Incubation of maize branching enzyme, mBEI and mBEII, with 100 μM diethylpyrocarbonate (DEPC) rapidly inactivated the enzymes.
Treatment of the DEPC-inactivated enzymes with 100–500 mM hydroxylamine restored the enzyme activities. Spectroscopic data
indicated that the inactivation of BE with DEPC was the result of histidine modification. The addition of the substrate amylose
or amylopectin retarded the enzyme inactivation by DEPC, suggesting that the histidine residues are important for substrate
binding. In maize BEII, conserved histidine residues are in catalytic regions 1 (His320) and 4 (His508). His320 and His508
were individually replaced by Ala via site-directed mutagenesis to probe their role in catalysis. Expression of these mutants
inE. coli showed a significant decrease of the activity and the mutant enzymes hadK

以新鲜蚯蚓为原料,经过保温抽提、乙醇沉淀、DEAE-SepharoseFastFlow离子交换层析、Lysine-Sepharose4B亲和层析以及SDS-PAGE制备电泳等纯化步聚,得到一种纯度达95%以上的蚯蚓纤溶酶.该酶具有强烈的溶解纤维蛋白的作用及蛋白酶活性,平板法测得其比活性为90OUK单位/毫克蛋白,TAME法测得其比活性为2500O单位/毫克蛋白.酶学性质研究表明其最适反应温度为65℃,最适反应PH值为8.5.该酶的分子量为33kD,等电点为pH3.5.还对该酶进行了氨基酸组成分析,并测定了其N端部分序列. 相似文献

Abstract. Uptake of phytotoxic amounts of metal by higher plants or algae can result in inhibition of several enzymes, and in increase in activity (= induction) of others. Two mechanisms of enzyme inhibition predominate: (1) binding of the metal to sulphydryl groups, involved in the catalytic actionor structural integrity of enzymes, and (2) deficiency of an essential metal in metalloproteins or metal-protein complexes, eventually combined with substitution of the toxic metal for the deficient element. Metal accumulation in the cellular compartment of the enzyme is a prerequisite for enzyme inhibition in vivo. The induction of some enzymes is considered to play a significant role in the stress metabolism, induced by metal phytotoxicity. Peroxidase induction is likely to be related to oxidative reactions at the biomembrane; several enzymes of the intermediary metabolism might be stimulated to compensate for metal-sensitive photosynthetic reactions. The induction of enzymes and metal-specific changes in isoperoxidase pattern can be used as diagnostic criteria to evaluate the phytotoxicity of soils, contaminated by several metals. Lines for future research on metal phytotoxicity are proposed, involving the study of inhibition and induction of enzymes at the different cell membranes (especially the plasmamembrane) in vivo. 相似文献

The reproductive and diapausing adult females of brachypterous morph and macropterous females with reproductive arrest of non-diapause type, originating from the laboratory cultures of Pyrrhocoris apterus, were studied for their feeding and drinking behaviour, digestive enzyme activities, and carbohydrate and lipid contents. The highest feeding and drinking activities were observed in reproductive brachypters, the lowest in macropters. Macropters also differed from brachypters by lower activities of gut lipase, peptidase and protease, lower concentration of haemolymph sugars, and lower weight of fat body, which probably reflects their low feeding activity. The total content of fat body lipids was also lower in macropters (0.6 mg) than in reproductive and diapausing brachypters (4.6 and 7.5 mg, respectively) on day 14. A very high amount of glycogen was found in the fat body of diapausing brachypters, 363 μg on day 14, as opposed to 15 and 80 μg in macropterous and reproductive brachypterous females, respectively. The obtained data indicate that the most important difference between macropterous and brachypterous females with different types of reproductive arrest consists of an enhanced mobilization of lipids for dispersal in macropters and accumulation of energetic reserves for hibernation in brachypters. 相似文献

Originally classified as waste products, these compounds more recently have been investigated extensively by ecologists and pharmacologists, and many complex biological functions have been discovered. Secondary metabolites occur nearly in all living organisms, within bacteria as well as in mammals, and are especially prominent in those organisms lacking an immune system. Functions of plant secondary metabolites comprise attractants, such as color pigments and scents, repellents such as antifeedants against insects and mammals, or toxins that affect growth and development of animal and microbial predators. Conversely, insects can employ plant-synthesized compounds to their own advantage, such as signals for feeding and oviposition and location of prey. Microbes also use secondary metabolites as carbon sources, and bacteria utilize them for quorum-sensing, an aspect recently discovered. Despite the diversity of recognized functions, the biochemical processes underlying these interactions are few. Primarily, they relate to the ability of these small molecules to bind to receptor regions of various proteins such as keys into locks. This review attempts a summary of current knowledge of secondary plant metabolism with focus on history of discovery, development of analytical techniques, theories of origin and function, signal pathways, biosynthesis, and assessment of biological activities. Outlined is current utilization by, and future perspectives in, different disciplines, such as chemosystematics, chemical ecology, and agricultural biotechnology. Examples illustrate the strong potential of research in secondary metabolism, particularly in comparison to more established disciplines such as developmental biology and physiology. 相似文献

The targeting, insertion, and topology of membrane proteins have been extensively studied in both prokaryotes and eukaryotes. However, the mechanisms used by viral membrane proteins to generate the correct topology within cellular membranes are less well understood. Here, the effect of flanking charges and the hydrophobicity of the N-terminal hydrophobic segment on viral membrane protein topogenesis are examined systematically. Experimental data reveal that the classical topological determinants have only a minor effect on the overall topology of p9, a plant viral movement protein. Since only a few individual sequence alterations cause an inversion of p9 topology, its topological stability is robust. This result further indicates that the protein has multiple, and perhaps redundant, structural features that ensure that it always adopts the same topology. These critical topogenic sequences appear to be recognized and acted upon from the initial stages of protein biosynthesis, even before the ribosome ends protein translation. 相似文献

采用盆栽控制试验,研究了土壤不同NaCl浓度(0(CK)、1‰、2‰、3‰、4‰、5‰和6‰)处理45 d对四季竹叶片脱落率和细胞膜透性以及立竹器官K+、Na+、Ca2+和Cl-等矿质离子的吸收、运输和分配的影响.结果表明,1‰~2‰NaCl处理对四季竹叶片脱落率和离子渗漏率无显著影响,3‰~6‰ NaCl处理显著提高了叶片脱落率和离子渗漏率,四季竹的盐胁迫伤害随土壤盐浓度的增大而加剧.随着Na+、Cl-在四季竹立竹各器官中的显著增加,竹根、竹秆、竹枝K+含量逐渐下降,Ca2+含量变化较小,并且K+、Ca2+在竹根、竹秆中的向上选择性运输能力逐渐减弱.由于竹叶在低浓度(1‰~2‰)和高浓度(3‰~6‰)盐胁迫下分别对Ca2+和K+具有较高的选择性吸收能力,随盐浓度的增大,竹叶K+含量迅速升高,Ca2+含量先升高后下降,这对维持竹叶的营养平衡和正常生长具有重要意义.3‰~6‰NaCl处理时,Na+、Cl-在竹叶中的浓度显著高于立竹其他器官,不仅降低了竹叶的渗透势,有利于水分的向上运输,而且四季竹还可以通过叶片脱落的方式降低体内的盐分含量,减轻盐离子毒害. 相似文献

Epidemiological studies suggest that intrauterine undernutrition plays an important role in the development of arterial hypertension and endothelial dysfunction in adulthood. We have evaluated the effect of the Renin Angiotensin System inhibition on the blood pressure and the mesenteric arteriolar reactivity of the intrauterine undernourished rats. Wistar rats were fed either normal or 50% of the normal intake diets, during the whole gestational period. In this study only the male offspring was used. At 16 weeks of age, the rats were used for the study of blood pressure, microvascular reactivity studied in vivo-in situ to Angiotensin II (Ang II), Bradykinin (Bk) and Acetylcholine (Ach) before and after either losartan (10 mg/kg/15 days) or enalapril (15 mg/kg/21 days) treatment. We also evaluated the mesenteric and plasmatic Angiotensin Converting Enzyme (ACE), renal function, lipid plasmatic content, and insulin and glucose metabolism. Intrauterine undernutrition induced hypertension and increased response of mesenteric arterioles to Ang II and decreased vasodilation to Bk and Ach. The treatments with losartan or enalapril normalized the blood pressure levels and significantly improved the arteriolar responses to Bk, Ach and reduced the response to Ang II. No differences have been detected to ACE activity, renal function, lipid content and insulin and glucose metabolism. This study shows for the first time that Renin Angiotensin System inhibitors can normalize the cardiovascular alterations induced by intrauterine undernutrition. 相似文献
