利用改进的Alex-phalloidin活细胞染色方法和激光共聚焦显微镜技术,观察川百合(Lilium davidii Duch)花粉原生质体极性形成及萌发过程中微丝骨架的列阵变化.结果表明,花粉原生质体从贮存状态,经过水合、极性形成至萌发花粉管,其微丝结构从短小的梭形体,经过形成均匀的网状结构、向细胞边缘汇集的平行排列的束状结构,逐渐变成多层连续环绕细胞的微丝束结构.用酪氨酸磷酸酶抑制剂苯胂化氧(PAO)处理花粉原生质体,在微丝的汇合处,肌动蛋白聚集成小的团块,花粉的萌发受到抑制;而利用酪氨酸磷酸激酶抑制剂genistein处理细胞,微丝结构的列阵变化与对照相似.结果说明,在川百合花粉萌发过程中,有某种酪氨酸磷酸酶参与了反应.  相似文献

Effects of ozone exposure on polyamines in Pinus sylvestris L. were studied in a long-term experiment. Ten- to 15-year-old Scots pines were exposed to target ozone levels which began at ambient + 40 ppb in May, decreasing to ambient air only by September for 3 growing seasons. The amount of ozone applied followed the natural pattern of variation in ozone concentrations in Northern Finland. The free, soluble conjugated and insoluble conjugated polyamines were analyzed during the experiment and shortly after termination of exposure as well as at the beginning of the following growing season. A carry-over effect was observed as ozone-induced reduction of free spermidine in the oldest needle year class, which developed during the first exposure season of the experiment. This reduction was observed both after the second and the third ozone exposure season. Conversely, after termination of the experiment, levels of free polyamines increased in the following growing season, and soluble conjugated polyamines decreased in the developing needles. The post-treatment changes in polyamine concentrations are hypothesized to be caused by stress-induced injuries or delayed recovery of metabolic processes rather than protective responses. It is noteworthy that some responses in polyamines were found in the developing needles nine months after terminating the ozone exposure. This suggests that stress-induced injuries to older needles affected metabolism of new developing needles.  相似文献

After extensive analysis, Ulva lactuca dried algae, collected from the Monastir coastal zone, was proven to be successful as an adsorbent for the removal of certain inorganic pollutants. The main objective of this study was the nonlinear modeling of heavy metal removal from an aqueous solution, using a freely available and well analyzed biomaterial, as well as the evaluation of its efficacy on various metal ion sorptions. Although relatively low specific surface area, compared to more conventional adsorbents, the selected biomaterial displays very interesting retention capacities when used with aqueous inorganic pollutants. The pseudo, first and second-order kinetic models were used to investigate the kinetic retention mechanism. Assuming the nonlinear form, the results indicate that the retention mechanism is diffusion controlled. Concerning the heavy metal uptake capacity, it was found that the selected biomaterial has a retention capacity of 67 mg g−1 of Ni(II), 112 mg g−1 of Cu(II), 127 mg g−1of Cd(II) and 230 mg g−1 of Pb(II).  相似文献

The aim of this study was to compare the relative contribution of ammonia-oxidizing archaea (AOA) and bacteria (AOB) to nitrification during agricultural waste composting. The AOA and AOB amoA gene abundance and composition were determined by quantitative PCR and denaturing gradient gel electrophoresis (DGGE), respectively. The results showed that the archaeal amoA gene was abundant throughout the composting process, while the bacterial amoA gene abundance decreased to undetectable level during the thermophilic and cooling stages. DGGE showed more diverse archaeal amoA gene composition when the potential ammonia oxidation (PAO) rate reached peak values. A significant positive relationship was observed between the PAO rate and the archaeal amoA gene abundance (R2=0.554; P<0.001), indicating that archaea dominated ammonia oxidation during the thermophilic and cooling stages. Bacteria were also related to ammonia oxidation activity (R2=0.503; P=0.03) especially during the mesophilic and maturation stages.  相似文献

Different alternative configurations and strategies for the simultaneous biological removal of organic matter and nutrients (N and P) in wastewater have been proposed in the literature. This work demonstrates a new successful strategy to bring in enhanced biological phosphorus removal (EBPR) to a conventional nitrification/denitrification system by means of bioaugmentation with an enriched culture of phosphorus accumulating organisms (PAO). This strategy was tested in a sequencing batch reactor (SBR), where an 8 h configuration with 3 h anoxic, 4.5 h aerobic and 25 min of settling confirmed that nitrification, denitrification and PAO activity could be maintained for a minimum of 60 days of operation after the bioaugmentation step. The successful bioaugmentation strategy opens new possibilities for retrofitting full-scale WWTP originally designed for only nitrification/denitrification. These systems could remove P simultaneously to COD and N if they were bioaugmented with waste purge of an anaerobic/aerobic SBR operated in parallel treating part of the influent wastewater.  相似文献

The Stay-Green Rice (SGR) protein is encoded by the SGR gene and has been shown to affect chlorophyll (Chl) degradation during natural and dark-induced leaf senescence. An SGR homologue, SGR-like (SGRL), has been detected in many plant species. We show that SGRL is primarily expressed in green tissues, and is significantly downregulated in rice leaves undergoing natural and dark-induced senescence. As the light intensity increases during the natural photoperiod, the intensity of SGRL expression declines while that of SGR expression increases. Overexpression of SGRL reduces the levels of Chl and Chl-binding proteins in leaves, and accelerates their degradation in dark-induced senescence leaves in rice. Our results suggest that the SGRL protein is also involved in Chl degradation. The relationship between SGRL and SGR and their effects on the degradation of the light-harvesting Chl a/b-binding protein are also discussed.  相似文献

A new subgenus of Polia, Metallopolia subgen. n., two new species (Polia (Metallopolia) dysgnorima sp. n. and P. (M.) metagnorima sp. n.) from China (Sichuan, Yunnan, Gansu), and a new subspecies, P. (M.) subviolacea kanchenjunga ssp. n. from Nepal and SE Tibet (Sikkim) are described. Prenatal exposure to neurotoxicants such as lead (Pb) may cause stable changes in the DNA methylation (5mC) profile of the fetal genome. However, few studies have examined its effect on the DNA de-methylation pathway, specifically the dynamic changes of the 5-hydroxymethylcytosine (5hmC) profile. Therefore, in this study, we investigate the relationship between Pb exposure and 5mC and 5hmC modifications during early development. To study the changes in the 5hmC profile, we use a novel modification of the Infinium™ HumanMethylation450 assay (Illumina, Inc.), which we named HMeDIP-450K assay, in an in vitro human embryonic stem cell model of Pb exposure. We model Pb exposure-associated 5hmC changes as clusters of correlated, adjacent CpG sites, which are co-responding to Pb. We further extend our study to look at Pb-dependent changes in high density 5hmC regions in umbilical cord blood DNA from 48 mother-infant pairs from the Early Life Exposure in Mexico to Environmental Toxicants (ELEMENT) cohort. For our study, we randomly selected umbilical cord blood from 24 male and 24 female children from the 1st and 4th quartiles of Pb levels. Our data show that Pb-associated changes in the 5hmC and 5mC profiles can be divided into sex-dependent and sex-independent categories. Interestingly, differential 5mC sites are better markers of Pb-associated sex-dependent changes compared to differential 5hmC sites. In this study we identified several 5hmC and 5mC genomic loci, which we believe might have some potential as early biomarkers of prenatal Pb exposure.  相似文献

Previously we showed that dormancy break of spindle tree embryos after gibberellic acid (GA3) treatment was followed by an increase in arginine decarboxylase (ADC) activity (Béranger-Novat N. et al., Plant Sc. 102: 139–145, 1994). These results indicated that arginine decarboxylase pathway mediate hormone-induced growth responses in spindle tree embryos. In the present investigation we show that in GA3-treated embryos diamine oxidase (DAO) increases immediately after putrescine content and the increase in DAO activity paralleles the accumulation of putrescine at the beginning of the culture (before the visible appearance of the radicle). In this system polyamine oxidase (PAO) increases immediately after DAO activity and follows closely the increase in spermidine content. These results demonstrate a direct correlation between the biosynthesis and oxidation of putrescine and spermidine. At every stage of development DAO and putrescine levels are lower than spermidine and PAO levels. Dormant embryos can be distinguished from GA3-treated embryos by a complete lack of putrescine accumulation. In dormant embryos compared to GA3-treated embryos DAO changed more or less in parallel and on the whole seemed to follow the same content and distribution, but the kinetics of the activation of DAOs were different in dormant embryos with a delay of 1.5 day for the first and 1 day for the second peak. During the first days of culture at least up to 4 days the distribution of spermidine and PAO in GA3-treated embryos followed the same pattern observed in dormant embryos, but the levels of spermidine and PAO were greatly reduced in dormant embryos. On the other hand the kinetics of the activation of PAOs were different in dormant embryos with a delay of 1 day. The results suggest that dormant embryos are deficient in their ability to synthesize polyamines efficiently and support the view that spermidine catabolism (via PAO pool) is limiting in untreated embryos during the first days of culture.  相似文献

We used carefully defined heme-hemopexin complexes to investigate the role of hemopexin in the catabolism of heme in vivo. Uptake of rabbit [59Fe]heme-[125I]hemopexin by rat liver was rapid. The liver-associated 125I reached a maximum 5 minutes after injection, nearly 7-fold higher than apo-hemopexin, whereas liver-associated 59Fe increased with time. This together with an inverse relationship of [125I]hemopexin in the liver and serum during the course of heme transport suggests that hemopexin was released from the liver back to the circulation. Saturation of uptake with heme-hemopexin, reaching about 170 pmol [125I]hemopexin (gm liver)?1 5 minutes after injection of 11 nmol, indicates a receptor-mediated process.We conclude that hemopexin delivers heme to the liver via interaction with a finite number of receptors and returns to the circulation.  相似文献
