. When methanol or sodium acetate was added at HDNRfraction values of 60 and 44%, respectively, 100% denitrification was achieved without alkalinity addition. However, glucose required a HDNRfraction value of 70% for complete denitrification and denitrification was not complete when molasses was used at a HDNRfraction value of 70%. The EBCT and volumetric loading rate at which 100% denitrification efficiency could be achieved were 6.76 h and 2.84 kg NO3–N/m3 day, respectively. The maximum nitrogen removal rate was 5.05 kg NO3–N/m3 day observed with 89% removal efficiency. At short HRT, a clogging problem was observed near the bottom of the SPBR with excess growth of heterotrophic denitrifiers and gas accumulation within the pores of the SPBR. This problem may be eliminated by back-washing or by separating heterotrophic denitrification from sulfur-utilizing denitrification.  相似文献

  • 1 The population density, diversity and productivity of the microbial plankton in an oligotrophic maritime Antarctic lake were studied for a 15‐month period between December 1994 and February 1996.
  • 2 In the lake, concentrations of nutrients and dissolved organic carbon were uniformly low, temperature varied over a small annual range of 0.1–3 °C, and the surface was ice‐covered except during a period of approximately 6 weeks in summer.
  • 3 The total of 57 morphotypes of protozoa observed during the study is a higher taxonomic diversity than previously reported from continental Antarctic lakes, but lower than that found in more eutrophic maritime Antarctic lakes. Likewise, planktonic abundance and productivity were lower than has been reported in other lakes on Signy Island, but generally higher than those of lakes on the Antarctic continent.
  • 4 There were marked seasonal and interannual variations in planktonic population density.
  • 5 Chlorophyll a concentrations ranged from undetectable to 4.2 µg L‐1 and the greatest rate of primary productivity measured was 4.5 mg C m‐3 h‐1. The phytoplankton was dominated by small chlorophytes and chrysophytes, with phototrophic nanoflagellate abundance ranging from 1.1 × 103 to 1.2 × 107 L‐1.
  • 6 Bacterial densities of 3.6 × 108 to 1.9 × 1010 L‐1 were recorded and bacterial productivity reached a peak of 0.36 µg C L‐1 h‐1. Numbers of heterotrophic nanoflagellates between 5.0 × 104 and 1.8 × 107 L‐1, and of ciliates from undetectable to 1.1 × 104 L‐1 were observed. Naked amoebae were usually rare, but occasionally reached peaks of up to 1.5 × 103 L‐1.

1. This study focused on heterotrophic microorganisms in the two main basins (north and south) of Lake Tanganyika during dry and wet seasons in 2002. Bacteria (81% cocci) were abundant (2.28–5.30 × 106 cells mL?1). During the dry season, in the south basin, bacterial biomass reached a maximum of 2.27 g C m?2 and phytoplankton biomass was 3.75 g C m?2 (integrated over a water column of 100 m). 2. Protozoan abundance was constituted of 99% of heterotrophic nanoflagellates (HNF). Communities of flagellates and bacteria consisted of very small but numerous cells. Flagellates were often the main planktonic compartment, with a biomass of 3.42–4.43 g C m?2. Flagellate biomass was in the same range and often higher than the total autotrophic biomass (1.60–4.72 g C m?2). 3. Total autotrophic carbon was partly sustained by the endosymbiotic zoochlorellae Strombidium. These ciliates were present only in the euphotic zone and usually contributed most of the biomass of ciliates. 4. Total heterotrophic ciliate biomass ranged between 0.35 and 0.44 g C m?2. In 2002, heterotrophic microorganisms consisting of bacteria, flagellates and ciliates represented a large fraction of plankton. These results support the hypothesis that the microbial food web contributes to the high productivity of Lake Tanganyika. 5. As the sole source of carbon in the pelagic zone of this large lake is phytoplankton production, planktonic heterotrophs ultimately depend on autochthonous organic carbon, most probably dissolved organic carbon (DOC) from algal excretion.  相似文献

Extracellular phosphatases are an important part of the phosphorus cycle in aquatic environments. Phosphatase activity (PA) in plankton was studied in seven subtropical shallow lakes of different exploitation management and trophic status in the urban area of Wuhan City. Bulk PA was rather high (range 1.1–11 μmol l−1 h−1), although concentrations of soluble reactive phosphorus (SRP) were also high (range 27 μg P l−1 to ~1.5 mg P l−1) in all lakes. Cell-associated extracellular PA in phytoplankton was detected using the fluorescence-labelled enzyme activity technique. Phytoplankton species partly contributed to the bulk PA. We found explicit differences in the presence of cell-associated phosphatase within the main phytoplankton groups; species belonging to Chlorophyta and Dinophyta were regularly phosphatase-positive, while Cyanophyta and Bacillariophyceae were phosphatase-negative in all but one case. Furthermore, there is a certain potential of extracellular phosphatases produced by heterotrophic nanoflagellates in most of the lakes. This new finding compromises the ‘traditional’ interpretation of bulk phosphatase data as being due to overall phytoplankton or bacterial P regeneration.  相似文献

从汕尾健生鲍鱼场养殖水体及鲍鱼消化道中分离筛选到26株异养细菌,分析了其胞外产物,并采用API条带对其进行了种类鉴定。结果表明,就整体而言,成鲍消化道菌株产蛋白酶、淀粉酶、明胶酶和溶血能力均高于养殖水体的菌株,而产脂肪酶和卵磷脂酶菌株的比例则低于后者。无论是消化道还是养殖水体,均存在着分泌胞外产物能力极强的菌株,且大部分为鞘氨醇单胞菌。因此,在该养殖环境中,鞘氨醇单胞菌应视为一种主要的条件致病菌,同时消化道和养殖水体均应视为潜在致病菌的源泉。此外,在考虑细菌对水产经济动物的致病作用时,除了优势种类外,还应从菌群结构的角度出发,考虑整个细菌群落胞外产物的作用。  相似文献
