Apoptosis-induced directed fractionation and purification was used to identify the bioactive components of hard clams (HC), Meretrix lusoria. Two stereoisomers of epidioxysterol were previously identified as the active compounds in the ethyl acetate fraction (HC-EA). The molecular mechanism of HC-EA-induced apoptosis was also investigated in this study. Dissipation of mitochondrial membrane potential, release of mitochondrial cytochrome c into cytosol, and subsequent induction of pro-caspase-9 and -3 processing preceded apoptosis in HL-60 cells, confirmed by DNA fragmentation, chromatin condensation, changes in the cell membrane and the appearance of a sub-G1 DNA peak. Furthermore, treatment with HC-EA caused a rapid loss of intracellular glutathione content and stimulation of reactive oxygen species (ROS). Antioxidants such as catalase, N-acetylcysteine, pyrrolidine dithiocarbamate, and superoxide dismutase, but not allopurinol and diphenylene iodonium, significantly inhibited HC-EA-induced cell death. Apoptosis was completely prevented by a pan-caspase inhibitor, z-Val-Ala-Asp-fluoromethyl ketone (z-VAD-FMK). The induction of apoptosis by M. lusoria may prove to be a pivotal mechanism for its cancer chemopreventive action.  相似文献

我国的水稻育种经历了矮化育种、杂种优势利用和绿色超级稻培育3次飞跃,其间伴随矮化育种(第一次绿色革命)、三系杂交稻培育、二系杂交稻培育、亚种间杂种优势利用、理想株型育种和绿色超级稻培育等6个重要历程。育种目标从唯产量是举到高抗、优质和高产并重,育种理念从高产优质逐步提升为“少投入,多产出,保护环境”。水稻功能基因组研究为第二次绿色革命准备了大量的有重要利用价值的基因,水稻育种正迈向设计育种的新时代。基因组选择技术和转基因技术将为培育“少打农药,少施化肥,节水抗旱,优质高产” 绿色超级稻保驾护航。本文对我国水稻遗传育种的发展历程进行了概括,指出了各种育种方法和育种技术的优缺点,系统介绍了水稻细胞质雄性不育和光温敏雄性核不育以及籼粳杂种不育的分子机制的研究进展,综述了水稻株型、穗型、粒形和养分高效利用相关的重要功能基因,阐明了产量与开花期联动的关系,凸显了我国水稻基础研究在国际上的重要地位。特别指出,近年来,我国水稻生产方式发生了或正在发生巨大变革,育种理念也要与时俱进。未来,杂交育种技术要与现代育种技术紧密结合,选育水稻品种不仅要满足市场需求,而且更要具备绿色健康的特点,同时还要适应新耕作制度和新耕作方法。  相似文献

食粪行为广泛存在于草食性中小型哺乳动物中,是特指动物取食由盲肠内容物所形成的粪便的行为。这些动物具有特殊的结肠分离机制,能够产生两种不同的粪便。食粪行为延长了食物在消化道中的平均滞留时间;提高了对高纤维食物的消化率;弥补了食物中氨基酸和维生素的缺乏,满足了动物对这些营养物质的需求;是动物消化过程中的一个特殊组成部分。食粪行为的节律性是动物对取食和食粪的风险权衡的结果。食粪行为的发展是与动物草食性的特点紧密相关的,是动物对身体较小、食物质量低和天敌威胁等不利因素适应的结果。本对这些方面的研究进展进行了综述。  相似文献

A major feat of social beings is to encode what their conspecifics see, know or believe. While various non-human animals show precursors of these abilities, humans perform uniquely sophisticated inferences about other people''s mental states. However, it is still unclear how these possibly human-specific capacities develop and whether preverbal infants, similarly to adults, form representations of other agents'' mental states, specifically metarepresentations. We explored the neurocognitive bases of eight-month-olds'' ability to encode the world from another person''s perspective, using gamma-band electroencephalographic activity over the temporal lobes, an established neural signature for sustained object representation after occlusion. We observed such gamma-band activity when an object was occluded from the infants'' perspective, as well as when it was occluded only from the other person (study 1), and also when subsequently the object disappeared, but the person falsely believed the object to be present (study 2). These findings suggest that the cognitive systems involved in representing the world from infants'' own perspective are also recruited for encoding others'' beliefs. Such results point to an early-developing, powerful apparatus suitable to deal with multiple concurrent representations, and suggest that infants can have a metarepresentational understanding of other minds even before the onset of language.  相似文献

The mandibular third premolar (P3) of Australopithecus afarensis is notable for extensive morphological variability (e.g., metaconid presence/absence, closure of the anterior fovea, root number) and temporal trends in crown length and shape change over its 700 Ka time range. Hominins preceding A. afarensis have unicuspid, mesiodistally elongated P3s with smaller talonids, and subsequent australopiths have bicuspid, more symmetrically-shaped P3 crowns with expanded talonids. For these features, A. afarensis is intermediate and, thus, evinces the incipient stages of P3 molarization. Here, we examine A. afarensis P3 Phase II microwear and compare it with that of Australopithecus africanus and Cercocebus atys, an extant hard-object specialist, to assess whether the role of the P3 in food processing changed over time in A. afarensis. Premolar Phase II microwear textures are also compared with those of the molars to look for evidence of functional differentiation along the tooth row (i.e., that foods with different mechanical properties were processed by separate regions of the postcanine battery). Microwear textures were also examined along the mesial protoconid crest, the site of occlusion with the maxillary canine, of the A. afarensis P3 and compared with the same region in Pan troglodytes to determine whether microwear can be useful for identifying changes in the occlusal relationship between the P3 and maxillary canine in early Australopithecus. Finally, temporal trends in P3 Phase II and mesial microwear are considered. Results indicate that 1) both the P3 and molar Phase II facets of A. afarensis have less complex microwear textures than in A. africanus or C. atys; 2) A. afarensis P3 and molar Phase II textures differ, though not to the extent seen in taxa that eat hard and tough items; 3) microwear along the A. afarensis mesial protoconid crest is clearly distinct from that of the P. troglodytes, indicating that there is no honing equivalent in A. afarensis; and 4) there is little evidence of change over time in A. afarensis P3 microwear on either the mesial or Phase II facet. In sum, these results provide no evidence that A. afarensis routinely loaded either its premolars or molars to process hard objects or that A. afarensis P3 function changed over time.  相似文献

Autophagy plays a key role in the pathophysiology of schizophrenia as manifested by a 40% decrease in BECN1/Beclin 1 mRNA in postmortem hippocampal tissues relative to controls. This decrease was coupled with the deregulation of the essential ADNP (activity-dependent neuroprotector homeobox), a binding partner of MAP1LC3B/LC3B (microtubule-associated protein 1 light chain 3 β) another major constituent of autophagy. The drug candidate NAP (davunetide), a peptide fragment from ADNP, enhanced the ADNP-LC3B interaction. Parallel genetic studies have linked allelic variation in the gene encoding MAP6/STOP (microtubule-associated protein 6) to schizophrenia, along with altered MAP6/STOP protein expression in the schizophrenic brain and schizophrenic-like behaviors in Map6-deficient mice. In this study, for the first time, we reveal significant decreases in hippocampal Becn1 mRNA and reversal by NAP but not by the antipsychotic clozapine (CLZ) in Map6-deficient (Map6+/?) mice. Normalization of Becn1 expression by NAP was coupled with behavioral protection against hyperlocomotion and cognitive deficits measured in the object recognition test. CLZ reduced hyperlocomotion below control levels and did not significantly affect object recognition. The combination of CLZ and NAP resulted in normalized outcome behaviors. Phase II clinical studies have shown NAP-dependent augmentation of functional activities of daily living coupled with brain protection. The current studies provide a new mechanistic pathway and a novel avenue for drug development.  相似文献

The part of the primate visual cortex responsible for the recognition of objects is parcelled into about a dozen areas organized somewhat hierarchically (the region is called the ventral stream). Why are there approximately this many hierarchical levels? Here I put forth a generic information-processing hierarchical model, and show how the total number of neurons required depends on the number of hierarchical levels and on the complexity of visual objects that must be recognized. Because the recognition of written words appears to occur in a similar part of inferotemporal cortex as other visual objects, the complexity of written words may be similar to that of other visual objects for humans; for this reason, I measure the complexity of written words, and use it as an approximate estimate of the complexity more generally of visual objects. I then show that the information-processing hierarchy that accommodates visual objects of that complexity possesses the minimum number of neurons when the number of hierarchical levels is approximately 15.  相似文献

Mirror mediated object discrimination was investigated in a captive female, human-reared, western lowland gorilla (Gorilla gorilla gorilla) approximately 26 years of age. The gorilla was trained to find a stimulus that was only visible by use of the mirror. The gorilla could not reach the goal object except by observing it in the mirror and her hand movement was not visible through the mirror. Further, it was hypothesized that this mirror mediated object discrimination would enhance the probability of self-directed behavior.Gallup’s (1970) marking paradigm, excluding the use of general anesthesia, was utilized in assessing self-directed behavior. Mirror-gazing, face-directed, and mark-directed behavior with a mirror were compared before versus after discrimination mirror training. The results supported the hypotheses that gorillas are capable of mirror mediated object discrimination and that mirror training involving a discrimination task would facilitate self-directed behavior.  相似文献
