Here we describe in more depth the previously published application of the fluorescent probe 4,5-diaminofluorescein (DAF-2)
in order to reliably measure low levels of nitric oxide (NO) as released from human endothelial cells invitro. The used approach is based on the following considerations a) use low concentrations of DAF-2 (0.1 μM) in order to reduce
the contribution of DAF-2 auto-fluorescence to the measured total fluorescence, and b) subtract the DAF-2 auto-fluorescence
from the measured total fluorescence. The advantage of this method is the reliable quantification of NO in a biological system
in the nanomolar range once thoroughly validated. Here we focus in addition to the previous publication (Leikertet al.,FEBS Lett 2001, 506:131–134) on aspects of validation procedures as well as limitations and pitfalls of this method.

The responsiveness of spontaneously active neurons in the subfornical organ (SFO) of adult ducks to angiotensin II (ANGII), norepinephrine (NE), isoproterenol (Iso, -agonist), phenylephrine (Phe,
1-agonist) and clonidine (Clo,
2-agonist) was investigated in brain slices with extracellular recording technique. 64% (n=90) of the neurons increased their activity after superfusion with ANGII, the rest were unresponsive. Application of NE activated 10 and inhibited 8 neurons (n=22); the excitation being correlated with an excitatory ANGII responsiveness of the same neurons and the inhibition with the absence of an ANGII responsiveness. Iso activated 74% (n=58) and Clo inhibited 88% (n=16) of the investigated neurons. Phe did not have an effect on the majority (60%) of the neurons and produced both excitatory and inhibitory actions on the remaining cells. These results offer a plausible explanation for the dose dependent dipsogenic effect of Iso and the failure of NE to elicit dose dependent drinking, which can be explained by its dual, excitatory and inhibitory effect on SFO neurons. It is further concluded, that peripherally applied Iso exerts its dipsogenic action in high concentration by a direct excitatory effect on SFO neurons via the open blood brain barrier. Under physiological conditions, afferent neuronal input of still unknown origin might specifically modulate the activity of SFO neurons, because plasma concentrations of NE are probably not high enough to activate SFO neurons from the blood side of the blood brain barrier.Abbreviations

HvLhcb1 a major light-harvesting chlorophyll a/b-binding protein in barley, is a critical player in sustainable growth under Fe deficiency. Here, we demonstrate that Fe deficiency induces phosphorylation of HvLhcb1 proteins leading to their migration from grana stacks to stroma thylakoid membranes. HvLhcb1 remained phosphorylated even in the dark and apparently independently of state transition, which represents a mechanism for short-term acclimation. Our data suggest that the constitutive phosphorylation-triggered translocation of HvLhcb1 under Fe deficiency contributes to optimization of the excitation balance between photosystem II and photosystem I, the latter of which is a main target of Fe deficiency. 相似文献

This article examines the cultivation of transnational connections, cosmopolitanism and global class consciousness among members of elite social clubs in Paris. Drawing from interviews with members, it compares how – according to their respective characteristics – various social clubs promote different kinds of bourgeois cosmopolitanism, while rejecting the more recent internationalism of upper-middle-class service clubs such as the Rotary. Each club's peculiar ethos, practice and representations of social capital are related to the features of competing clubs through relations of mutual symbolic distinction; for example, some clubs emphasize the ‘genuineness’ of links while stigmatizing others for the accent they put on utility. The varied forms of cosmopolitanism that they promote partly replicate these logics of distinction, eliciting struggles over the authenticity or inauthenticity of transnational connections. Yet, clubs also oppose each other according to the unequal emphasis that they place on international ties per se, which creates a competing axiology within the symbolic economy of social capital accumulation. 相似文献

In the present article, we report on the kinetics of brain penetration in rats of the H3R antagonist 1,1′‐[1,1′‐biphenyl‐4,4′‐diylbis(methylene)]bis‐[piperidine] ( 1 ), which had shown a favorable in vitro pharmacological profile and in vivo potency in preventing scopolamine‐induced amnesia. Two different approaches were employed: high‐performance liquid chromatography/electrospray‐mass spectrometry (HPLC/ESI‐MS) and ex vivo binding against the labeled agonist [3H]‐(R)‐α‐methylhistamine ([3H]RAMHA). Starting from the structure of 1 , the rigid piperidine ring was replaced by a flexible dipropylamino group (see 2 ) or by a morpholino ring (see 3 ), endowed with lower basicity. The effect of replacement on rat plasma and brain disposition in the 24 h after administration was analyzed. High (μM ) and persistent concentrations of 1 were found in rat plasma, while plasma levels were significantly lower (range: 0–200 nM ) for the other two derivatives. This could be explained, among other factors, by the higher stability, observed for 1 , to liver metabolic cleavage. The applied chemical modulation had an important effect on in vivo brain disposition, as, despite the comparable physico‐chemical properties, 2 did not show the tendency to accumulate within the brain, as stated by its brain vs. plasma concentration ratios, if compared to 1 . Plant regeneration from mesophyll protoplasts of Agrobacterium rhizogenes-transformed Astragalus melilotoides Pall. was here developed. The protoplasts were isolated directly from the leaves of the hairy root-induced plants. The highest
yield of protoplasts was obtained from fully expanded leaves of young plants. Their viability was up to 72 ± 2.3 %. The highest

Suppression of gene expression by small interfering RNA (siRNA) has proved to be a gene-specific and cost effective alternative
to other gene suppression technologies. Short hairpin RNAs (shRNAs) generated from the vector-based expression are believed
to be processed into functional siRNAs in vivo, leading to gene silencing. Since an shRNA library carries a large pool of potential siRNAs, such a library makes it possible
to knock down gene expression at the genome wide scale. Although much of research has been focused on generating shRNA libraries
from either individually made gene specific sequences or cDNA libraries, there is no report on constructing randomized shRNA
libraries, which could provide a good alternative to these existing libraries. We have developed a method of constructing
shRNAs from randomized oligonucleotides. Through this method, one can generate a partially or fully randomized shRNA library
for various functional analyses. We validated this procedure by constructing a p53-specific shRNA. Western blot revealed that
the p53-shRNA successfully suppressed expression of the endogenous p53 in MCF-7 cells. We then made a partially randomized
shRNA library. Sequencing of 15 randomly picked cloned confirmed the randomness of the library. Therefore, the library can
be used for various functional assays, such as target validation when a suitable screening or selection method is available. 相似文献
