The basidiomycete Phlebiop sis gigantea (Fr.) Jülich has been used in Swedish forestry as a biocontrol agent against the root and butt-rot pathogen Heterobasidion annosum s.l. on freshly cut Picea abies stumps since the early 1990s. Until 2005, the commercial preparation of this biological stump treatment, Rotstop®, has been based on a single strain of P. gigantea that has been applied on more than 47,000 ha annually in Fennoscandia. This paper reports on the spread of genetic material from the Rotstop® biocontrol strain of P. gigantea to resident populations of P. gigantea. We conclude that the inoculated fungus remained to a large extent restricted to the treated plots and did not spread to the adjacent areas, dominated by the natural spore deposition from resident populations of the fungus. Furthermore, the study demonstrates high genetic diversity and low geographic differentiation in P. gigantea populations in the geographical area around the Baltic Sea.  相似文献

In ectomycorrhizae auxins are proposed to attenuate elicitor-induced defence reactions in the host plant. To examine this hypothesis we compared the elicitor-induced accumulation of peroxidase isoforms between suspension-cultured spruce (Picea abies[L.] Karst.) cells incubated in media with and without auxins. In spruce cells changes in ionically and covalently wall-bound as well as symplasmic peroxidase (EC 1.11.1.7) activities were observed when elicitors from the following fungal species were applied: (1) Hebeloma crustuliniforme, an ectomycorrhizal partner of spruce; (2) Suillus variegatus, an ectomycorrhizal fungus incompatible with spruce; (3) Heterobasidion annosum, a spruce pathogen. Activity staining after SDS-PAGE and western blotting showed an accumulation of an ionically wall-bound 38-kDa peroxidase isoform. In addition, two covalently wall-bound isoforms (34 and 53 kDa) that could be released from spruce cell walls by cellulase and pectinase treatment were also induced by elicitors from these fungi. Moreover, in cells cultured without auxins all the elicitors triggered a rapid and transient accumulation of ionically wall-bound peroxidases, which reached a maximum activity 48 h after elicitor application. This early and transient peroxidase accumulation was diminished and delayed in cells cultured in the presence of auxins. In contrast, activity of peroxidases released into the culture medium of spruce cells or into the medium of protoplasts was suppressed by the elicitors of Hebeloma crustuliniforme. However, this suppression was attenuated by the action of auxins. It is suggested that under natural conditions, in infected spruce roots, the elicitors of the compatible fungus cause both suppression of the peroxidase (which is secreted to the free space of the roots), and induction of wall-bound and symplasmic peroxidases. On the other hand, auxins synthesized by the fungus could weaken these different elicitor-mediated effects.  相似文献

Lignin biosynthesis is a major carbon sink in gymnosperms and woody angiosperms. Many of the enzymes involved are encoded for by several genes, some of which are also related to the biosynthesis of other phenylpropanoids. In this study, we aimed at the identification of those gene family members that are responsible for developmental lignification in Norway spruce (Picea abies (L.) Karst.). Gene expression across the whole lignin biosynthetic pathway was profiled using EST sequencing and quantitative real-time RT-PCR. Stress-induced lignification during bending stress and Heterobasidion annosum infection was also studied. Altogether 7,189 ESTs were sequenced from a lignin forming tissue culture and developing xylem of spruce, and clustered into 3,831 unigenes. Several paralogous genes were found for both monolignol biosynthetic and polymerisation-related enzymes. Real-time RT-PCR results highlighted the set of monolignol biosynthetic genes that are likely to be responsible for developmental lignification in Norway spruce. Potential genes for monolignol polymerisation were also identified. In compression wood, mostly the same monolignol biosynthetic gene set was expressed, but peroxidase expression differed from the vertically grown control. Pathogen infection in phloem resulted in a general up-regulation of the monolignol biosynthetic pathway, and in an induction of a few new gene family members. Based on the up-regulation under both pathogen attack and in compression wood, PaPAL2, PaPX2 and PaPX3 appeared to have a general stress-induced function. The fungus Phlebiopsis gigantea has been used in Europe as a biological agent for the control of conifer root and butt (caused by Heterobasidion annosum ) for nearly 40 years. P. gigantea competes with H. annosum for the woody resource within conifer stumps, and is applied to stump surfaces at felling. Three distinct biological control products based on P. gigantea have been developed: PG Suspension in the UK, PG IBL in Poland and Rotstop in Finland. The formulations are of oidia, which are maintained in a sucrose suspension, sawdust, or a wettable powder, respectively. PG Suspension and PG IBL are applied to pine stumps, while Rotstop is equally as effective on pine as on Norway spruce stumps. For each product, isolates of P. gigantea are selected from the wild and are screened for their competitive ability against H. annosum before formulation. Viability and purity checks are undertaken throughout the production cycle and during routine use. The increasing use of mechanized harvesting machines to fell and process trees is having an impact on this biological control system, the formulations having to be compatible with the mechanical application systems and vice versa. This paper compares the formulation, testing and application of the three products, and considers some aspects of their future development.  相似文献

1 A four-arm olfactometer was used to test the effect of permethrin ‘Gori 920®’ and the wax treatment ‘CereNat® E30’ on the response of mature adult Hylobius abietis to olfactory stimuli from seedlings of Pinus sylvestris and Picea abies. 2 Also investigated in the olfactometer was the effect of Rotstop®, a biofungicide for the control of Fomes root and butt rot, Heterobasidion annosum, on the responses of mature adult female H. abietis to stump material of both P. sylvestris and P. abies. 3 Hylobius abietis was significantly more attracted to untreated P. sylvestris seedlings compared to those treated with CereNat® E30 or permethrin Gori 920®. No differences in response to P. abies seedlings were found. 4 With Rotstop®, no changes in the attractiveness of P. sylvestris stump material were found, but that of P. abies was significantly increased compared to untreated material.  相似文献

Internal transcribed spacer (ITS) sequences of the rDNA repeat unit of Heterobasidion annosum were used to design specific primers for the detection and quantification of this important forest pathogen by polymerase chain reaction (PCR). Specificity of detection was cross‐checked against a variety of other fungi (saprophytes, root pathogens, mycorrhizal fungi) which may occur in the same environment. As little as 1 pg fungal DNA (equiv. to 10–40 genomes) could be detected in 200 ng spruce root DNA (from 1 mg fresh spruce root). The Heterobasidion‐specific primers allowed simultaneous detection of Armillaria spp. in multiplex PCR. The method was successfully applied to increment cores of Norway spruce from the forest region Tharandter Wald (Saxonia, Germany), Oberbärenburg (East Ore Mountains, Saxonia) and Oberschleissheim (north of Munich, Bavaria).  相似文献

Using taxon-specific competitive-priming polymerase chain reaction (TSCP-PCR) the intersterility groups (ISGs) S and P of the root and butt rot pathogen Heterobasidion annosum were found in an active pine mortality center in the forest region Tharandter Wald (Saxonia, Germany). All isolates from spruce were typed as S, and all isolates from pine were P, except for two isolates which were identified as hybrid 'SP’genets. These results indicate the possibility of gene flow in nature between the two ISGs in Europe.  相似文献
