Laboratory studies with Neomegalotomus parvus(Westwood) (Hemiptera: Alydidae) with one nymph per Petri dish in multiple-choice tests indicated that seeds of pigeon pea [Cajanus cajan(L.) Mills.], lablab (Dolichos lablabL.), and soybean [Glycine max(L.) Merrill] were visited before seeds of common bean (Phaseolus vulgarisL.) and rice (Oryza sativaL.). The percentage of individuals engaging in dabbing/antennation resulting in probing, and percentage probing resulting in feeding, were higher on common bean (97%) and pigeon pea (87%) seeds than on lablab (55%), soybean (50%), or rice (5%) seeds. No significant differences were found in preference (number of flanges) among pigeon pea, common bean, and lablab, and preference (insects on foods) varied throughout the assessment period (5 d). In tests using 10 nymphs per dish, pigeon pea was the preferred food (number of flanges and insects on plants) throughout the period (5 d). In no-choice tests, the average duration of a feeding session and the longest feeding session were greater on lablab and common bean than on pigeon pea, soybean, or rice seeds. The number of feeding sessions was greater on seeds of common bean, pigeon pea, and soybean than on those of lablab or rice. Laboratory tests with N. parvusadults indicated that pigeon pea seeds were located faster, followed by common bean, soybean, and rice. When pods were tested, dabbing/antennation time was shorter on pigeon pea than on soybean, and probing time was longer on soybean than on pigeon pea or common bean. On pigeon pea, 100% of the insects probed the host, while on common bean and soybean pods, and on rice panicles, these values dropped to 71.8%, 46.0%, and 10.5%, respectively. Adults showed similar feeding times on pigeon pea, common bean, and soybean pods, but did not feed on rice panicles. Electronmicroscopical analysis showed the presence of two apical lobes with 12 peg sensilla on the labial tip. Sensillum tips were stained with silver nitrate solution, indicating a permeability of the cuticle and, therefore, their function as taste receptors.  相似文献

The stem borer Busseola fusca (Fuller) (Lepidoptera: Noctuidae) is a major pest of maize, Zea mays L., and sorghum, Sorghum bicolor (L.) Moench (both Poaceae), in sub-Saharan Africa. Like in many other lepidopteran insects, the success of B. fusca in recognizing and colonizing a limited variety of plants is based on the interaction between its sensory systems and the physicochemical characteristics of its immediate environment. The sensilla on the maxillary galeae of B. fusca larvae are typical of Lepidoptera and comprise two uniporous styloconic sensilla, which are contact chemoreceptors, three basiconic sensilla, and two aporous sensilla chaetica. The maxillary palp is two-segmented and has eight small basiconic sensilla at the tip, which were also found to be gustatory. The antennae of B. fusca larvae are short and simple. The sensilla of the antenna are composed of two aporous sensilla chaetica, three multiporous cone-shaped basiconic sensilla, three small basiconic sensilla, and one aporous styloconic sensillum. The basiconic sensillum located on the third antennal segment displayed a contact chemoreception response. The other basiconic sensilla did not show any action potential activity in tip-recording tests. The significant and positive dose–response curve obtained for the antennal basiconic sensillum with sucrose indicated for the first time the presence of gustatory chemoreceptors on the antennae of a lepidopteran larva.  相似文献

In recent years, primary cilia have emerged as key regulators in development and disease by influencing numerous signaling pathways. One of the earliest signaling pathways shown to be associated with ciliary function was the non-canonical Wnt signaling pathway, also referred to as planar cell polarity (PCP) signaling. One of the best places in which to study the effects of planar cell polarity (PCP) signaling during vertebrate development is the mammalian cochlea. PCP signaling disruption in the mouse cochlea disrupts cochlear outgrowth, cellular patterning and hair cell orientation, all of which are affected by cilia dysfunction. The goal of this protocol is to describe the analysis of PCP signaling in the developing mammalian cochlea via phenotypic analysis, immunohistochemistry and scanning electron microscopy. Defects in convergence and extension are manifested as a shortening of the cochlear duct and/or changes in cellular patterning, which can be quantified following dissection from developing mouse mutants. Changes in stereociliary bundle orientation and kinocilia length or positioning can be observed and quantitated using either immunofluorescence or scanning electron microscopy (SEM). A deeper insight into the role of ciliary proteins in cellular signaling pathways and other biological phenomena is crucial for our understanding of cellular and developmental biology, as well as for the development of targeted treatment strategies.  相似文献

The presence of human blood-group antigens in developing and adult hypothyroid rat cochleas was analyzed using antibodies directed against antigens H and B. During postnatal development, hypothyroid rat cochleas exhibited a highly selective expression of both B and H antigens, mainly at the hair cell level. Labeling for antigen B was found throughout the hair cells, whereas the antibody directed against antigen H selectively labeled the apical part of these cells. These immunostaining patterns were similar to those found in normal (euthyroid) rat cochleas, but antigenic expression periods were clearly prolonged. Thus, whereas in normal rat cochleas, the B and H antigenic expression disappears from postnatal day (PD) 9 on, in cochleas of hypothyroid rats the reactivity was intense until PD15; it decreased from this developmental stage, and was negative or only faintly positive at PD30. Therefore, in congenital hypothyroidism, hair cell immunoreactivity is present at developmental stages that are negative in normal rat cochleas. These results suggest that human blood-group antigen expression on the developing cochlear hair cells of rats is modulated by thyroxine and that thyroxine is necessary for the temporal expression pattern and secretion of normal glycoproteins.  相似文献

Olfactory responses at the receptor level have been thoroughly described in Drosophila melanogaster by electrophysiological methods. Single sensilla recordings (SSRs) measure neuronal activity in intact individuals in response to odors. For sensilla that contain more than one olfactory receptor neuron (ORN), their different spontaneous spike amplitudes can distinguish each signal under resting conditions. However, activity is mainly described by spike frequency.Some reports on ORN response dynamics studied two components in the olfactory responses of ORNs: a fast component that is reflected by the spike frequency and a slow component that is observed in the LFP (local field potential, the single sensillum counterpart of the electroantennogram, EAG). However, no apparent correlation was found between the two elements.In this report, we show that odorant stimulation produces two different effects in the fast component, affecting spike frequency and spike amplitude. Spike amplitude clearly diminishes at the beginning of a response, but it recovers more slowly than spike frequency after stimulus cessation, suggesting that ORNs return to resting conditions long after they recover a normal spontaneous spike frequency. Moreover, spike amplitude recovery follows the same kinetics as the slow voltage component measured by the LFP, suggesting that both measures are connected.These results were obtained in ab2 and ab3 sensilla in response to two odors at different concentrations. Both spike amplitude and LFP kinetics depend on odorant, concentration and neuron, suggesting that like the EAG they may reflect olfactory information.  相似文献

The effect of insect age on the neural responsiveness of gustatory sensilla was investigated. Electrophysiological recordings were obtained from type A and type D sensilla on the pro- and meso-thoracic tarsi, and from sensilla on the labellum of the turnip root fly,Delia floralis (Fallen) in response to potassium chloride, sucrose and sinigrin. The age of the fly did have an effect on the numbers of sensilla responding to the test stimuli and on the magnitude of the response, but there was no consistent pattern in these effects among sensilla. The labellar sensilla were more responsive to sucrose than the tarsal sensilla and the proportion of flies whose labellar sensilla responded to sucrose was initially low, but increased after day 2 of adult life. In contrast, the number of flies whose tarsal sensilla responded to stimulation with sucrose was initially high and decreased as the flies aged. There was a similar decline in the proportion of tarsal sensilla responding to potassium chloride. Neither the proportion of flies whose tarsal sensilla responded to sinigrin nor the magnitude of the response was influenced by the age of the fly. These finding are discussed in relation to the feeding and host selection behaviour of the fly.  相似文献

In arachnids, pedipalps are highly variable appendages that may be used in feeding, courtship, defense, and agonistic encounters. In cosmetid harvestmen, adults have pedipalps that feature flattened femora, spoon‐shaped tibiae, and robust tarsal claws. In contrast, the pedipalps of nymphs are elongate with cylindrical podomeres and are adorned with delicate pretarsi. In this study, we used scanning electron microscopy to examine the distribution of cuticular structures (e.g., sensilla chaetica, pores) on the elements of the pedipalps of adults and nymphs of three species of cosmetid harvestmen. Our results indicate that there is considerable ontogenetic variation in the morphology of the trochanter, femur, patella, tibia, and tarsus. The pretarsus of the nymph has a ventral patch of setae that is absent from the adult tarsal claw. We observed this structure on all three cosmetid species as well as on the pedipalps of an additional seven morphospecies of nymphs collected in Belize and Costa Rica. This structure may represent a previously unrecognized autapomorphy for Cosmetidae. Examinations of the pedipalps of antepenultimate nymphs of additional gonyleptoidean harvestmen representing the families Ampycidae, Cranaidae, Manaosbiidae, and Stygnidae revealed the occurrence of unusual, plumose tarsal setae, but no setal patches on the tarsal claw.  相似文献
