  • Pollution of coasts by toxic metals and metalloids is a worldwide problem for which phytoremediation using halophytes and associated microbiomes is becoming relevant. Metal(loid) excess is a constraint for plant establishment and development, and plant growth promoting rhizobacteria (PGPR) mitigate plant stress under these conditions. However, mechanisms underlying this effect remain elusive. The effect of toxic metal(loid)s on activity and gene expression of ROS‐scavenging enzymes in roots of the halophyte Spartina densiflora grown on real polluted sediments in a greenhouse experiment was investigated.
  • Sediments of the metal‐polluted joint estuary of Tinto and Odiel rivers and control, unpollutred samples from the Piedras estuary were collected and submitted to ICP‐OES. Seeds of S. densiflora were collected from the polluted Odiel marshes and grown in polluted and unpolluted sediments. Rhizophere biofilm‐forming bacteria were selected based on metal tolerance and inoculated to S. densiflora and grown for 4 months. Fresh or frozen harvested plants were used for enzyme assays and gene expression studies, respectively.
  • Metal excess induced SOD (five‐fold increase), whereas CAT and ascorbate peroxidase displayed minor induction (twofold). A twofold increase of TBARs indicated membrane damage. Our results showed that metal‐resistant PGPR (P. agglomerans RSO6 and RSO7 and B. aryabhattai RSO25) contributed to alleviate metal stress, as deduced from lower levels of all antioxidant enzymes to levels below those of non‐exposed plants. The oxidative stress index (OSI) decreased between 50 and 75% upon inoculation.
  • The results also evidenced the important role of PAL, involved in secondary metabolism and/or lignin synthesis, as a pathway for metal stress management in this halophyte upon inoculation with appropriate PGPR, since the different inoculation treatments enhanced PAL expression between 3.75‐ and five‐fold. Our data confirm, at the molecular level, the role of PGPR in alleviating metal stress in S. densiflora and evidence the difficulty of working with halophytes for which little genetic information is available.

The present study aims to monitor the surface water quality of different regions in Delhi (India). With many physical and chemical properties, all samples had a high load of pollution in which Najafgarh drain (Nd) exhibited maximum and laboratory tap water (Ltw) minimum contamination. Water samples contained notable amounts of heavy metals including Cr, Cd, As, Cu, Pb and Hg. A total of 88 Hg-resistant bacteria were isolated from all the regions except Ltw. Among all the samples, the density of Hg-resistant bacteria was highest in sample of Nd and their morphotype heterogeneity was highest in sample collected from river Yamuna nearby Kashmiri gate (Kg). Different strains showed different patterns of resistance to different heavy metals and antibiotics. Multiple antibiotic resistance (MAR) indices were high in two samples, the highest reported in a sample taken from river Yamuna nearby Majnu ka tila (Mkt) (0.34). The 12.5% and 24.45% isolates showed β- and α-hemolytic natures, respectively that might be of pathogenic concern. In this account, high concentrations of heavy metals and their resistant bacteria in surface water have severely damaged the quality of water and their resources and produced high risk to the associated life forms.  相似文献

Pollution is one of the most important factors inhibiting growth in the environment; therefore the effects of zinc pollution were studied in Acer velutinum specie. Two years old seedlings of Acer velutinum specie were prepared from nursery, the concentrations of zero and one hundred and thirty mg per liter of zinc chloride solution were added to the soil of seedlings pots after calculating and after passing a three-month period of seedling growth, the plant organs were removed, then the amount of zinc concentration in the samples was determined and data were analyzed. The results of the analysis showed that the highest amount of accumulation on the stem, root and soil in treatment concentrations is 87.75, 65.68 and 24.78 mg per kg and accumulation of zinc in total chlorophyll and sugar in treatment concentrations is 4.61 and 0.6028 mg per g, respectively, and accordingly Acer velutinum specie is suitable for refinement of soils contaminated with zinc.  相似文献

The aim of the present study was to investigate the effects of environmental pollutants, such as heavy metals and pesticides on ion transport across the skin of the leech (Hirudo medicinalis). We wanted to examine the suitability of this epithelium as a model system for studies concerning the mechanisms of toxic action caused by environmental pollutants. For this purpose we performed Ussing chamber experiments to test three representative heavy metals and pesticides, respectively, for their effects on current flow across leech dorsal integument. Two representatives of each substance class showed distinct effects on ion transport across this epithelium. The heavy metal ions Pb2+ and Hg2+ produced a significant inhibition of amiloride-sensitive Na+ transport across leech skin in concentrations below or close to their limiting values in waste water. Therefore, it seems feasible to use leech skin for future investigations of the toxic actions of these heavy metals. The fact that Pb2+ and Hg2+ exerted their effects only when applied apically points to a specific action of these divalent cations on ion channels in the apical membrane. However, this inhibition does not seem to be a general feature of divalent cations because Cd2+ did not influence ion transport across leech skin at all. Since current flow through amiloride-sensitive Na+ channels in typical vertebrate tight epithelia is stimulated by numerous divalent cations, the pronounced inhibition of amiloride-sensitive Na+ channels in leech skin by Pb2+ and Hg2+ might lead to a further differentiation of amiloride-sensitive Na+ channels. The two widespread pesticides lindane and promecarb exerted their effects only at comparativ high concentrations. This low sensitivity restricts the usefulness of leech skin as a subject for further analysis of toxicity mechanisms, at least for these two pesticides.  相似文献

研究了胶州湾海水、海洋生物体中重金属含量.结果表明,沿岸河口地区海水中重金属污染主要受沿岸工业排放废水的影响;海洋生物体内重金属含量不仅受海水中的重金属含量大小的支配,温度对其也有一定的影响.海洋生物体中重金属含量为底栖生物大于鱼类,以沉积物为主要饵料的杂食性底栖动物内重金属含量大于鱼类;海洋生物对于Cu比其它重金属具有较强的富集能力.  相似文献

Copper (Cu) chaperones constitute a family of small Cu+-binding proteins required for Cu homeostasis in eukaryotes. The ATX1 family of Cu chaperones specifically delivers Cu to heavy metal P-type ATPases. The plant Arabidopsis thaliana expresses the ATX1-like Cu chaperone CCH, which exhibits a plant-specific carboxy-terminal domain (CTD) with unique structural properties. We show that CCH homologues from other higher plants contain CTDs with structural properties similar to Arabidopsis CCH. Furthermore, we identify a new ATX1-like Cu chaperone in Arabidopsis, AtATX1, which functionally complements yeast atx1Delta and sod1Delta associated phenotypes, and localizes to the cytosol of Arabidopsis cells. Interestingly, AtATX1, but not full-length CCH, interacts in vivo with the Arabidopsis RAN1 Cu-transporting P-type ATPase by yeast two-hybrid. We propose that higher plants express two types of ATX1-like Cu chaperones: the ATX1-type with a predominant function in Cu delivery to P-type ATPases, and the CCH-type with additional CTD-mediated plant-specific functions.  相似文献

Streptomyces lividans 1326 carries inducible mercury resistance genes on the chromosome, which are arranged in two divergently transcribed operons. Expression of the genes is negatively regulated by the repressor MerR, which binds in the intercistronic region between the two operons. The merR gene was expressed in E. coli using a T7 RNA polymerase/promoter expression system, and MerR was purified to around 95% homogeneity by ammonium sulfate precipitation, gel filtration and affinity chromatography. Gel filtration showed that the native MerR is a dimer with a molecular mass of 31 kDa. Two DNA binding sites were identified in the intercistronic mer promoter region by footprinting experiments. No evidence for cooperativity in the binding of MerR to the adjacent operator sequences was observed in gel mobility shift assays. The dissociation constants (KD) for binding of MerR were: binding site I, 8.5 × 10−9 M; binding site II, 1.2 × 10−8 M; and for the complete promoter/operator region 1 × 10−8 M. The half-life of the MerR-DNA complex was 19.4 min and 18.8 min for binding site I and binding site II, respectively. The KD value for binding of mercury(II)chloride to MerR, again determined by mobility shift assay, was 1.1 × 10−7 M. Received: 18 August 1998 / Accepted: 5 May 1999  相似文献

本文利用油松年轮研究了承德市自1800年以来大气污染的历史过程,结果表明:避暑山庄油松年轮内S含量自本世纪初开始升高,比19世纪初增加1—2倍,至70—80年代达3—5倍,而在最近8—10年则达到10倍以上。 其中碧峰门古松木质部年轮内S含量由40—50μg·g-1达到目前的572.9μg·g-1,从而指示出承德市大气SO2浓度由城市化初期的<0.1 mg·m-3增加到目前的30mg·m-3,这一历史过程主要与城市化过程尤其是近50年来工业化的出现并加剧有密切的关系。随着1927—1945年大庙铁矿的开采,年轮内Fe明显增高。Mn在整个历史时期表现为增加趋势,由1840—1850年的4.1μg·g-1增加到1980—1990年的10.4μg·g-1。Pb在1980年前的含量增加不明显,但在最近10年内形成的木质部中,Pb含量明显增加,超过1920—1930年的560%。Cu、Zn、Ni含量的变化不表现线性增加,除个别点的某些时期变化较大外,基本上很平缓。19世纪初油松年轮内S及重金属含量最低,可视为油松内元素自然背景值,与该值比较,1980—1990年这10年内年轮S含量增加了10倍,Fe,2.4倍;Pb,1.8倍;Mn,1.5倍;Ni,1.0倍;Zn,0.8倍。 日益加剧的大气污染尤其是SO2的污染,造成承德市大气质量严重下降,同时危及古松的生存,造成古松大量死亡,必须引起高度重视。  相似文献
